Medaka Fish Parkinson’s Disease Model

The teleost fish has been widely used in creating neurodegenerative models. Here we describe the teleost medaka fish Parkinson’s disease (PD) models we developed using toxin treatment and genetic engineering. 1-Methyl-4-phenyl-1,2,3,4–tetrahydropyridine (MPTP), 6-hydroxydopamine (6-OHDA), proteasome inhibitors, lysosome inhibitors and tunicamycin treatment in our model fish replicated some salient features of PD: selective dopamine cell loss and reduced spontaneous movement with the last three toxins producing inclusion bodies ubiquitously in the brain. Despite the ubiquitous distribution of the inclusion bodies, the middle diencephalic dopaminergic neurons were particularly vulnerable to these toxins, supporting the idea that this dopamine cluster is similar to the human substantia nigra. PTEN-induced putative kinase 1 (PINK1) homozygous mutants also showed reduced spontaneous swimming movements. These data indicate that medaka fish can serve as a new model animal of PD. In this review we summarize our previous data and discuss future prospects

Purpose: The aim of this study was to elucidate whether intrasynovial corticosteroid injections for trigger digit reduced the volume of the tendon and pulley on high-resolution ultrasonography. Methods: Twenty-three digits of 20 patients with trigger digit were included. Each affected finger was graded clinically according to the following classification: grade I for pre-triggering, grade II for active triggering, grade III for passive triggering, and grade IV for presence of contracture. Axial ultrasound examinations were performed before an intrasynovial corticosteroid injection and at an average of 31 days after the injection. The transverse diameter, thickness, and crosssectional area of the tendon and the thickness of the pulley were measured by two independent, blinded researchers. Results: At least 1 grade of improvement was achieved in this study group by the time of the second examination. The transverse diameter and cross-sectional area of the tendon and the thickness of the pulley significantly decreased (P<0.05). Conclusion: The injection of a single dose of betamethasone improved clinical symptoms by reducing the volume of both the tendon and pulley, which may be related to the fact that tendon and pulley ruptures are delayed by corticosteroid injections.

Purpose: It has been suggested that resonance frequency analysis (RFA) can measure changes in the stability of dental implants during osseointegration. This retrospective study aimed to evaluate dental implant stability at the time of surgery (primary stability; PS) and secondary stability (SS) after ossseointegration using RFA, and to investigate the relationship between implant stability and cortical bone thickness. Methods: In total, 113 patients who attended the Tohoku University Hospital Dental Implant Center were included in this study. A total of 229 implants were placed in either the mandibular region (n=118) or the maxilla region (n=111), with bone augmentation procedures used in some cases. RFA was performed in 3 directions, and the lowest value was recorded. The preoperative thickness of cortical bone at the site of implant insertion was measured digitally using computed tomography, excluding cases of bone grafts and immediate implant placements. Results: The mean implant stability quotient (ISQ) was 69.34±9.43 for PS and 75.99±6.23 for SS. The mandibular group had significantly higher mean ISQ values than the maxillary group for both PS and SS (P<0.01). A significant difference was found in the mean ISQ values for PS between 1-stage and 2-stage surgery (P<0.5). The mean ISQ values in the non-augmentation group were higher than in the augmentation group for both PS and SS (P<0.01). A weak positive correlation was observed between cortical bone thickness and implant stability for both PS and SS in all cases (P<0.01). Conclusions: Based on the present study, the ISQ may be affected by implant position site, the use of a bone graft, and cortical bone thickness before implant therapy
Cell Lineage, Self-Renewal, and Epithelial-to-Mesenchymal Transition during Secondary Neurulation

Secondary neurulation (SN) is a critical process to form the neural tube in the posterior region of the body including the tail. SN is distinct from the anteriorly occurring primary neurulation (PN); whereas the PN proceeds by folding an epithelial neural plate, SN precursors arise from a specified epiblast by epithelial-to-mesenchymal transition (EMT), and undergo self-renewal in the tail bud. They finally differentiate into the neural tube through mesenchymal-to-epithelial transition (MET). We here overview recent progresses in the studies of SN with a particular focus on the regulation of cell lineage, self-renewal, and EMT/MET. Cellular mechanisms underlying SN help to understand the functional diversity of the tail in vertebrates.

