      • Effect of a Fluorescent Dye Conjugated with a Cell Penetrating Peptide for Cancer Therapy

        We experienced cancer therapy using conjugated CPPs with ZW800-1 leaded to instant cell death like necrosis. The coupling reaction of ZW800-1, to clinical protamine (Pro) was performed to synthesis the new type of agent .The reaction between clinical protamine (Pro) and an NHS ester of dye is a solution phase reaction with the complete modification of the protamine peptides which feature a single reactive amine at the N-terminal and single carboxyl group at the C-terminal . cell death studies by conjugation of a protamine with ZW800-1 shows that a new types of agent has more efficient cytotoxic activities than each material alone, causing rapid cell death. This study aimed to identify a new type of agent for cancer cell therapy and to assess the antitumor effects of the newly synthesized agent using the Protamineconjugated ZW800-1 on a cancer cell line.

      • Mutation Analysis of IDH1/2 Genes in Unselected De novo Acute Myeloid Leukaemia Patients in India - Identification of A Novel IDH2 Mutation

        IDH1/2 mutations which result in alternation in DNA methylation pattern are one of the most common methylation associated mutations in Acute myeloid leukaemia. IDH1/2 mutations frequently associated with higher platelet level, normal cytogentics and NPM1 mutations. Here we analyzed IDH1/2 mutations in 200 newly diagnosed unselected Indian adult AML patients and investigated their correlation with clinical, cytogenetic parameters along with cooperating NPM1 mutation. We detected 5.5% and 4% mutations in IDH1/2 genes, respectively. Except IDH2 c.515_516GG>AA mutation, all the other identified mutations were reported mutations. Similar to reported c.515G>A mutation, the novel c.515_516GG>AA mutation replaces $172^{nd}$ arginine to lysine in the active site of the enzyme. Even though there was a preponderance of IDH1/2 mutations in NK-AML, cytogenetically abnormal patients also harboured IDH1/2 mutations. IDH1 mutations showed significant higher platelet count and NPM1 mutations. IDH2 mutated patients displayed infrequent NPM1 mutations and lower WBC count. All the NPM1 mutations in the IDH1/2 mutated cases showed type A mutation. The present data suggest that IDH1/2 mutations are associated with normal cytogenetics and type A NPM1 mutations in adult Indian AML patients.

        Agnihotra Yajna: A Prototype of South Asian Traditional Medical Knowledge

        This study conceptualizes the principle of agnihotra yajna. The perusal of ancient and modern literature reveals that the functioning of the human body is impossible without maintaining an energetic continuum driven by sunlight. The seven major chakras existing over the spinal cord help to maintain this energetic continuum. Agnihotra yajna is proposed to balance the chakra system as a whole by minimizing entropy. Offerings of natural elements to fire lit in a copper pyramid during agnihotra liberate various volatile compounds having potent pharmacological actions. Attempts were made to enhance the efficacy of fumes by incorporating two to three pieces of coconut endosperm and “navadhanya” (nine grains) to the conventional fire oblations. This investigation clearly demonstrates that the purpose behind the practice of agnihotra yajna is “letting incessant flow of energy (LIFE)” through our meridian lines and acupuncture points. The volatile organic compounds in smoke were analyzed using the gas chromatographyemass spectrometry method, and the results are discussed.

        Therapeutic Role of Yoga in Type 2 Diabetes

        Yoga originated in India more than 5,000 years ago and is a means of balancing and harmonizing the body, mind, and emotions. Yoga practice is useful in the management of various lifestyle diseases, including type 2 diabetes. Psycho-neuro-endocrine and immunemechanisms are involved in the beneficial effects of yoga on diabetes. Incorporation of yoga practice in daily life helps to attainglycaemic control and reduces the risk of complications in people with diabetes. In this review, we briefly describe the role ofvarious yoga practices in the management of diabetes based on evidence from various clinical studies.

        Experimental studies on domestic refrigeration system with brazed plate heat exchanger as condenser

        In view of Kyoto protocol, there is a pressing need to reduce the energy consumption and environmental impacts of domestic appliances. In the total energy consumption of household appliances, domestic refrigerator plays a vital role. Alternate refrigerants and improvement in the performance of the components can contribute to tackle the above issue in a domestic refrigeration system. In this paper, the performance of a domestic refrigeration system with brazed plate heat exchanger as condenser, and working with refrigerants such as R290/R600a and R134a was studied using experimental method. The result showed that the system with water-cooled brazed plate heat exchanger reduces the per day energy consumption of a system from 21% to 27% and increases the COP from 52% to 68%, when compared to conventional system. The compressor discharge temperature and dome temperature are also dampened. For R134a and R290/R600a, the TEWI of the system with water-cooled brazed plate heat exchanger is lower than that of the system with air-cooled condenser by 26.8% and 21%, respectively. Among the refrigerants, R290/R600a showed higher performance than R134a.

