        Genome-wide comparative analysis of the codon usage patterns in plants

        Codon usage analysis has been a classical area of study for decades and is important for evolution, mRNA translation, and new gene discovery. Recently, genome sequencing has made it possible to perform studies of the entire genome in plant kingdoms. The base composition of the coding sequence, codon usage pattern, codon pairs, and related indicators of relative synonymous codon usage (RSCU), including the Fop, Nc, RSCU, CAI and GC contents, were analyzed. We found that the GC content of single-celled algae is the highest, whereas dicotyledons are the lowest. Moreover, the base composition of plants is similar within the same family. In addition, the GC content of the second base of the codon is lower than the first and third base. In conclusion, the codon usage characteristics are opposite in Gramineae, single-celled algae, fern and dicotyledon, moss, and Pinaceae. Furthermore, the degree of codon usage bias is decreasing with evolution. Therefore, we hypothesize that the lower the plants, the more that they must optimize codons and that higher plants no longer need to optimize codons.

        Extraction of bioethanol from fermented sweet sorghum bagasse by batch distillation

        Extraction of bioethanol, a potential alternative to fossil fuel in the transport industry, from sweet sorghum stems [Sorghum bicolor (L.) Moench] using solid-state fermentation (SSF) technology has become a popular research topic worldwide. Because SSF technology can directly convert fermentable sugars into target products without juice squeezing and water input, this method can potentially reduce energy and water consumption. However, ethanol extraction from fermented sweet sorghum bagasse requires further investigation. We used batch solid-state distillation to investigate the optimal operating parameters in a distillation column (diameter, 400 mm) via a single-factor experiment. Results showed that the optimal steam flow rate and loading height were 8-10 kg·h−1 and 700-1,000 mm, respectively. Under optimal conditions, an energy consumption of 3.82 tons of steam per ton of ethanol and distillate concentration of 60.9% (v/v) were obtained. The pseudo-first-order rate equation was used to describe the distillation kinetics, and good correlations were obtained. Therefore, solid-state distillation can be effectively used to extract ethanol from fermented sweet sorghum bagasse.

        Extraction of Pigment from Sugarcane Juice Alcohol Wastewater and Evaluation of Its Antioxidant and Free Radical Scavenging Activities

        Pigment was extracted to decolor the sugarcane juice alcohol wastewater and its antioxidant activities were evaluated. The pigment was extracted using ion exchange fiber along with macroporous resin D-101 as the absorbent and an ethanol-water mixture (6:4, v/v) as the eluent. Total flavonoid and phenolic contents of the pigment were determined. DPPH radical scavenging ability and reducing power of 1.0 mg/mL of the pigment were approximately 78.5 and 16.8%, respectively, compared to 1.0 mg/mL of the tea polyphenol (positive control). The hydroxyl radical scavenging ability of 10.0 mg/mL of the pigment was about 92.5% compared to that of positive control. The DPPH radical scavenging ability of the pigment was enhanced significantly with thermal treatment at 70 and 100oC. The result shows the proposed procedure can be effective to decolor such wastewater and to get antioxidative pigment.
