      • <i>Sophora flavescens</i> Aiton Decreases MPP <sup>+</sup> -Induced Mitochondrial Dysfunction in SH-SY5Y Cells

        <P><I>Sophora flavescens</I> Aiton (SF) has been used to treat various diseases including fever and inflammation in China, South Korea and Japan. Several recent reports have shown that SF has anti-inflammatory and anti-apoptotic effects, indicating that it is a promising candidate for treatment of Parkinson’s disease (PD). We evaluated the protective effect of SF against neurotoxin 1-methyl-4-phenylpyridinium ion (MPP<SUP>+</SUP>)-induced mitochondrial dysfunction in SH-SY5Y human neuroblastoma cells, an <I>in vitro</I> PD model. SH-SY5Y cells were incubated with SF for 24 h, after which they were treated with MPP<SUP>+</SUP>. MPP<SUP>+</SUP>-induced cytotoxicity and apoptosis were confirmed by 3-(4,5-dimethyl-thiazol-2-yl)-2,5-diphenyl tetrazolium bromide assay and terminal deoxynucleotidyl transferase-mediated biotinylated UTP nick end labeling assay. MitoSOX red mitochondrial superoxide indicator, tetramethylrhodamine methyl ester perchlorate and Parkin, PTEN-induced putative kinase 1 (PINK1), and DJ-1 immunofluorescent staining were conducted to confirm the mitochondrial function. In addition, western blot was performed to evaluate apoptosis factors (Bcl-2, Bax, caspase-3 and cytochrome c) and mitochondrial function-related factors (Parkin, PINK1 and DJ-1). SF suppressed MPP<SUP>+</SUP>-induced cytotoxicity, apoptosis and collapse of mitochondrial membrane potential by inhibiting the increase of reactive oxidative species (ROS) and DNA fragmentation, and controlling Bcl-2, Bax, caspase-3 and cytochrome <I>c</I> expression. Moreover, it attenuated Parkin, PINK1 and DJ-1 expression from MPP<SUP>+</SUP>-induced decrease. SF effectively suppressed MPP<SUP>+</SUP>-induced cytotoxicity, apoptosis and mitochondrial dysfunction by regulating generation of ROS, disruption of mitochondrial membrane potential, mitochondria-dependent apoptosis and loss or mutation of mitochondria-related PD markers including Parkin, PINK1 and DJ-1.</P>

        Proteomic change by Korean Red Ginseng in the substantia nigra of a Parkinson's disease mouse model

        Background: Recent studies have shown that Korean Red Ginseng (KRG) successfully protects against dopaminergic neuronal death in the nigrostriatal pathway of a Parkinson's disease (PD) mouse model induced by 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) administration; however, the mechanism has yet to be identified. Therefore, in this study we used two-dimensional electrophoresis to investigate the effects of KRG on the changes in protein expression in the substantia nigra (SN) of MPTP-treated mice. Methods: Male C57BL/6 mice (9 wk old) were intraperitoneally administered MPTP (20 mg/kg) four times at 2-h intervals, after which KRG (100 mg/kg) was orally administered once a day for 5 d. Two hours after the fifth KRG administration, a pole test was conducted to evaluate motor function, after which the brains were immediately collected. Survival of dopaminergic neurons was measured by immunohistochemistry, and protein expression was measured by two-dimensional electrophoresis and Western blotting. Results: KRG alleviated MPTP-induced behavioral dysfunction and neuronal toxicity in the SN. Additionally, the expression of eight proteins related to neuronal formation and energy metabolism for survival were shown to have changed significantly in response to MPTP treatment or KRG administration. KRG alleviated the downregulated protein expression following MPTP administration, indicating that it may enhance neuronal development and survival in the SN of MPTP-treated mice. Conclusion: These findings indicate that KRG may have therapeutic potential for the treatment of patients with PD.

        Understanding the light soaking effect of ZnMgO buffer in CIGS solar cells

        <P>This study investigated the mechanism underlying the light soaking effect of a ZnMgO buffer in Cu(In,Ga)Se<SUB>2</SUB> (CIGS) solar cells, where the cell efficiency increased with an increase of light soaking time. The ZnMgO buffer layer was deposited by an atomic layer deposition method. With light soaking, the cell efficiency of ZnMgO/CIGS cells increased mainly by the increase of the fill factor and partly by the increase of the open-circuit voltage. With light soaking, the electron carrier concentration of the ZnMgO layer increased and the XPS intensity of the hydroxyl bond in the ZnMgO layer decreased. Based on the above results and the comparison of other buffers in literature, we assumed that the hydrogen atoms broken away from the hydroxyl bond by photon irradiation occupied the interstitial sites of the ZnMgO layer as a donor atom and also passivated the defects at the ZnMgO/CIGS interface.       • Acupuncture Resolves Persistent Pain and Neuroinflammation in a Mouse Model of Chronic Overlapping Pain Conditions

