        HPLC-DAD Phenolic Profiling and In Vitro Antioxidant Activities of Three Prominent Nigerian Spices

        Spices and herbs have antioxidant, anti-inflammatory, and anti-microbial properties, amongst others. These characteristics are attributable to their composition, such as high polyphenol and flavonoid contents that are responsible for their antioxidative properties. Methanolic extracts of Ocimum basilicum (OB), Xylopia aethiopica (XA), and Piper guineensis (PG) were evaluated to profile their phenolic compounds and in vitro antioxidant properties. High performance liquid chromatography with diode-array detection phenolic compounds profiling revealed that quercetin, quercitrin, and isoquercitrin are the most prevalent phenolic compound in OB, XA, and PG, respectively. All the extracts possessed good antioxidant activity. XA showed the highest total phenolic content of 29.50 ㎎ gallic acid equivalents/g, a total flavonoid content of 21.17 ㎎ quercetin equivalents/g, 2,2-diphenyl-1-picrylhydrazyl and 2,2’-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid) radical scavenging abilities of 29% and 88.23%, respectively, and a nitric oxide scavenging activity of 44.13 ㎎/g. Thus, the XA methanolic extract demonstrated a high content of phenolic compounds and significant antioxidative properties, with prospective potency to prevent oxidative damage and promote better cardiovascular health.

        Influence of Sourdough Fermentation on Amino Acids Composition, Phenolic Profile, and Antioxidant Properties of Sorghum Biscuits

        Biscuits were produced from sorghum with and without the addition of sourdough. The influence of sourdough fermentation on the amino acids composition, phenolic profile, and antioxidant properties of the biscuits were evaluated. Phenolic compounds of the biscuits were identified and quantified using gas chromatography/mass spectrometer. The total phenol contents (TPC), total flavonoid contents (TFC), ferric reducing antioxidant properties (FRAP), 2,2’-azinobis(3-ethylbenzothiazoline-6-sulphonic) (ABTS) scavenging ability, 1,1-diphenyl-2-picryl-hydrazy (DPPH) scavenging ability, and nitric oxide (NO) scavenging ability of the biscuits were determined. Addition of sourdough increased the total essential amino acids, total non essential amino acids, hydrophobic, and aromatic amino acids contents of the biscuits. Essential amino acid index, biological value, and predicted protein efficiency were higher in biscuits with sourdough than in biscuits without sourdough. Six phenolic compounds were identified and quantified in the biscuits. Ferulic acid was the most prominent phenolic compound, followed by chlorogenic acid. TPC, TFC, FRAP, ABTS, DPPH, and NO scavenging abilities increased significantly with the addition of sourdough. Sorghum biscuits with sourdough could be useful in dietary interventions to prevent protein-energy malnutrition. Similarly, the presence of bioactive phenolic compounds and their antioxidant efficacy suggest health benefits in the management of oxidative stress and degenerative diseases.

        Chemical Compositions and Antioxidant Properties of Orange Fleshed Sweet Potato Leaves and the Consumer Acceptability in Vegetable Soup

        The purpose of this study is to determine the chemical compositions, anti-nutrient compositions, antioxidant properties, and phenolic profile of the leaves of orange fleshed sweet potato varieties [King J (UMUSPO1) and mother’s delight (UMUSPO2)] in Nigeria and their suitabilities in soup preparation. Freshly harvested leaves of the two varieties of orange fleshed sweet potato were sorted, cleaned, dried, and milled into powder. The dried leaves were assessed for their chemical compositions viz proximate and mineral compositions, the anti-nutrient compositions, antioxidant properties, and phenolic profile. The overall acceptability of the leafy vegetables in the preparation of a local soup (Edikang Ikong) was evaluated. The ash, fat, and protein contents of UMUSPO1 leaves were higher than UMUSPO2, similarly UMUSPO1 leaves had higher mineral contents. Phytate and saponin contents were higher in UMUSPO2, however, the calculated molar ratios were below critical levels. The 2,2-diphenyl-1-picrylhydrazyl hydrate radical scavenging activity and ferric reducing antioxidant power assay were higher in UMUSPO1. Seven phenolic compounds were identified and quantified in both leaves with gallic acid being the most abundant. The taste of soup prepared with UMUSPO2 was rated higher, however, no significant difference was observed in the overall acceptability of the soups. The two leaves are good sources of nutritional antioxidants and can be suitable for the management of some disease conditions linked to oxidative stress.

