        원발성 간암에서 PIVKA-II 및 Lens Culinaris Agglutinin-A 반응성 Alpha-fetoprotein(AFP-L3)의 임상적 유용성

        Background/Aims: Des-γ-carboxy prothrombin(DCP), a protein induced by vitamin K absence or antagonist-II(PIVKA-II), and Lens culinaris agglutinin-A reactive AFP-L3 have been reported to be useful markers for the diagnosis of hepatocellular carcinoma (HCC). In the present study, both the PIVKA-II and AFP-L3 were analyzed and compared with a conventional AFP to determine its usefulness, specificity and sensitivity in the diagnosis of HCC. Methods: Sera were collected from 108 patients consisting of 17 patients with chronic hepatitis, 22 patients with liver cirrhosis and 69 patients with HCC. The AFP-L3 was determined by an lectin affinity electrophoresis coupled with an antibody-affinity blotting. Level of DCP was measured by an enzyme immunoassay with an anti-DCP monoclonal antibody. Results: The sensitivity and specificity of PIVKA-II and AFP L3 were 49.3% and 89.5%, and 32.5% and 85.7%, respectively. No significant correlation was found between the PIVKA-II or AFP L3 and serum AFP. No correlation was found etween the PIVKA-II or AFP L3 and the characteristics of HCC.       • Development of Strip Casting Process for Fabrication of Wrought Mg Alloys

        <P>AZ31 and experimental ZMA611 alloys were strip cast into 2 mm thick strips. The as-cast AZ31 alloy strip consists of columnar dendrites. On the other hand, as-cast ZMA611 alloy strip shows equiaxed dendritic structure through the thickness of strip. Strip cast AZ31 alloy in H24 condition has equivalent yield and tensile strengths with similar ductility compared to commercial ingot cast AZ31-H24 alloy, indicating that strip casting is a viable process for the fabrication of Mg alloy strips. The ZMA611 alloy has a large volume fraction of fine dispersoid particles in the microstructure, resulting from the beneficial effect of strip casting on microstructural refinement. It has been shown that the ZMA611 alloy has superior tensile properties compared to commercial ingot cast AZ31-H24 alloy, suggesting the possibility of the development of new wrought Mg alloy sheets by strip casting.</P>

        스케일이 있는 급수관내의 유도초음파의 전파 특성에 관한 연구

        배관 내의 스케일은 통수비를 감소시키므로 배관을 적절히 관리하기 위해서는 스케일량에 대한 정량적인 평가가 필수적이다. 상수도 급수관내의 스케일량을 평가하기 위해서 유도초음파를 사용하였다. 가변각 웻지를 사용하여 유도 초음파의 여러 모드를 발생시키고, 노후 배관을 따라서 전파하는 특성을 분석하였다. 실험적으로 관내의 스케일이 증가함에 따라서 f×d=1.5㎒·㎜인 경우에 F(M,2) 모드들의 진폭이 크게 감소하는 것을 알 수 있었다. 본 연구의 결과로 상수도 급수관의 스케일량 평가에 적합한 유도 초음파는 F(M,2) 모드들임을 알 수 있었다. Since the scale in pipes reduces the flow rate, a quantitative evaluation of the scale is essential for the proper maintenance of pipes. Guided waves were employed to estimate the amount of scale in water supplying pipes. Using variable angle wedge, several modes of guided waves were generated and their propagation charcteristics along the pipes with scale were analyzed. It was experimentally observed that the amplitude of F(M,2) modes at f×d=1.5㎒·㎜ decreased significantly with increasing amount of scale. The present study showed that F(M,2) modes were optimal to evaluate the scale in water supplying pipes.

      • Microstructure and Mechanical Properties of Twin-Roll Strip Cast Mg Alloys

        <P>Development of wrought Mg alloys, particularly in sheet form, is essential to support the growing interest for lightweight components in the automotive industry. However, development of Mg alloy sheets has been quite slow due to the complexity of sheet production originated from limited deformability of Mg. In this respect, twin-roll strip casting, a one-step processing of flat rolled products, can be an alternative for the production of Mg alloy sheets. In this study, AZ31 and experimental ZM series alloys are twin-roll strip cast into 2 mm thick sheets. The microstructure of the as-cast AZ31 alloy sheet consists of columnar zones near the roll side and equiaxed zones in the mid-thickness region. On the other hand, as-cast ZM series alloy sheets show equiaxed dendritic structure through the thickness of sheet. These alloys were subjected to various thermo-mechanical treatments and their tensile properties were evaluated. Twin-roll strip cast AZ31 alloy in H24 condition has equivalent yield and tensile strengths with similar ductility compared to commercial ingot cast AZ31-H24 alloy, indicating that twin-roll strip casting is a viable process for the fabrication of Mg alloy sheets.       • 후방복사 레일리 표면탄성파를 이용한 부식열화평가

        후방복사 레일리 표면탄성파를 사용하여 부식에 의한 재료 손상을 평가하였다. 입사각, 입사점을 정밀하게 제어할 수 있는 자동화된 system을 이용하여 부식열화된 12Cr 합금강과 부식·피로 열화된 TMCP강에 대하여 후방복사 초음파 신호를 측정하였다. 손상의 정도에 후방복사 프로파일의 세기와 레일리 표면탄성파의 속도가 달라짐을 확인하였다. 이러한 결과는 레일리 표면탄성파가 노후 재료의 부식손상을 비파괴적으로 평가하는데 유용한 도구임을 보여주었다.

