        Comparative analysis of type 2 diabetes-associated gut microbiota between Han and Mongolian people

        Due to the different rates of diabetes in different ethnic groupsand the structural differences in intestinal microbiota, thisstudy evaluated the changes in diabetes-related intestinal microbiotain two ethnic groups. Fifty-six stool samples werecollected from subjects from the Han and Mongolian ethnicgroups in China, including participants without diabetes(non-diabetic, ND) and with type 2 diabetes (T2D). The 16SrDNA gene V3 + V4 area was extracted from microbiota,amplified by PCR, and used to perform high-throughput sequencingand screen differential microbiota associated withethnicity. The results showed that there were 44 T2D-relatedbacterial markers in the Han subjects, of which Flavonifractor,Alistipes, Prevotella, Oscillibacter, Clostridium XlVa,and Lachnospiracea_incertae_sedis were most closely relatedto diabetes. There were 20 T2D-related bacterial markers inthe Mongolian subjects, of which Fastidiosipila and Barnesiellawere most closely related to diabetes. The commonmarkers of T2D bacteria in the two ethnic groups were Papillibacterand Bifidobacterium. There were 17 metabolic pathwayswith significant differences between the ND and T2Dgroups in the Han group, and 29 metabolic pathways in theMongolian group. The glutamatergic metabolic pathway wasthe only common metabolic pathway in two ethnic groups. The composition and function of diabetes-related bacteriawere significantly different among the different ethnic groups,which suggested that the influence of ethnic differences shouldbe fully considered when studying the association betweendiabetes and bacteria. In addition, the common bacterialmarkers found in diabetic patients of different ethnic groupsin this study can be used as potential targets to study the pathogenesisand treatment of diabetes.

        Lifetime Prediction and Aging Behaviors of Nitrile Butadiene Rubber under Operating Environment of Transformer

        Based on the actual operating environment of transformer, the aging tests of nitrile butadiene rubber (NBR) were conducted systematically under four conditions: in air, in transform oil, under compression in air and under compression in transform oil to studythe effect of high temperature, transform oil and compression stress simultaneously on the thermal aging behaviors of nitrile butadiene rubber and predict the lifetime. The effects of liquid media and compression stress simultaneously on the thermal aging behaviors of nitrile butadiene rubber were studied by using characterization methods such as IR spectrosc-opy, thermogravimetric measurements, Differential Scanning Calorimetry (DSC) measurements and mechanical property measurements. The changes in physical properties during the aging process were analyzed and compared. Different aging conditions yielded materials with different properties. Aging at 70°C under compression stress in oil, the change in elongation at break was lower than that aging in oil, but larger than that aging under compression in air. The compression set or elongation at break as evaluation indexes, 50% as critical value, the lifetime of NBR at 25°C was predicted and compared. When aging under compression in oil, the prediction lifetime was lower than in air and under compression in air, and in oil. It was clear that when predicting the service lifetime of NBR in oil sealing application, compression and media liquid should be involved simultaneously. Under compression in oil, compression set as the evaluation index, the prediction lifetime of NBR was shorter than that of elongation at break as the evaluation index. For the life prediction of NBR, we should take into account of the performance trends of NBR under actual operating conditions to select the appropriate evaluation index.

      • Inotodiol Inhabits Proliferation and Induces Apoptosis through Modulating Expression of cyclinE, p27, bcl-2, and bax in Human Cervical Cancer HeLa Cells