        Endoscope-Assisted Microsurgical Removal of an Epidermoid Tumor within the Cavernous Sinus

        Epidermoid tumor of the cavernous sinus is rare. The aim of this case report is to discuss the role of neuroendoscopes in the removal of such lesions. A 21-year-old man presented with 6-year history of progressive headache, diplopia, and visual disturbance. Work-up revealed an epidermoid tumor located in the right cavernous sinus. An extradural transcavernous approach was utilized via a traditional frontotemporal craniotomy with endoscopic assistance. The postoperative course was uneventful with immediate improvement of the patient’s headache. Postoperative magnetic resonance imaging demonstrated complete removal of the tumor. There were no signs of recurrence during a 2-year follow-up period. The endoscope is a useful tool for removing epidermoid tumors from the cavernous sinus and enhances visualization of areas that would otherwise be difficult to visualize with microscopes alone. Endoscopes also help minimize the retraction of neurovascular structures.

        Multiresponse Optimization and Prediction of Antioxidant Properties of Aqueous Ginger Extract

        The influence of extraction temperature, powder concentration, and extraction time on the antioxidant properties of aqueous ginger extract was investigated. The possibility of estimating the antioxidant properties of the extract from its absorbance and colour properties was also investigated. Results indicated that powder concentration was the most significant factor to consider in optimizing antioxidant extraction. However, temperature and time still influenced the 2,2’-azinobis-(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) radical scavenging activity while extraction temperature influenced the 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity of the extract. Using the total phenol content, total flavonoid content, ABTS radical scavenging activity, and DPPH radical scavenging activity of the extract, the multiresponse optimization condition for extraction of antioxidant based on the experimental range studied is 96℃, 2.10 g/100 mL, and 90 min. The absorbance of the ginger extract at 610 nm could be exploited for rapid estimation of its total flavonoid and polyphenol with a R<SUP>2</SUP> of 0.713 and 0.753, respectively.

        Evaluation of Non-Watertight Dural Reconstruction with Collagen Matrix Onlay Graft in Posterior Fossa Surgery

        Objective : Many surgeons advocate for watertight dural reconstruction after posterior fossa surgery given the significant risk of cerebrospinal fluid (CSF) leak. Little evidence exists for posterior fossa dural reconstruction utilizing monolayer collagen matrix onlay graft in a non-watertight fashion. Our objective was to report the results of using collagen matrix in a non-watertight fashion for posterior fossa dural reconstruction. Methods : We conducted a retrospective review of operations performed by the senior author from 2004-2011 identified collagen matrix (DuraGen) use in 84 posterior fossa operations. Wound complications such as CSF leak, infection, pseudomeningocele, and aseptic meningitis were noted. Fisher's exact test was performed to assess risk factor association with specific complications. Results : Incisional CSF leak rate was 8.3% and non-incisional CSF leak rate was 3.6%. Incidence of aseptic meningitis was 7.1% and all cases resolved with steroids alone. Incidence of palpable and symptomatic pseudomeningocele in follow-up was 10.7% and 3.6% respectively. Postoperative infection rate was 4.8%. Previous surgery was associated with pseudomeningocele development (p<0.05). Conclusion : When primary dural closure after posterior fossa surgery is undesirable or not feasible, non-watertight dural reconstruction with collagen matrix resulted in incisional CSF leak in 8.3%. Incidence of pseudomeningocele, aseptic meningitis, and wound infection were within acceptable range. Data from this study may be used to compare alternative methods of dural reconstruction in posterior fossa surgery.

        COX I 및 RAG 2 유전자 염기서열 분석에 의한 tiger grouper Epinephelus fuscoguttatus와 giant grouper E. Lanceolatus 간 잡종의 동정

        Interspecific hybrids between tiger grouper Epinephelus fuscoguttatus and giant grouper E. lanceolatus were genetically identified based on the partial sequence analysis of mitochondrial cytochrome coxidase I (COX I) gene and nuclear recombination activating gene 2 (RAG 2) gene. Out of 585 base positions of RAG 2, a total of five nucleotide substitutions were detected between the two parental species (E. fuscoguttatus and E. lanceolatus). The hybrids had two distinct types of RAG 2 sequences corresponding to those of both parental species. Mitochondrial COX I gene sequencing showed that hybrids had sequences identical to E. fuscoguttatus. Molecular data clearly demonstrate that hybridization does occur between E. fuscoguttatus and E. lanceolatus, but with E. fuscoguttatus as the maternal parent.