        Effective charge separation in site-isolated Pt-nanodot deposited PbTiO<sub>3</sub> nanotube arrays for enhanced photoelectrochemical water splitting

        <P><B>Abstract</B></P> <P>Highly uniform, self-supported PbTiO<SUB>3</SUB> nanotube arrays are fabricated on a transparent conducting glass by an all solution-based, hard-templating procedure. A new concept of site-isolation has been realized by Pt-sol infiltration only in the internal core of deposited nanotube arrays and thus physically separating electron and hole reaction sites on inside and external surface of the nanotubes, respectively. The effective charge separation by the site-isolated Pt-nanodot deposited PbTiO<SUB>3</SUB> nanotube photoanode leads to greatly enhanced photocurrent generation and H<SUB>2</SUB> evolution efficiencies relative to those of the particulate-type photoanode or PbTiO<SUB>3</SUB> nanotube without Pt infiltration in photoelectrochemical water splitting under visible light.         Anti-Helicobacter pylori activities of FEMY-R7 composed of fucoidan and evening primrose extract in mice and humans

        Helicobacter pylori-eliminating effects of FEMY-R7, composed of fucoidan and evening primrose extract, were investigated in mice and humans. Male C57BL/6 mice were infected with the bacteria by intragastric inoculation (1×10<SUP>9</SUP> CFU/mouse) 3 times at 2-day intervals, and simultaneously, orally treated twice a day with 10 or 100 mg/kg FEMY-R7 for 2 weeks. In Campylobcter-like organism-detection test, FEMY-R7 markedly reduced the urease-positive reactivity. In a clinical sudy, human subjects, confirmed to be infected with Helicobacter pylori, were orally administered twice a day with a capsule containing 150 mg FEMY-R7 for 8 weeks. FEMY-R7 significantly decreased both the Delta over baseline-value in urea breath test and the serum pepsinogens I and II levels. The results indicate that FEMY-R7 not only eliminates H. pylori from gastric mucosa of animals and humans, but also improves gastric function.

        Metabolic syndrome and quality of life (QOL) using generalised and obesity-specific QOL scales

        <P>OBJECTIVES: We investigated the association between metabolic syndrome (MS) and health-related quality of life (HRQOL) assessed using generalised and obesity-specific QOL instruments. METHODS: We recruited 456 outpatients [age: 19-81 years, body mass index (BMI): 16.3-36.7 kg/m2] in the primary care division from 12 general hospitals in Korea. HRQOL was measured using EuroQol comprising the health states descriptive system (EQ-5D) and visual analogue scale (EQ-VAS) as a general instrument. The Korean Obesity-related QOL scale (KOQOL) composed of six domains was used as a disease-specific QOL instrument. MS was defined on the basis of International Diabetes Federation (IDF) criteria with Korean-specific waist circumference cutoffs (men: 90 cm, women: 85 cm). RESULTS: Subjects with MS displayed significantly higher impairment of EQ-5D and KOQOL. Binary logistic regression analysis of MS patients with controls for age, gender, smoking, alcohol, exercise, education, income, marital status and medication history disclosed odds ratio (OR) values of 2.13 (1.33-3.41) for impaired total KOQOL, 2.07 (1.31-3.27) for impaired physical health, 1.63 (1.03-2.60) for impaired work-related health, 2.42 (1.45-4.04) for impaired routine life, 2.08 (1.27-3.40) for impaired sexual life and 2.56 (1.59-4.11) for diet distress. Among the EQ-5D dimensions, only pain/discomfort displayed a significantly increased OR of 1.60 (1.01-2.56) in MS group. CONCLUSIONS: Subjects with MS displayed a significantly impaired HRQOL compared with those without MS. MS and HRQOL were more strongly associated in obesity-specific QOL than in generalised QOL.</P>

        Oxidative stress is important for the pathogenesis of nonalcoholic fatty liver disease (NAFLD), a chronic disease that ranges from hepatic steatosis to nonalcoholic steatohepatitis (NASH). The nuclear factor erythroid 2-related factor 2-Kelch-like ECH associated protein 1 (Nrf2-Keap1) pathway is essential for cytoprotection against oxidative stress. In this study, we found that oxidative stress or inflammatory biomarkers and TUNEL positive cells were markedly increased in NASH patients compared to normal or simple steatosis. In addition, we identified that the hepatic mRNA levels of Nrf2 target genes such as Nqo-1 and GSTA-1 were significantly increased in NASH patients. Ezetimibe, a drug approved by the Food and Drug Administration for the treatment of hypercholesterolemia, improves NAFLD and alleviates oxidative stress. However, the precise mechanism of its antioxidant function remains largely unknown. We now demonstrate that ezetimibe activates Nrf2-Keap1 pathway which was dependent of autophagy adaptor protein p62, without causing cytotoxicity. Ezetimibe activates AMP-activated protein kinase (AMPK), which in turn phosphorylates p62 (p-S351) via their direct interaction. Correspondingly, Ezetimibe protected liver cells from saturated fatty acid-induced apoptotic cell death through p62-dependent Nrf2 activation. Furthermore, its role as an Nrf2 activator was supported by methione- and choline- deficient (MCD) diet-induced NASH mouse model, showing that ezetimibe decreased the susceptibility of the liver to oxidative injury. These data demonstrate that the molecular mechanisms underlying ezetimibe's antioxidant role in the pathogenesis of NASH.
