        Effects of climate change on the physiology of giant kelp, Macrocystis pyrifera, and grazing by purple urchin, Strongylocentrotus purpuratus

        As global warming continues over the coming century, marine organisms will experience a warmer, more acidic ocean. Although these stressors may behave antagonistically or synergistically and will impact organisms both directly (i.e., physiologically) and indirectly (i.e., through altered species interactions), few studies have examined the complexities of these effects in combination. To address these uncertainties, we examined the independent and combined effects of elevated temperature and $pCO_2$ on the physiology of the adult sporophyte stage of giant kelp, Macrocystis pyrifera, and the grazing of the purple sea urchin Strongylocentrotus purpuratus. While elevating $pCO_2$ alone had no effect on M. pyrifera growth or photosynthetic carbon uptake, elevating temperature alone resulted in a significant reduction in both. However, when M. pyrifera was grown under elevated temperature and $pCO_2$ together, growth and photosynthetic carbon uptake significantly increased relative to ambient conditions, suggesting an interaction of these factors on photosynthetic physiology. S. purpuratus held under future conditions generally exhibited reduced growth, and smaller gonads than urchins held under present-day conditions. However, urchins fed kelp grown under future conditions showed higher growth rates, partially ameliorating this effect. Feeding rates were variable over the course of the experiment, with only the first feeding rate experiment showing significantly lower rates for urchins held under future conditions. Together, these data suggest that M. pyrifera may benefit physiologically from a warmer, more acidic (i.e., higher $pCO_2$) ocean while S. purpuratus will likely be impacted negatively. Given that kelp-urchin interactions can be important to kelp forest structure, changes to either of these populations may have serious consequences for many coastal environments.

        Disinfection performance of adsorption using graphite adsorbent coupled with electrochemical regeneration for various microorganisms present in water

        The disinfection performance of the process of adsorption using a graphitic material combined withelectrochemical regeneration for a range of microorganisms including bacteria, fungi, yeast and protozoain a laboratory scale sequential batch reactor is demonstrated. The bacterial species studied werePseudomonas aeruginosa,Staphylococcus aureus and Legionella pneumophila. A 3.0 log10 reduction in theconcentration of P. aeruginosa cells was achieved with the adsorbent that was regenerated at 30 mA cm 2with 100% regeneration on each adsorption cycle. The process was quite effective in removing S. aureuspresent in water with a significantly higher reduction in the number of cells (ca. 9-log10 reduction) atrelatively low current density (10 mA cm 2). Similarly, L. pneumophila were removed from water with aca. 7.5-log10 reduction in the number of bacterial cells. The SEM images confirmed the adsorption of L. pneumophila onto the adsorbent and its electrochemical regeneration at 20 mA cm 2 that is considered arefractory pathogen against chlorination. The process was also found to be suitable for disinfecting fungalspores, Aspergillus awamori and yeasts including Saccharomyces cerevisiae and Rhodosporidium turoloidesHowever, the removal of Cryptosporidium parvum from water was not demonstrated successfully. Thepreliminary results suggest that using a chloride free environment and a relatively high current densitycould be useful in disinfecting C. parvum.

      • OH reactivity at a rural site (Wangdu) in the North China Plain: contributions from OH reactants and experimental OH budget

        <P>Abstract. In 2014, a large, comprehensive field campaign was conducted in the densely populated North China Plain. The measurement site was located in a botanic garden close to the small town Wangdu, without major industry but influenced by regional transportation of air pollution. The loss rate coefficient of atmospheric hydroxyl radicals (OH) was quantified by direct measurements of the OH reactivity. Values ranged between 10 and 20 s−1 for most of the daytime. Highest values were reached in the late night with maximum values of around 40 s−1. OH reactants mainly originated from anthropogenic activities as indicated (1) by a good correlation between measured OH reactivity and carbon monoxide (linear correlation coefficient R2 = 0.33) and (2) by a high contribution of nitrogen oxide species to the OH reactivity (up to 30 % in the morning). Total OH reactivity was measured by a laser flash photolysis-laser-induced fluorescence instrument (LP-LIF). Measured values can be explained well by measured trace gas concentrations including organic compounds, oxygenated organic compounds, CO and nitrogen oxides. Significant, unexplained OH reactivity was only observed during nights, when biomass burning of agricultural waste occurred on surrounding fields. OH reactivity measurements also allow investigating the chemical OH budget. During this campaign, the OH destruction rate calculated from measured OH reactivity and measured OH concentration was balanced by the sum of OH production from ozone and nitrous acid photolysis and OH regeneration from hydroperoxy radicals within the uncertainty of measurements. However, a tendency for higher OH destruction compared to OH production at lower concentrations of nitric oxide is also observed, consistent with previous findings in field campaigns in China. </P>

        Comparative adsorption–regeneration performance for newly developed carbonaceous adsorbent

