      • Dynamic Structure of Bacteriorhodopsin Revealed by $^{13}C$ Solid-state NMR

        We demonstrate here a dynamic structure of bacteriorhodopsin (bR) as revealed by $^{13}$ C NMR studies on [3_$^{13}$ C]_,[1-$^{13}$ C]Ala- and/or Val-labeled wild type and a variety of site-directed mutants at ambient temperature. For this purpose, well-resolved (up to twelve) I$^{13}$ C NMR peaks were assigned with reference to the displacement of peaks due to the conformation-dependent I$^{13}$ C chemical shifts and reduced peak-intensities due to site-directed mutations. Revealed bR structure was not rigid as anticipated from 2D crystals of hexagonal array but a dynamically heterogeneous, undergoing a variety of local fluctuations depending upon specific site with frequency range of 10$^2$ -10$^{8}$ Hz. In particular, dynamics- dependent suppression of peaks turned out to be very sensitive to the motion of 10$^{-4}$ s and 10$^{-5}$ s interfered with frequency of magic angle spinning and proton decoupling, respectively. It is also noteworthy that such dynamic feature is strongly dependent upon the manner of 2D crystalline packing: $^{13}$ C NMR peaks of monomeric bR yielded either highly broadened or completely suppressed signals, depending upon the type of $^{13}$ C-labeled amino-acid residues.

      • Influence of Growth and Yield by Diffrence of Seeding Accuracy and Depth on Green Soybeans Cultivation used Up-cut Type Ridging and Mulching Seeder

        A cultivation outline of green soybeans could not set mechanical seeding technics to have difference of mulched soil temperature at any seeding season. Especially, the early cropping type of green soybeans seeding that used mulcting method to get high emergence rate on low temperature season have been high labor load to seed by manual operation. Therefore, the purpose of this study was to develop high efficiency and precision mechanical seeding technique which have ridging and mulching same time. We developed prototype seeding machine combined with up-cut rotary, mulching seeder and the molding implement. After that, we made control device to move diching device for up and down direction. We investigated the seeding accuracy and seeding depth of the prototype seeder, and evaluated the growth and yield, respectively. As a result, it is showed that the seeding accuracy is as same as manual seeding. Although the seeding depth was shallow compared to the conventional manual seeding, it can increase seeding depth than conventional seeding. However, the measured seeding depths were shallower than the set values. This is reason why was influenced of soil compression, posture of the seeding part and covered soil after seeding. The plant length at harvest time might become longer as the seeding depth got deeper. So, we considered that it is possible to do multiple simultaneous work used up-cut ridging and mulching seeder.

      • Noninvasive diagnosis of NAFLD

        Nonalcoholic fatty liver disease (NAFLD) and nonalcoholic steatohepatitis (NASH) is an important cause of chronic liver injury in many countries around the world. The degree of liver fibrosis must be estimated to determine the prognosis, surveillance, and optimal treatment for NAFLD/NASH, similar to the situation for other liver diseases. Liver biopsy remains the gold standard for evaluating the degree of hepatic fibrosis. However, liver biopsy is an invasive and expensive method and is associated with a relatively high risk of complications. Furthermore it is impossible to enforce liver biopsy in all NAFLD patients, because the number of NAFLD patients has reached 80-100 million in the U.S. and about 5-10 million NAFLD patients are estimated to exist even in Korea. Thus, a rapid and noninvasive method of detecting fibrosis in patients with NAFLD is of major clinical interest. Several noninvasive investigations, such as serum biomarkers, scoring systems, imaging method have been developed to establish the diagnosis of NASH. First, we validated scoring systems (FIB4 index, AST/ALT ratio, AST to platelet ratio, Age-platelet index, NAFLD fibrosis score, BARD score) in biopsy-proven Asian population (n=576). Next, we investigated the clinical usefulness of ultrasound-based elastography such as transientelastography (Fibroscan) and Acoustic Radiation Force Impulse (ARFI) Elastography for detecting the advanced fibrosis in NAFLD patients. The major advantages of transient elastography and ARFI elastography, as compared with liver biopsy, are that these techniques are painless, rapid, and have no associated complications, and are, therefore, very easily accepted by the patients. It is not uncommon for patients to present with the complications of previously unrecognized cirrhosis despite being under long-standing medical care, because these patients often do not manifest the classic physical changes associated with cirrhosis. By using these noninvasive scoring systems and/or imaging methods, efficient recruitment of NASH patients with advanced stage may be facilitated.

        Interaction of NpO+2 with Cl- in Na-Ca-Cl-type solutions at ionic strength of 6M: Effect of presence of Ca ion on interaction

        The interaction of NpO+ 2 with Cl- was studied using visibleenear-infrared spectroscopy in NaCl-Ca- Cl2-NaClO4, NaCl-NaClO4, and CaCl2-NaClO4 solutions with ionic strength (I) of 6M. The spectra of NpO+2 around 980 nm varied with Cl- concentration in the NaCl-CaCl2-NaClO4 and NaCl-NaClO4 solutions at [Cle] ≥ 3.5M, but not in the CaCl2-NaClO4 solution. Assuming the 1:1 interaction between NpOþ+ and Cl-, the apparent equilibrium constants at I = 6M were evaluated. The presence of Ca2+ was found to destabilize overall interaction between NpOþ 2 and Cle. The observations were consistent with the density functional theory calculation.

        Preclinical Efficacy and Clinical Feasibility of a Novel Aerosol- Exposure Protection Mask for Esophagogastroduodenoscopy

        Background/Aims: This study aimed to assess the efficacy of a novel aerosol-exposure protection (AP) mask in preventingcoronavirus disease in healthcare professionals during upper gastrointestinal endoscopy and to evaluate its clinical feasibility. Methods: In Study 1, three healthy volunteers volitionally coughed with and without the AP mask in a cleanroom. Microparticleswere visualized and counted with a specific measurement system and compared with and without the AP mask. In Study 2,30 patients underwent endoscopic resection with the AP mask covering the face, and the SpO2 was measured throughout theprocedure. Results: In Study 1, the median number of microparticles in volunteers 1, 2, and 3 with and without the AP mask was 8.5 and 110.0,7.0 and 51.5, and 8.0 and 95.0, respectively (p<0.01). Using the AP mask, microparticles were reduced by approximately 92%. Themedian distances of microparticle scattering without the AP mask were 60, 0, and 68 in volunteers 1, 2, and 3, respectively. In Study 2,the mean SpO2 was 96.3%, and desaturation occurred in three patients. Conclusion: The AP mask could provide protection from aerosol exposure and can be safely used for endoscopy in clinical practice.

        Complications of endoscopic resection in the upper gastrointestinal tract

        Endoscopic resection (ER) is widely utilized as a minimally invasive treatment for upper gastrointestinal tumors; however, complications could occur during and after the procedure. Post-ER mucosal defect leads to delayed perforation and bleeding; therefore, endoscopic closure methods (endoscopic hand-suturing, the endoloop and endoclip closure method, and over-the-scope clip method) and tissue shielding methods (polyglycolic acid sheets and fibrin glue) are developed to prevent these complications. During duodenal ER, complete closure of the mucosal defect significantly reduces delayed bleeding and should be performed. An extensive mucosal defect that comprises three-quarters of the circumference in the esophagus, gastric antrum, or cardia is a significant risk factor for post-ER stricture. Steroid therapy is considered the first-line option for the prevention of esophageal stricture, but its efficacy for gastric stricture remains unclear. Methods for the prevention and management of ER-related complications in the esophagus, stomach, and duodenum differ according to the organ; therefore, endoscopists should be familiar with ways of preventing and managing organ-specific complications.
