      • Survival and colonization of <i>Escherichia coli</i> O157:H7 on spinach leaves as affected by inoculum level and carrier, temperature and relative humidity

        <P><B>Abstract</B></P><P><B>Aims: </B> To determine survival and colonization of <I>Escherichia coli</I> O157:H7 on spinach leaves as affected by inoculum level and carrier, temperature and relative humidity (r.h.).</P><P><B>Methods and Results: </B> Spinach leaves were inoculated with suspensions of <I>E.?coli</I> O157:H7 in distilled water (DW) and 0·1% peptone water (PW) and incubated at 4, 12 and 25°C and 43, 85 and 100% r.h. The number of <I>E.?coli</I> O157:H7 on leaves (5·6 or 1·9 log CFU per leaf) inoculated using DW as a carrier medium increased significantly at 25°C and 100% r.h. within 120 h but remained constant or decreased significantly under other test conditions. <I>E.?coli</I> O157:H7 on leaves (5·4 log CFU per leaf) inoculated using PW as a carrier increased significantly within 72 and 24 h, respectively, at 12 or 25°C and 100% r.h.; counts using a low inoculum (2·2 log CFU per leaf) increased significantly within 24 h at 25°C.</P><P><B>Conclusions: </B> <I>Escherichia coli</I> O157:H7 can colonize on spinach leaves at 12 or 25°C in a 100% r.h. environment. Organic matter in the inoculum carrier may provide protection and nutrients which enhance survival and colonization.</P><P><B>Significance and Impact of the Study: </B> Colonization of <I>E.?coli</I> O157:H7 on spinach leaves as affected by organic matter in the inoculum, temperature and r.h. was determined. These observations will be useful when developing strategies to prevent growth of <I>E.?coli</I> O157:H7 on pre‐ and postharvest spinach.</P>

        Peroxiredoxin II promotes hepatic tumorigenesis through cooperation with Ras/Forkhead box M1 signaling pathway

        <P>The current study was carried out to define the involvement of Peroxiredoxin (Prx) II in progression of hepatocellular carcinoma (HCC) and the underlying molecular mechanism(s). Expression and function of Prx II in HCC was determined using H-ras(G12V)-transformed HCC cells (H-ras(G12V)-HCC cells) and the tumor livers from H-ras(G12V)-transgenic (Tg) mice and HCC patients. Prx II was upregulated in H-ras(G12V)-HCC cells and H-ras(G12V)-Tg mouse tumor livers, the expression pattern of which highly similar to that of forkhead Box M1 (FoxM1). Moreover, either knockdown of FoxM1 or site-directed mutagenesis of FoxM1-binding site of Prx II promoter significantly reduced Prx II levels in H-ras(G12V)-HCC cells, indicating FoxM1 as a direct transcription factor of Prx II in HCC. Interestingly, the null mutation of Prx II markedly decreased the number and size of tumors in H-ras(G12V)-Tg livers. Consistent with this, knockdown of Prx II in H-ras(G12V)-HCC cells reduced the expression of cyclin D1, cell proliferation, anchorage-independent growth and tumor formation in athymic nude mice, whereas overexpression of Prx II increased or aggravated the tumor phenotypes. Importantly, the expression of Prx II was correlated with that of FoxM1 in HCC patients. The activation of extracellular signal-related kinase (ERK) pathway and the expression of FoxM1 and cyclin D1 were highly dependent on Prx II in H-ras(G12V)-HCC cells and H-ras(G12V)-Tg livers. Prx II is FoxM1-dependently- expressed antioxidant in HCC and function as an enhancer of Ras(G12V) oncogenic potential in hepatic tumorigenesis through activation of ERK/FoxM1/cyclin D1 cascade.</P>

