Based on the reported cDNA sequences of $BmK{\alpha}Txs$, the genes encoding toxin $BmK{\alpha}Tx11$ and $BmK{\alpha}Tx15$ were amplified by PCR from the Chinese scorpion Buthus martensii Karsch genomic DNA employing synthetic oligonucleotides. Sequences analysis of nucleotide showed that an intron about 500 bp length interrupts signal peptide coding regions of $BmK{\alpha}Tx11$ and $BmK{\alpha}Tx15$. Using cDNA sequence of $BmK{\alpha}Tx11$ as probe, southern hybridization of BmK genome total DNA was performed. The result indicates that $BmK{\alpha}Tx11$ is multicopy genes or belongs to multiple gene family with high homology genes. The similarity of $BmK{\alpha}$-toxin gene sequences and southern hybridization revealed the evolution trace of $BmK{\alpha}$-toxins: $BmK{\alpha}$-toxin genes evolve from a common progenitor, and the genes diversity is associated with a process of locus duplication and gene divergence.

We adopt the PASTEL catalog combined with SIMBAD radial velocities as a testing standard to validate the stellar parameters (effective temperature $T_{eff}$, surface gravity log g, metallicity [Fe/H] and radial velocity $V_r$) from the first data release (DR1) of The Large Sky Area Multi-Object Fiber Spectroscopic Telescope (LAMOST) survey. After applying data reduction and temperature constraints to the sample obtained by cross-identification, we compare the stellar parameters from DR1 and PASTEL. The results show that the DR1 results are reliable under certain conditions. We derive a dispersion of 110 K, 0.19 dex, 0.11 dex and $4.91kms^{-1}$ in specified effective temperature ranges, for $T_{eff}$, log g, [Fe/H] and $V_r$ respectively. Systematic errors are negligible except for those of $V_r$. In addition, for stars with PASTEL [Fe/H] < -1:5, the metallicities in DR1 are systematically higher than those in PASTEL.

In this research, the reactive radical species in photocatalytic activities of a composite photocatalyst made fromPET filaments loaded with Ag-TiO2 nanoparticles under visible light irradiation, in comparison with those of as-modifiedAg-TiO2 nanoparticles, were reported and its photocatalytic mechanism was investigated. The PET filaments were modifiedby using tetrabutyl titanate as the TiO2 precursor and silver nitrate as the dopant in a hydrothermal process. The Ag decoratedTiO2 nanoparticles were synthesized and deposited on the surfaces of PET filaments in the hydrothermal process at 120 oCfor 180 min. The morphology, phase structure, chemical binding state, and optical properties of the PET-Ag-TiO2nanoparticles composites were systemically studied by means of field emission scanning electron microscopy (FESEM), X-raydiffraction (XRD), transmission electron microscopy (TEM), X-ray photoelectron spectroscopy (XPS), photoluminescence (PL)spectroscopy, diffuse reflectance spectroscopy (DRS), and ultraviolet photoelectron spectroscopy (UPS) techniques. Thephotocatalytic activities of the PET-Ag-TiO2 nanoparticles composites were evaluated in the photodegradation of bothmethylene blue (MB) and methyl orange (MO) dyes under visible light irradiation. It was found that, unlike in the asobtainedTiO2 nanoparticles and Ag decorated TiO2 nanoparticles, the photoinduced holes (h+) were the major reactiveradical species in both PET-Ag-TiO2 composite photocatalyst and the PET filaments loaded with TiO2 nanoparticles in theMB photodegradation process. The experimental results also indicated that the PET-Ag-TiO2 nanoparticles composites led tothe improvement of the separation efficiency of photogenerated electron-hole pairs. The enhanced photocatalytic activity ofthe PET filaments coated with Ag decorated TiO2 nanoparticles was ascribed to both the incorporation of Ag nanoparticlesinto TiO2 nanoparticles and the possible infiltration of Ag/Ti nanoparticles into PET polymers. In addition, the wavelengthand intensity of monochromatic light had great influences on the photodegradation rate of dye used, which was closelycorrelated with the maximum absorption wavelength of the dye to be degraded.
Enhanced Allergic Inflammation of Der p 2 Affected by Polymorphisms of MD-2 Promoter

Purpose: Myeloid differentiation-2 (MD-2) has been associated with endotoxin and inflammatory disorders because it can recognize lipopolysaccharide (LPS) binding and attenuate Toll-like receptor 4 (TLR4)-mediated signaling. However, its role in allergic inflammation has yet to be clarified. We examined whether single nucleotide polymorphisms (SNPs) in MD-2 promoter can affect MD-2 expression and aimed to clarify the relationship between Der p 2 allergy and SNPs of MD-2 promoter. Methods: The function of SNPs of MD-2 promoter and the effects of cytokines and immunoglobulin on the secretion and mRNA expression were investigated in 73 allergic subjects with different MD-2 gene promoter variants. Peripheral blood mononuclear cells were cultured with or without LPS in the presence of Dermatophagoides pteronyssinus group 2 allergen (Der p 2), followed by mRNA extraction and cytokine expression analysis. The culture supernatants were collected for cytokine measurement. Results: Patients with the MD-2 promoter SNPs (rs1809441/rs1809442) had increased mRNA expressions of MD-2, ε heavy chain of IgE (Cε), and interleukin (IL)-8; however, only MD-2 and IL-8 were further up-regulated after Der p 2 stimulation. Patients with SNPs of MD-2 promoter tended to have high levels of IL- 1β, IL-6, IL-8, IL-10, and tumor necrosis factor (TNF)-α after Der p 2 and LPS stimulation. Increased secretions of IL-6, IL-8, and IL-10 were found to be up-regulated by Der p 2 stimulation, and an increased secretion of IFN-γ and decreased secretion of IL-4 were noted after LPS stimulation. Conclusions: The high levels of proinflammatory cytokines secreted by Der p 2 were predetermined by MD-2 promoter SNPs (rs1809441/rs1809442). Through cytokine secretion by Der p 2 and LPS, these SNPs may serve as an indicator of the pathological phenotype of Der p 2-induced allergic inflammation.