        <P><B>Abstract</B></P> <P>Patients with chronic overlapping pain conditions have decreased levels of catechol-O-methyltransferase (COMT), an enzyme that metabolizes catecholamines. Consistent with clinical syndromes, we previously demonstrated that COMT inhibition in rodents produces persistent pain and heightened immune responses. Here, we sought to determine the efficacy of manual acupuncture in resolving persistent pain and neuroinflammation in the classic inbred C57BL/6 strain and the rapid-wound healing MRL/MpJ strain. Mice received subcutaneous osmotic minipumps to deliver the COMT inhibitor OR486 or vehicle for 13 days. On day 7 after pump implantation, acupuncture was performed at the Zusanli (ST36) point or a non-acupoint for 6 consecutive days. Behavioral responses to mechanical stimuli were measured throughout the experiment. Immunohistochemical analysis of spinal phosphorylated p38 mitogen-activated protein kinase, a marker of inflammation, and glial fibrillary acidic protein, a marker of astrogliosis, was performed on day 13. Results demonstrated that ST36, but not sham, acupuncture resolved mechanical hypersensitivity and reduced OR486-dependent increases in phosphorylated p38 and glial fibrillary acidic protein in both strains. The magnitude of the analgesic response was greater in MRL/MpJ mice. These findings indicate acupuncture as an effective treatment for persistent pain linked to abnormalities in catecholamine signaling and, furthermore, that analgesic efficacy may be influenced by genetic differences.</P> <P><B>Perspective</B></P> <P>Chronic overlapping pain conditions remain ineffectively managed by conventional pharmacotherapies. Here, we demonstrate that acupuncture alleviates persistent pain and neuroinflammation linked to heightened catecholaminergic tone. Mice with superior healing capacity exhibit greater analgesic efficacy. Findings indicate acupuncture as an effective treatment for chronic overlapping pain conditions and provide insight into treatment response variability.</P> <P><B>Highlights</B></P> <P> <UL> <LI> Sustained inhibition of catechol-O-methyltransferase led to persistent pain and neuroinflammation in mice. </LI> <LI> ST36, but not sham, acupuncture alleviated pain. </LI> <LI> ST36 acupuncture reduced spinal phosphorylated p38 expression and astrocyte activation. </LI> <LI> The magnitude of the analgesic response was greater in rapid-healing MRL/MpJ mice. </LI> </UL> </P>

        A Study on the Mediating Effect of Wage Level in the Relationship between Education Level and Self-Efficacy of Disabled People

        본 연구는 장애인의 교육수준과 자기효능감과의 관계에서 임금수준의 매개효과를 검증하는 것을 목적으로 한다. 이를 위하여 장애인고용패널조사 2차 웨이브 3차자료를 활용하여 분석하였으며, 회귀분석과 Sobel test를 실시하였다. 본 연구의 연구결과는 다음과 같다. 첫째, 교육수준이 높을수록 임금수준이 높은 것으로 나타났다. 둘째, 교육수준이 높을수록 자기효능감이 높은 것으로 나타났다. 셋째, 장애인의 임금수준이 장애인의 교육수준과 자기효능감 사이를 매개하는 것으로 나타났다. 이러한 연구결과를 바탕으로 본 연구에서는 다음과 같은 제언을 하였다. 첫째, 진학 과정에서 장애인에 대한 차별을 줄여야 하며, 현행 수능에 중점을 두고 있는 교육제도를 개선해야할 필요성이 있다. 둘째, 장애인의 교육환경에 대한 지속적인 개선이 필요하다. 셋째, 장애인의 임금수준 향상을 위한 제도적 지원이 필요하다. The purpose of this study is to verify the mediating effect of wage level in the relationship between education level and self-efficacy of disabled people. To this end, this study analyzed the Panel Survey of Employment for the Disabled (PSED) 2nd wave 3rd data, and conducted Regression analysis and Sobel test. The research results of this study are as follows. First, higher education level leads higher wage level. Second, higher education level leads higher self-efficacy. Third, The wage level of disabled people was found to mediate between education level and self-efficacy of disabled people. Based on these research results, this study made the following suggestions. First, there are needs to reduce discrimination against disabled people in the process of entering colleges and universities and to improve the education system which focuses on the College Scholastic Ability Test(CSAT). Second, it is necessary to continuously improve the educational environment of disabled people. Third, institutional support is needed to improve the wage level of disabled people.

      • Biodiversity of Arthropods between GM and non-GM Rice Field

        Biodiversity of arthropods in Cnaphalocrocis medinalis (Lepidoptera: Crambidae) resistant GM rice (CryIAc1) and non-GM rice fields. Sampling was conducted 15 times using sweeping net and electric aspirator. Biodiversity was analyzed with species richness and Shannon diversity index (H’). Total 28,275 arthropod individuals (12,413 in GM rice and 15,862 individuals in non-GM rice) were collected and there were 22 families, 34 genera and 36 species belonging to 8 orders. There were 19 families, 29 genera, 31 species in GM rice and 20 families 32 genera 34 species in non-GM rice fields. There was no significant difference in species richness and species diversity (H’) between GM and non-GM rice. Species diversity was significantly higher in non GM rice in the insect pest group only in middle and late of August .