        Inonotus obliquus is a medicinal mushroom that has been used as an effective agent to treat various diseases such as diabetes, tuberculosis and cancer. Inotodiol, an included triterpenoid shows significant anti-tumor effect. However, the mechanisms have not been well documented. In this study, we aimed to explore the effect of inotodiol on proliferation and apoptosis in human cervical cancer HeLa cells and investigated the underlying molecular mechanisms. HeLa cells were treated with different concentrations of inotodiol. The MTT assay was used to evaluate cell proliferating ability, flow cytometry (FCM) was employed for cell cycle analysis and cell apoptosis, while expression of cyclinE, p27, bcl-2 and bax was detected by immunocytochemistry. Proliferation of HeLa cells was inhibited by inotodiolin a dose-dependent manner at 24h (r=0.9999, p<0.01). A sub-$G_1$ peak (apoptotic cells) of HeLa cells was detected after treatment and the apoptosis rate with the concentration and longer incubation time (r=1.0, p<0.01), while the percentage of cells in S phase and $G_2$/M phase decreased significantly. Immunocytochemistry assay showed that the expression of cyclin E and bcl-2 in the treated cells significantly decreased, while the expression of p27 and bax obviously increased, compared with the control group (p<0.05). The results of our research indicate that inotodiol isolated from Inonotus obliquus inhibited the proliferation of HeLa cells and induced apoptosis in vitro. The mechanisms may be related to promoting apoptosis through increasing the expression of bax and cutting bcl-2 and affecting the cell cycle by down-regulation the expression of cyclin E and up-regulation of p27. The results further indicate the potential value of inotodiol for treatment of human cervical cancer.

        Long non-coding RNAs in Sus scrofa ileum under starvation stress

        Objective: In this study, we aimed to identify long non-coding RNAs (lncRNAs) that play important roles in starvation stress, analyze their functions, and discover potential molecular targets to alleviate starvation stress to provide a theoretical reference for subsequent in-depth research. Methods: We generated a piglet starvation stress animal model. Nine Yorkshire weaned piglets were randomly divided into a long-term starvation stress group (starved for 72 h), short-term starvation stress group (starved for 48 h), and the control group. LncRNA libraries were constructed using high-throughput sequencing of piglet ileums. Results: We obtained 11,792 lncRNAs, among which, 2,500 lncRNAs were novel. In total, 509 differentially expressed (DE)lncRNAs were identified in this study. Target genes of DElncRNAs were predicted via cis and trans interactions, and functional and pathway analyses were performed. Gene ontology functions and Kyoto encyclopedia of genes and genomes analysis revealed that lncRNA-targeted genes mainly participated in metabolic pathways, cellular processes, immune system processes, digestive systems, and transport activities. To reveal the mechanism underlying starvation stress, the interaction network between lncRNAs and their targets was constructed based on 26 DElncRNAs and 72 DEmRNAs. We performed an interaction network analysis of 121 DElncRNA–DEmRNA pairs with a Pearson correlation coefficient greater than 0.99. Conclusion: We found that MSTRG.19894.13, MSTRG.16726.3, and MSTRG.12176.1 might play important roles in starvation stress. This study not only generated a library of enriched lncRNAs in piglets, but its outcomes also provide a strong foundation to screen key lncRNAs involved in starvation stress and a reference for subsequent in-depth research.         DC-Link Capacitor Voltage Balanced Modulation Strategy Based on Three-Level Neutral-Point-Clamped Cascaded Rectifiers

        This study proposes a new modulation strategy to deal with unbalanced output voltage that is based on three-level neutral-point-clamped cascaded rectifiers. The fundament idea is to reallocate the value of the voltage levels generated by each of the modules on the basis of space vector pulse width modulation. This proposed modulation strategy can reduce the switching frequency while maintaining the mutual-module voltage balance. First, an analysis of unbalanced output voltage is reflected. Then a new modulation strategy is introduced in detail. Internal module capacitor voltages are balanced by the selection of redundant vectors. Moreover, the voltage balance ability is calculated. Finally, the feasibility of this modulation strategy is verified through experimental results.

        Liver cancer is one of leading digestive malignancies with high morbidity and mortality. There is an urgent need for the development of novel therapies for this deadly disease. It has been proven that asparagus polysaccharide, one of the most active derivates from the traditional medicine asparagus, possesses notable antitumor properties. However, little is known about the efficacy of asparagus polysaccharide as an adjuvant for liver cancer chemotherapy. Herein, we reported that asparagus polysaccharide and its embolic agent form, asparagus gum, significantly inhibited liver tumor growth with transcatheter arterial chemoembolization (TACE) therapy in an orthotopic hepatocellular carcinoma (HCC) tumor model, while significantly inhibiting angiogenesis and promoting tumor cell apoptosis. Moreover, asparagine gelatinous possessed immunomodulatory functions and showed little toxicity to the host. These results highlight the chemotherapeutic potential of asparagus polysaccharide and warrant a future focus on development as novel chemotherapeutic agent for liver cancer TACE therapy.