        Temporal and Spatial PFC Temperature Profiles in KSTAR 2010

        <P>In and outboard plasma facing components (PFCs) of Korea Superconducting Tokamak Advanced Research (KSTAR) have been fully installed in 2010 for D-shaped diverted plasmas. Before the start of plasma operation, the PFCs were baked up to 200 by hot nitrogen gas circulation system to remove impurities including water. The surface temperature of the PFC tiles was monitored by 200 thermocouple sensors during the plasma operation [plasma shot or glow discharge cleaning (GDC)], and the temporal and spatial (poloidal) temperature profiles are obtained. Depending on the heat flux on each tile, the surface temperature shows time-dependent behavior. After 1-h morning He GDC, the temperature of the PFCs at the inboard side has reached to 40 . After an H-mode shot, the temperature of divertor tiles around the striking points was substantially increased. The time-averaged total heat flux after a few of L- and H-modes in 2010 was estimated to be approximately 0.010 .</P>

      • Antiviral activities of compounds from aerial parts of Salvia plebeia R. Br

        Ethnopharmacological relevance: Salvia plebeia R. Br. is an edible plant widely spread in many countries. It has been used as a traditional medicine to treat common cold, flu, cough, hepatitis, hemorrhoids, etc. The purpose of the study is to explicate antiviral compounds responsible for its traditional use for the common cold or flu. Materials and methods: The methanolic extract of the aerial parts of S. plebeia was extracted with CHCl<SUB>3</SUB>, EtOAc, and n-BuOH, successively. The EtOAc and CHCl<SUB>3</SUB> fractions were subjected to a successive of chromatographic method, which led to the isolation of fourteen compounds. Inhibition activities of the isolated compounds were evaluated against influenza A (H1N1) neuraminidase. Results: Chemical investigation of the methanolic extracts of S. plebeia resulted in the isolation of two novel benzoylated monoterpene glycosides, named as plebeiosides A (1) and B (2), together with twelve known compounds including four flavonoids (4-5, 7, 10), two sesquiterpenoids (8, 12), four phenolics (9-10, 13-14), a steroid (6), and a triterpenoid (3). Their chemical structures were elucidated based on spectroscopic data and absolute stereochemistries of 1 and 2 were determined by comparison of optical rotations of their hydrolysates with literature values. Compounds 5, 7, 9, and 11 exhibited potent enzymatic inhibition against H1N1 neuraminidase (IC<SUB>50</SUB> values ranging from 11.18+/-1.73 to 19.83+/-2.28μM). Furthermore, two flavonoids (5 and 7) and one rosmarinic acid methyl ester (9) reduced cytopathic effects of the H1N1 virus during replication. Conclusions: The antiviral activities of the flavonoids and phenolics isolated from the extracts of S. plebeia supported the traditional application of this medicine on common cold or flu. In this study, benzoylated monoterpene glycosides were first found to exist in this species. Moreover, the present study suggested potential of three compounds (5, 7, and 9) to be new lead structures for the development of new neuraminidase inhibitors in the future.

      • Modeling and simulation of HTS cables for scattering parameter analysis

        <P>Most of modeling and simulation of high temperature superconducting (HTS) cables are inadequate for high frequency analysis since focus of the simulation's frequency is fundamental frequency of the power grid, which does not reflect transient characteristic. However, high frequency analysis is essential process to research the HTS cables transient for protection and diagnosis of the HTS cables. Thus, this paper proposes a new approach for modeling and simulation of HTS cables to derive the scattering parameter (S-parameter), an effective high frequency analysis, for transient wave propagation characteristics in high frequency range. The parameters sweeping method is used to validate the simulation results to the measured data given by a network analyzer (NA). This paper also presents the effects of the cable-to-NA connector in order to minimize the error between the simulated and the measured data under ambient and superconductive conditions. Based on the proposed modeling and simulation technique, S-parameters of long-distance HTS cables can be accurately derived in wide range of frequency. The results of proposed modeling and simulation can yield the characteristics of the HTS cables and will contribute to analyze the HTS cables. (C) 2016 Elsevier B.V. All rights reserved.</P>

