A Thirteen-week Oral Dose Subchronic Toxicity Study of Isaria sinclairii in Rats
Mi Young Ahn,Jea Woong Han,Sang Deok Jee,Jae Sam Hwang,Seok Jo Hwang,Yoo Na Hong,Sung Nam Kim 한국독성학회 2007 Toxicological Research Vol.23 No.2
Isaria sinclairii (IS) was orally administered at doses of 0, 0.04, 0.2, and 1 g/㎏/day over a 13-week period. There were no observed clinical signs or deaths related to treatment in all the groups tested. Therefore, the approximate lethal oral dose of I. sinclairii was considered to be higher than 1 g/kg in rats. Throughout the administration periods, no significant changes in diet consumption, ophthalmologic findings, organ weight, clinical pathology (hematology, clinical chemistry, coagulation, and urinalysis) or gross pathology were detected. Minor changes were found in hematological parameters for the 0.04 g/㎏/day and 0.2 g/㎏/day IS treated groups (triglyceride reductions of 20.1~46.6% and platelet increases), but all changes were within physiological range. Microscopic examination failed to identify any treatment-related histopathologic changes in the organs of the IS-treated rats other than nuclear enlargement (cellular atypia) of the tubular regions in the medulla of the kidney in the high dose group. From these results, one can conclude that the no-observed effect level (NOAEL) of I. sinclarii is less than 0.04 g/㎏/day in rats.

The aim of the present work was to investigate the effect of NaCl concentration on the emulsifying and rheological properties of porcine myofibrillar protein (MF)-stabilized soybean oil and fish oil emulsion (SO-EMs and FO-EMs). Emulsions (EMs) were prepared from 1% MF with 10% SO or FO at various NaCl concentration (0-0.5 M). The emulsifying ability index (EAI) of the EMs increased with increasing NaCl concentration for both oil types. Conversely, increasing NaCl manifested decrease in the emulsion stability index (ESI). In addition, creaming index (CI) also increased with NaCl concentration. From the microscopic observation, droplets of the EMs were more aggregated at relatively higher NaCl concentrations, especially for FO-EMs. All EMs had a gel-like structure owing to G' > G" from the rheological analysis. Comparing the oil types, the emulsifying capacity of SO-EMs was more stable than that of FO-EMs at all NaCl concentrations as determined from the CI value and microscopic observation. Therefore, it can be concluded that SO-EMs and FO-EMs are more stable at relatively lower concentrations of NaCl. In addition, the dispersed stability of SO-EMs was better than that of FO-EMs at the same concentration of NaCl.

This study was conducted to evaluate the inhibitory effects of Yongseollan(YSL) on the production of nitric oxide (NO) and the expression of inducible nitric oxide synthase (iNOS) in lipopolysaccharide (LPS)-activated RAW264.7 cells. YSL is tropical plant originating from Mexico. The biological activity of this plant is not yet evaluated systematically. The aim of the present work is to investigate a potential anti-inflammatory activity of YSL. The RAW264.7 cells were cultured in D MEM/F12 medium for 24 hrs. After serum starvation, cells were treated with YSL for 1 hr, followed by stimulating NO production with a LPS. We found that YSL has an inhibitory effect on the production of NO, iNOS expression and $phospho-I{\kappa}B$ expression. YSL also inhibited tumor necrosis factor $(TNF)-{\alpha}$, interleukin (IL)-6, and $IL-1{\beta}$. Moreover, YSL inhibited cyclooxygenase (COX)-2 expression and prostanglandin E2 (PGE2). These findings showed that YSL could have some anti-inflammatory effects which might play a role in therapy in Gram-negative bacterial infections.

Background: The most widely used circulating tumor cell (CTC) isolation techniques rely on antibody-based capture of CTCs, which express epithelial cell surface markers that are absent from normal leucocytes. Among these, epithelial cell adhesion molecule (EpCAM) is most commonly used. Although this platform is the most standardized of any current technology, it suffers from relatively low sensitivity. Survivin is a member of the inhibitor of apoptosis protein gene family that is highly expressed in most malignancies. Several studies have shown the prognostic value of survivin in various cancers, including lung cancer. But the diagnostic significance of survivin for non-small cell lung cancer (NSCLC) remains controversial. The purpose of this prospective study was to evaluate the diagnostic value of survivin-expressing CTCs in peripheral blood of patients with NSCLC. Methods: Blood samples were collected from patients with NSCLC before treatment and healthy volunteers. The EpCAMand survivin- expressing CTCs were detected by real-time quantitative PCR. Results: To date, 43 patients with NSCLC stages I-IV and 15 healthy controls, all aged 52-79 years were enrolled in the study. Survivin mRNA was detected in the CTCs. Survivin- expressing CTCs were upregulated with more than 2-fold difference as compared with controls (p=0.004). EpCAM was not significantly different between NSCLC patients and controls (p=0.409). No correlation between the survivin-expressing CTCs levels and the stage of disease and histology can be made at this point. Conclusions: We demonstrated the significant difference in the levels of survivin-expressing CTCs between NSCLC patients and controls. These results suggest that survivin might be useful molecular marker for CTCs.

Background: Normal thyroid function influences basal metabolic rate and general body metabolism and thyroid dysfunction is often associated with dyslipidemia and increased cardiovascular risk in general polulation. It is known that about 25% of patients with end-stage renal disease(ESRD) on hemodialysis had subclinical thyroid dysfunction. We investigated that subclinical hypothyroid dysfunction associate with dyslipidemia and cardiovascular event in hemodialysis patients. Hence, we conducted our study to elucidate whether subclinical thyroid dysfunction treated or not in ESRD patients. Methods: All hemodialysis patients in Red Cross Hospital within a period of one year were included in the study. Thyroxine-3 (T3), free thyroxine- 4 fT4, Thyroid Stimulating Hormone (TSH) and lipid profiles in the serum were estimated and death from all causes and CV events served as outcome variables over a median follow-up period of 12 months. Participants were divided into groups based on the thyroid function. Statistical analysis was carried out by using SPSS. Results: Among the 92 cases, 28.2% of the patients were having thyroid disorder. In which 2.1% were of hypothyroid, 13.7% were of subclinical hypothyroid, and 0.8% were of hyperthyroid, 11.6% were of subclinical hyperthyroid. We observed negative correlation between TC & fT3. Total cholesterol was raised in hypothyroidism in comparison to euthyroid ESRD patients. However, thyoroid function didn’t have significant association with lipid profiles. Cardiovascular disease significantly often occurred in subclinical thyroidism patients than other group Conclusions: Our results show that the blood thyroid hormone level isn’t affected by dyslipidemia but affected by occurrence of CVD. Therefore, the management of subclinical hypothyroidism in hemodialysis patients should be considered. Keywords: Thyroid function; Dyslipidemia; Cardiovascular events; End stage renal disease (ESRD)