        A proprietary adsorbent material called Nyex 1000 was developed by the Arvia Technology Ltd. (UK based waste water treatment company). Nyex 1000 was being employed for a number of commercial applications dealing with the removal of organic contaminants from industrial effluents. This adsorbent material had small adsorptive capacity. With the aim to address small adsorptive capacity, a new graphite based adsorbent material was developed. The particle design was accomplished through successive chemical, thermal and mechanical treatments of raw graphite material (natural large flake graphite, to be called here as NLFG). The chemical treatment of the NLFG was carried out through electrochemical intercalation using dilute (50%) sulfuric acid in an electrochemical cell. Chemically treated NLFG then went through thermal treatment at 850 °C and followed by mechanical treatments consisting of compression (4536 kgf cm−2) and chopping at 18,000 rpm for 30 s. The developed adsorbent material, (exfoliated compacted graphite, to be called here as ECG) and NLFG were characterized using state of the art techniques including SEM, BET surface area, XRD, Zeta potential, Boehm surface titration, bed electrical conductivity and laser size analysis. The characterization results showed significant increase in internal specific surface area from 1 to 17 m2 g−1. It was attributed to the development of partially porous particle surface verified by SEM results. The XRD, Boehm surface titration, Zeta potential results endorsed the associated chemical and physical changes appeared in the composition of the NLFG as a result of chemical, thermal and mechanical treatments. Adsorption-regeneration studies were conducted using developed ECG and existing Nyex 1000 materials. The pollutants used for adsorption–regeneration studies were acid violet 17, phenol, humic acid, ethane thiol and methyl propane thiol dissolved in aqueous solution. The results were compared and it was found that ECG showed significantly improved adsorption capacity with many folds. Both adsorbent materials, ECG and Nyex 1000 delivered 100% electrochemical regeneration efficiencies.

        SUNSHINE, EARTHSHINE AND CLIMATE CHANGE: II. SOLAR ORIGINS OF VARIATIONS IN THE EARTH'S ALBEDO

        There are terrestrial signatures of the solar activity cycle in ice core data (Ram & Stoltz 1999), but the variations in the sun's irradiance over the cycle seem too small to account for the signature (Lean 1997; Goode & Dziembowski 2003). Thus, one would expect that the signature must arise from an indirect effect(s) of solar activity. Such an indirect effect would be expected to manifest itself in the earth's reflectance. Further, the earth's climate depends directly on the albedo. Continuous observations of the earthshine have been carried out from Big Bear Solar Observatory since December 1998, with some more sporadic measurements made during the years 1994 and 1995. We have determined the annual albedos both from our observations and from simulations utilizing the Earth Radiation Budget Experiment (ERBE) scene model and various datasets for the cloud cover, as well as snow and ice cover. With these, we look for inter-annual and longer-term changes in the earth's total reflectance, or Bond albedo. We find that both our observations and simulations indicate that the albedo was significantly higher during 1994-1995 (activity minimum) than for the more recent period covering 1999-2001 (activity maximum). However, the sizes of the changes seem somewhat discrepant. Possible indirect solar influences on the earth's Bond albedo are discussed to emphasize that our earthshine data are already sufficiently precise to detect, if they occur, any meaningful changes in the earth's reflectance. Still greater precision will occur as we expand our single site observations to a global network.

        Chronic response of rat brain nirepinephrine and serotonin levels to endurance training. J. Appl. Physiol.: Respirat. Environ. Exercis Physiol. 46(1(:19-23, 1979. - Norepinephrine(NE)and serotonin(5-HT) concentrations were determined in three area of the rat brain following 8자 exposure to differing physical activity and dietary regimens. Animals were divided into sedentary and exercise groups, and one-half of each received either a normal or atherogenic diet. The exercise consisted of 30min of treadmill running, 5days/자 Brains were weighed and sectioned into cerebral cortex, cerebellum, and remainder(midbrain), and Iluorometrically analyzed for NE and 5-HT content. In most brain areas, NE and 5-HT levels were significantly greater among exercise-normal diet and exercise-fat dite compared to both sedentary groups. In the cerebral cortex, exercise-normal diet rats possessed higher levels of NE and lower concentrations of 5-HT than sedentary animals.

      • The mechanism of Torsin ATPase activation

        <P><B>Significance</B></P><P>Torsin activity critically depends on accessory cofactors that activate their ATPase activity by a poorly understood mechanism. This study establishes the mechanistic framework for Torsin activation, which relies on a complementation of the fragmentary Torsin active site. Given these unusual properties and the fact that Torsin activation is dysregulated in the congenital movement disorder primary dystonia, our results suggest that pharmacological manipulation of Torsin activation may present a novel therapeutic opportunity.</P><P>Torsins are membrane-associated ATPases whose activity is dependent on two activating cofactors, lamina-associated polypeptide 1 (LAP1) and luminal domain-like LAP1 (LULL1). The mechanism by which these cofactors regulate Torsin activity has so far remained elusive. In this study, we identify a conserved domain in these activators that is predicted to adopt a fold resembling an AAA+ (ATPase associated with a variety of cellular activities) domain. Within these domains, a strictly conserved Arg residue present in both activating cofactors, but notably missing in Torsins, aligns with a key catalytic Arg found in AAA+ proteins. We demonstrate that cofactors and Torsins associate to form heterooligomeric assemblies with a defined Torsin–activator interface. In this arrangement, the highly conserved Arg residue present in either cofactor comes into close proximity with the nucleotide bound in the neighboring Torsin subunit. Because this invariant Arg is strictly required to stimulate Torsin ATPase activity but is dispensable for Torsin binding, we propose that LAP1 and LULL1 regulate Torsin ATPase activity through an active site complementation mechanism.</P>