<P>Interferon-tau (IFNT), a type I interferon (IFN), is known as pregnancy recognition signaling molecule secreted from the ruminant conceptus during the preimplantation period. Type I IFNs, such as IFN-alpha and IFN-beta, are known to activate cell-death pathways as well as induce apoptosis. In cows, induction of apoptosis with DNA fragmentation is induced by IFNT in cultured bovine endometrial epithelial cells. However, the status of cell-death pathways in the bovine endometrium during the preimplantation period still remains unclear. In the present study, we investigated the different cell-death pathways, including apoptosis, pyroptosis, and autophagy, in uterine tissue obtained from pregnant cows and <I>in vitro</I> cultured endometrial epithelial cells with IFNT stimulation. The expression of <I>CASP7</I>, <I>8,</I> and <I>FADD</I> (apoptosis-related genes) was significantly higher in pregnant day 18 uterine tissue in comparison to non-pregnant day 18 tissue. The expression of <I>CASP4</I>, <I>11,</I> and <I>NLRP3</I> (pyroptosis-related genes) was significantly higher in the pregnant uterus in comparison to non-pregnant uterus. In contrast, autophagy-related genes were not affected by pregnancy. We also investigated the effect of IFNT on the expression of cell-death pathway-related genes, as well as DNA fragmentation in cultured endometrial epithelial cells. Similar to its effects in pregnant uterine tissue, IFNT affected the increase of apoptosis-related (<I>CASP8</I>) and pyroptosis-related genes (<I>CASP11</I>), but did not affect autophagy-related gene expression. IFNT also increased γH2AX-positive cells, which is a marker of DNA fragmentation. These results suggest that apoptosis- and pyroptosis-related genes are induced by IFNT in the pregnant bovine endometrial epithelial cells.</P>

OPTICAL AND NEAR-INFRARED POLARIMETRY FOR A HIGHLY DORMANT COMET 209P/LINEAR

<P>We conducted an optical and near-infrared polarimetric observation of the highly dormant Jupiter-Family Comet, 209P/LINEAR. Because of its low activity, we were able to determine the linear polarization degrees of the coma dust particles and nucleus independently, that is P-n = 30.3(-0.9)(+1.3)% at alpha = 92 degrees.2 and P-n = 31.0(-0.7)(+1.0)% at alpha = 99 degrees.5 for the nucleus, and P-c = 28.8(-0.4)(+0.4)% at alpha = 92 degrees.2 and 29.6(-0.3)(+0.3)% at alpha = 99 degrees.5 for the coma. We detected no significant variation in P at the phase angle coverage of 92 degrees.2-99 degrees.5, which may imply that the obtained polarization degrees are nearly at maximum in the phase-polarization curves. By fitting with an empirical function, we obtained the maximum values of linear polarization degrees P-max = 30.8% for the nucleus and P-max = 29.6% for the dust coma. The P-max of the dust coma is consistent with those of dust-rich comets. The low geometric albedo of P-v. =. 0.05 was derived from the slope-albedo relationship and was associated with high P-max. We examined P-max-albedo relations between asteroids and 209P, and found that the so-called Umov law seems to be applicable on this cometary surface.</P>
OPTICAL-INFRARED AND HIGH-ENERGY ASTRONOMY COLLABORATION AT HIROSHIMA ASTROPHYSICAL SCIENCE CENTER

The Hiroshima Astrophysical Science Center (HASC) was founded in 2004 at Hiroshima University, Japan. The main mission of this institute is the observational study of various transient objects including gamma-ray bursts, supernovae, novae, cataclysmic variables, and active galactic nuclei by means of multi-wavelength observations. HASC consists of three divisions; the optical-infrared astronomy division, high-energy astronomy division, and theoretical astronomy division. HASC is operating the 1.5m optical-infrared telescope Kanata, which is dedicated to follow-up and monitoring observations of transient objects. The high-energy division is the key operation center for the Fermi gamma-ray space telescope. HASC and the high-energy astronomy group in the department of physical science at Hiroshima University are closely collaborating with each other to promote multi-wavelength time-domain astronomy. We report the recent activities of HASC and some science topics pursued by this multi-wavelength collaboration.