        척수손상 환자를 위한 병원기반 사회복귀지원 프로그램의 개발 및 효과

        본 연구는 척수손상 환자를 대상으로 병원기반 사회복귀 지원프로그램을 개발하고, 프로그램 시행 전·후를 비교 분석함으로써 프로그램의 효과성을 평가하고자 하였다. 재활병원에 입원해 있는 25명의 척수손상 환자를 대상으로 2~3개월 동안 일주일에 2번 평균 6.1개의 프로그램을 실시하였다. 프로그램 시행 전, 후 COPM, HADS, AD-R, WHOQOL-BREF를 통해 작업 수행도 및 만족도, 불안과 우울, 장애수용, 삶의 질을 비교하였으며, 퇴원 시 CIQ로 지역사회통합 수준과 퇴원 후의 사회복귀여부를 측정하였다. 자료분석은 기술통계와 대응표본 t검정(paired t-test)을 실시하였다. 연구결과 병원기반 사회복귀 지원프로그램 실시 후 작업 수행도 및 만족도, 장애수용, 삶의 질에 향상을 보였다(p<0.05). 또한 퇴원 후 25명의 척수손상 환자 중에서 10명이 가정, 학업, 직업으로 복귀하였다. 이를 통해 병원기반 사회복귀 지원프로그램이 척수손상 환자들이 퇴원 후 그들이 속한 지역사회로 돌아가는데 효과적임을 알 수 있었다. 척수손상 환자들이 가정과 사회로의 빠른 복귀를 위해서는 사회복귀를 위한 다양한 프로그램 개발과 더불어 체계적인 사회복귀 연계시스템이 이루어져야 할 것으로 사료된다. The purpose of this research attempted to developed the Community Reintegration support Program(H-CRsP) on patients with Spinal Cord Injury(SCI) and identity the effect. To assess the effectiveness of a Hospital based H-CRsP with a comprehensive team approach for treating inpatients with Spinal Cord Injury(SCI), offered by the Korea National Rehabilitation Hospital. Participants with SCI were recruited from the Korean National Rehabilitation Hospital. Twenty-five participants who met inclusion criteria were provided with a H-CRsP. A H-CRsP with modules related to the training of daily living skills, driving, vocations and school support activities, leisure or recreational activities, peer counseling, sexual rehabilitation, and others support activities. The 25 enrolled patients had been treated with average of 6.1 programs 2 times a week for over 2 or 3months. COPM, HADS, WHOQOL-BREF, AD-R were administered before the treatment(pre-test) and also at the time of discharge(post-test) to compare the treatment outcomes in the 25 program participants. Measured level of community integration by CIQ at discharge. The data were analyzed by such statistical methods as frequency and paired t-test analysis. Significant improvements were found on the Occupational Performance and Satisfaction, Acceptance of Disability, Quality of Life score in the COPM, AD-R, WHOQOL-BREF (p<0.05). In addition, 10 of the 25 patients returned to home, work or school. These results demonstrated the effectiveness of the H-CRsP in helping patients with SCI return their communities to which they belong. It considered development of the variety of programs and prepare systematically related system.

        Quantification of main wall erosion/deposition pattern in n = 2 RMP KSTAR H-mode plasmas

        <P><B>Abstract</B></P> <P>Erosion/deposition pattern was studied using cavity sample at KSTAR main wall region. The cavity sample has a different two slit opening directions. The amorphous carbon film inside the cavity structure was installed on mid-plane manipulator system and was exposed to H-mode plasmas with n = 2 RMP. The most of characterized amorphous hydrocarbon film in cavity structure was eroded after exposing to H-mode plasmas. The erosion rate of bottom plate sample was in a range from 0.32 nm s<SUP>−1</SUP> to 0.77 nm s<SUP>−1</SUP> in a different slit opening directions, respectively. But the upper samples were deposited and the maximum deposition rate of it was 0.42 nm s<SUP>−1</SUP>. Thickness profiles measured by ellipsometer were compared with simulated profile using Monte Carlo Ballistic Simulation. The main parameter for simulation was the incident angle of particle. The incident angle of particle influenced the shape and width of simulated profile. The incident angle used in simulation was in the range of 80°–89° and it was well matched with 81.9° derived from EFIT.</P>