        Microsatellite 마커를 이용한 대왕바리(Epinephelus lanceolatus) 친어 집단의 가계도 분석 효율

        현재 IUCN의 취약 등급인 대왕바리(giant grouper, Epine-phelus lanceolatus) 친어의 효율적인 관리 시스템 구축을 위한 기반연구로서 기 개발되어 있는 동종의 microsatellite 마커를 이용한 가계도 분석 효율을 조사하였다. 대왕바리 친어32마리를 8개의 microsatellite 마커로 분석한 결과 총 52개의 대립유전자가 검출되었으며, 기대치 이형접합율은 0.663, 근친교배계수는 0.011로 조사되어 현재 확보된 대왕바리 친어는 유전 다양성이 비교적 잘 유지되고 있었다. 하지만 유효집단 크기가 35로 추정됨으로써 지속적인 친어 확보의 필요성을 보였다. 해당 마커를 이용한 동일 유전자형 출현확률은 무작위 집단에서 6.85×10-11 그리고 한쪽 부모의 유전자형 확보 및 양친의 유전자형이 확보된 상태에서의 부권 부정률은 각각 0.00835, 0.00027로 나타났으며, 주좌표분석 결과 친어의 유전자형은 중복되지 않았다. 따라서 본 연구에 이용한 8개의 microsatellite 마커로도 유전자형 데이터베이스를 기반으로 한 대왕바리 친어 관리 시스템 구축이 가능할 것이며, 이를 활용한 유전 다양성이 높은 자손 생산 및 유전적으로 유사한 개체의 중복 확보를 방지할 수 있어 친어 확보의 효율성을 높일 수 있을 것이다. Giant grouper (Epinephelus lanceolatus) is a endangered species considered as a vul-nerablegrade-organism in the International Union for Conservation of Nature (IUCN) red list. As a fun-damental baseline study for establishing a giant grouper broodstock management system, the effici-ency for parentage analysis was evaluated by using microsatellite makers previously available in this species. The eight microsatellites generated a total 52 alleles from 32 individuals, the mean expected heterozygosity was 0.663, and mean inbreeding coefficient was 0.011, consequently suggesting that the present broodstock has retained the high level of genetic diversity. However, our analysis also recommended the collection of more brood fish for more stable brood line, since the estimated value of the effective population size was proven to be 35. The average probability of identity was 6.85×10-11. NE-2P and NE-PP of paternity non-exclusion probabilities were 0.00835 and 0.00027, respectively. As the result of principle coordinate analysis, the genotype of broodstock was not overlapped, suggesting that the management system of giant grouper based on eight selected microsatellite markers might be effective, although further validation with extended number of brood fish might also be needed in future. Data of present study could be a useful basis to avoid the unwanted selection of broodfish that possess close genetic relationship with current broodstock, and consequently to establish effective broodstock management system allowing the production of progeny with high genetic diversity.

        Morphological and molecular evidence for the recognition of Hypoglossum sabahense sp. nov. (Delesseriaceae, Rhodophyta) from Sabah, Malaysia

        Culture isolates of the genus Hypoglossum (Delesseriaceae, Rhodophyta) were obtained and their development and morphological structure over many years were followed in the laboratory. Molecular data (rbcL, large subunit ribosomal DNA, and cytochrome c oxidase subunit I) were obtained from these strains and evidence presented to recognize the new species: Hypoglossum sabahense from Sabah, Malaysia. Because various aspects of morphology in culture specimens differ significantly from types based on field specimens we have to rely mainly on the molecular criteria in ascribing a new taxonomic name here. This also is complicated by the major lack of molecular phylogenetic evidence for Hypoglossum and other Delesseriaceae. The 'Germling Emergence Method' and 'serendipity' are proving valuable in discovering significant new taxa from laboratory cultures which otherwise might never be known.
