Hepatic glucose production by glucagon is crucial for glucose homeostasis during fasting, yet the underlying mechanisms remain incompletely delineated. Although CD38 has been detected in the nucleus, its function in this compartment is unknown. Here, we demonstrate that nuclear CD38 (nCD38) controls glucagon-induced gluconeogenesis in primary hepatocytes and liver in a manner distinct from CD38 occurring in the cytoplasm and lysosomal compartments. We found that the localization of CD38 in the nucleus is required for glucose production by glucagon and that nCD38 activation requires NAD+ supplied by PKCδ-phosphorylated connexin 43. In fasting and diabetes, nCD38 promotes sustained Ca2+ signals via transient receptor potential melastatin 2 (TRPM2) activation by ADP-ribose, which enhances the transcription of glucose-6 phosphatase and phosphoenolpyruvate carboxykinase 1. These findings shed light on the role of nCD38 in glucagon-induced gluconeogenesis and provide insight into nuclear Ca2+ signals that mediate the transcription of key genes in gluconeogenesis under physiological conditions.

The risk of coronary heart disease is linked to cholesterol levels. Previous studies have reported that the level of human plasma cholesteryl esters is influenced more by genetic factors than the level of free cholesterol. To assess the genetic nature and population differences, a serum cholesterol with free and esterified choleterol were measured from 22 pairs of twins who ranged from 12 to 26 in age. Thin layer chromatography was used to separate cholestaryl ester and free cholesterol from the rest of serum lipids. No signfcant contributions of genetic effects were found for any of cholesterol variables. Heterogeneity in variance for the total cholesterol was found in comparison of monozygotic versus dizygotic twins. This might indicated an association of cholesterol variations with twin types as reported previously.

Mebendazole (MBZ), a microtubule depolymerizing drug commonly used for the treatment of helminthic infections, has been suggested as a repositioning candidate for the treatment of brain tumors. However, the efficacy of MBZ needs further study to improve the beneficial effect on the survival of those patients. In this study, we explored a novel strategy to improve MBZ efficacy using a drug combination. When glioblastoma cells were treated with MBZ, cell proliferation was dose-dependently inhibited with an IC50 of less than 1 μM. MBZ treatment also inhibited glioblastoma cell migration with an IC50 of less than 3 μM in the Boyden chamber migration assay. MBZ induced G2-M cell cycle arrest in U87 and U373 cells within 24 h. Then, at 72 h of treatment, it mainly caused cell death in U87 cells with an increased sub-G1 fraction, whereas polyploidy was seen in U373 cells. However, MBZ treatment did not affect ERK1/2 activation stimulated by growth factors. The marked induction of autophagy by MBZ was observed, without any increased expression of autophagy-related genes ATG5/7 and Beclin 1. Co-treatment with MBZ and the autophagy inhibitor chloroquine (CQ) markedly enhanced the anti-proliferative effects of MBZ in the cells. Triple combination treatment with temozolomide (TMZ) (another autophagy inducer) further enhanced the anti-proliferative effect of MBZ and CQ. The combination of MBZ and CQ also showed an enhanced effect in TMZ-resistant glioblastoma cells. Therefore, we suggest that the modulation of protective autophagy could be an efficient strategy for enhancing the anti-tumor efficacy of MBZ in glioblastoma cells.

Endostatin, a carboxyl-terminal fragment of collagen X VIII is known as an anti-angiogenic agent, that specifically inhibits the proliferation of endothelial cell and the growth of several primary tumor. We report here the purification and characteriztion of recombinant murine endostatin (rmEndostain) which was expressed in a prokaryotic expression system. This rmEndostatin has similar physiochemical properties of Yeast-produced recombinant endostatin, and it also specifically inhibits the proliferation and migration of bovine capillary endothelial cells stimulated by basic fibroblast growth factor. The biological activity of rmEndostatin was also shown by its anti-angiogenic ability on the chorioallantoc membrane of chick embryo in vivo. In this article we demonstrate the refolding and purification of rmEndostatin expressed suing E. coli system, to a biologically active and soluble from. During the transportation and storage, the traditional cottage industries’ gochujang (fermented hot pepper paste) products are often showing pack burst spoilage. Thus, the present study focused on the prevention of pack burst spoilage of traditional cottage industries gochujang products by using high-pressure processing (HPP) method. The HPP treatment (550 Mpa, 4 min) significantly reduced (90%, p<0.05) the pack burst of gochujang products. The lightness (L*), pH, and moisture content of high-pressure processed gochujang products were not significantly changed, whereas redness (a*) and yellowness (b*) of gochujang products were reduced. The total aerobic bacterial counts did not decrease remarkably, however, a significant decline in yeasts and mold counts (1.26-4.49 log CFU/g, p<0.05) were noted in HPP treated gochujang products. Reduction in the yeast counts upon HPP treatment resulting in the control of aerogenesis in gochujang products, which subsequently reduced the pack burst spoilage. The present findings concluded that HPP treatment is effective for the prevention of pack burst spoilage in gochujang products.