        Castellated tungsten plasma-facing components exposed to H-mode plasma in KSTAR

        Heat load on the misaligned leading edges of tungsten castellated blocks based on tungsten (W), oxygen-free high conductive copper (OFHC-Cu), and copper-chrome-zirconium (CuCrZr) alloy are studied by COMSOL analysis and infrared (IR) measurements in KSTAR. IR measurements show that 1-3MW/m<SUP>2</SUP> of heat flux has been deposited on the blocks during the inter-ELM (edge localized mode) phase in H-mode plasmas. COMSOL analysis indicates that the temperature of 1mm leading edge in KSTAR under 3MW/m<SUP>2</SUP> would reach the recrystallization temperature within 2s and will be melted within 30s during a long pulse H-mode shot. Rounded and double chamfered blocks show much better thermal response meaning that shaping of divertor block enhances the heat load handling capability. It seems that a simple COMSOL analysis describes heat load patterns on the tungsten blocks of different shapes qualitatively well. Therefore, simple analysis would be useful to make a quick prediction on heat load patterns of blocks with arbitrary shapes.

      • Influence of <i>CYP2D6*10</i> on the pharmacokinetics of metoprolol in healthy Korean volunteers

        <P>Summary</P><P>Background and objective: </P><P>Genetic polymorphism of <I>CYP2D6</I> leads to differences in pharmacokinetics of CYP2D6 substrates. The <I>CYP2D6*10</I> allele is clinically important in Koreans because of its high frequency in Asians. We investigated whether the pharmacokinetics of metoprolol was altered by the presence of the <I>CYP2D6*10</I> allele in Korean subjects.</P><P>Methods: </P><P>One hundred and seven volunteers were recruited and grouped as <I>CYP2D6*1/*1</I>, <I>CYP2D6*1/*10</I> and <I>CYP2D6*10/*10</I> according to their genotypes. Metoprolol tartrate 100 mg (Betaloc<SUP>®</SUP>) was administered orally once to each subject in these three groups (<I>n</I> = 6, 7 and 5, respectively). The pharmacokinetic parameters of metoprolol and its metabolite, &agr;-hydroxymetoprolol, and the metabolic ratio for the three groups were estimated and compared.</P><P>Results and discussion: </P><P>The area under the plasma concentration–time curve (AUC<SUB>0→∞</SUB>), the maximum plasma concentration (<I>C</I><SUB>max</SUB>) and the elimination half-life (<I>T</I><SUB>1/2</SUB>) of metoprolol and &agr;-hydroxymetoprolol for the <I>CYP2D6*10/*10</I> group were all significantly different from those of the <I>CYP2D6*1/*1</I> group (<I>P</I> < 0·05). The AUC<SUB>0→∞</SUB>s of metoprolol were 443·7 ± 168·1, 995·6 ± 321·4 and 2545·3 ± 632·0 ng·h/mL, and the AUC<SUB>0→∞</SUB>s of &agr;-hydroxymetoprolol were 1232·0 ± 311·2, 1344·0 ± 288·1 and 877·4 ± 103·4 ng·h/mL for groups <I>CYP2D6*1/*1</I>, <I>*1/*10</I> and <I>*10/*10</I>, respectively. The corresponding <I>T</I><SUB>1/2</SUB> values of metoprolol were 2·7 ± 0·5, 3·2 ± 1·3 and 5·0 ± 1·1 h, while those of &agr;-hydroxymetoprolol were 5·4±1·5, 6·0 ± 1·4 and 10·5 ± 4·2 h, respectively. The metabolic ratios of the three groups were significantly different (<I>P</I> < 0·05).</P><P>Conclusion: </P><P>The <I>CYP2D6*10</I> allele altered the pharmacokinetics of metoprolol in Korean subjects and is likely to affect other drugs metabolized by the CYP2D6 enzyme, similarly.</P>