        Development of nanosilver embedded injectable liquid crystalline hydrogel from alginate and chitosan for potent antibacterial and anticancer applications

        Injectable hydrogels with macroscopic ordering have gained special attention in drug delivery due totheir excellent biocompatibility and minimally invasive surgical procedure. Nanosilver embeddedSchiff base hydrogel from biopolymers alginate and chitosan was developed and demonstrated its applicationin the injectable delivery of anticancer drug Dox.HCl. The ordered hydrogel structure was evidencedby FT-IR, DLS, XRD and Rheology. The self-assembled morphology of nanosilver- Schiff basehydrogels was studied by microscopic techniques such as SEM, TEM and AFM. The pH dependent anticanceractivity of the hydrogel was demonstrated by invitro drug release studies at pH 5.5 and 86.56 %release was obtained for 100 mgL-1 of Dox.HCl loaded hydrogel at 72 hours. The antibacterial activityattained from nanosilver can be assessed by Agar well diffusion assay and minimum inhibitory concentrationand minimum biocidal concentrations were determined as 62.5 lL/mL and 125 lL/mL, respectively. The invitro cell viability studies were done against skin cancer cell B16 by MTT assay whichshows 34.82 % of toxicity towards B16 cancer cells and the cell death was further confirmed byApoptosis assay and flow cytometry. The developed macroscopically ordered liquid crystalline hydrogelcan be act as a versatile platform for injectable drug delivery system.

      • Improvement of photodynamic therapy by conjugation of a cell-penetrating peptide with Methylene Blue

        Photodynamic therapy (PDT) causes tissue destruction by visible light in the presence of a photosensitizer (Ps) and oxygen, which promises alternative therapy that could be used an adjunct to chemotherapy and surgery for oncology. Methylene blue dye alone permeates into the cell and distributes through the cytoplasm, Protamine is one of the cell-penetrating peptides (CPPs), which has high arginine peptide with membrane translocating and nuclear localizing properties. So it makes localized to a specific spot of the cell by endocytosis and the localized conjugates enhances the generation of ROS enough to induce cell damages rapidly. In this study, we introduce the new type of PS and explore the effectiveness of PDT using MB-Pro in human colon cell lines, the new types of MB-Pro show more efficient photodynamic activities than MB alone, causing rapid and effective cell death.

        Introgression of Sub1 QTL alters aerenchyma-mediated gas exchange and stored carbohydrates to maintain yield under flooding stress in rice

        Floods can completely submerge rice felds. When rice is submerged, various anatomical and physiological changes occur at cellular and whole plant levels. New submergence tolerant varieties have been developed for the beneft of farmers in fooded areas. The present investigation comprised of evaluating a Sub 1introgressed CO 43 variety (CO 43 Sub 1 Nil) along with its donor (FR13A) and cultivated variety CO 43, for diferent anatomical and physiological traits underlying fooding stress tolerance. The results revealed that one of the adaptive trait of rice is the constitutive development of aerenchyma when submerged. It also showed that lesser porosity or air cavities in the leaf, stem and root aerenchymas of the tolerant varieties (FR13A and CO43 Sub 1 Nil) showed limited gas exchange and conserved the stored carbohydrates. The susceptible variety (CO 43) showed higher percentage increase in porosity in all the tissues studied. The plants also showed limited underwater elongation and higher survival percentage for seven days of fooding stress. The yield advantage of CO 43 Sub 1 Nil showed a two fold increase in all the yield components compared to CO 43 following submergence. This was due to its ability to utilize the stored carbohydrate immediately after withdrawal of stress to enhance photosynthetic rate and conductance, for greater biomass and yield. A strong positive correlation exists between leaf and stem carbohydrate contents and the survival percentage before and after submergence. The paper highlights the role of aerenchyma in regulating the gas exchange and stored carbohydrates in improving the tolerance of CO 43 Sub 1 Nil to fooding stress.

      • Characterization of CEBPA Mutations and Polymorphisms and their Prognostic Relevance in De Novo Acute Myeloid Leukemia Patients

        The CCAAT/enhancer-binding protein-alpha (CEBPA) is a transcriptional factor that plays a crucial role in the control of proliferation and differentiation of myeloid precursors. This gene was recognized as the target of genetic alterations and were associated with clinical complexity among AML. We here analyze the frequency and types of CEBPA mutations and polymorphisms in a de novo AML patients from South India and tried to find out associations of these variations with different clinical parameters and the prognostic significance in AML. Study was carried out in 248 de novo AML patients, cytogenetic analysis was performed from the bone marrow samples and was karyotyped. PCR-SSCP analysis and sequencing was performed for the detection of CEBPA gene variations. All the statistical analysis was performed using SPSS 17 (statistical package for social sciences) software. Pearson Chi-square test, Mann-Whitney U test, Kaplan-Meier survival analysis and log rank tests were performed. CEBPA mutations were detected in 18% and CEBPA polymorphisms were detected in 18.9% of AML cases studied. Most of the mutations occured at the C terminal region. Polymorphisms were detected in both N and C terminal region. with most common being, c.584_589dup ACCCGC and c.690G>T. A significant association was not observed for the mutation and polymorphism with respect to clinical and laboratory parameters. Survival advantage was observed for the mutated cases compared to non mutated cases, especially for the normal karyotype groups. Polymorphisms has no effect on the survival pattern of AML patients. CEBPA mutation and polymorphisms were observed with similar frequency and was identified in all the FAB subtypes as well as in cytogenetic risk groups in our study population, but CEBPA mutations alone confer a prognostic value for NK AML patients.

        Role of Yoga in the Management of Premature Ejaculation

        Premature ejaculation (PE) is a common male sexual problem. Various non-pharmacological and pharmacological treatment options are available currently. The importance of yoga in the management of various sexual dysfunctions is increasingly recognized. In this review, we are analyzing the role of yoga in the management of PE. This paper explores the mechanism of yoga practices in the management of PE and the recommended yoga practice for PE. It also discusses the scientific evidence of yoga practices, such as yoga postures (yogasana), breathing practices (pranayama), lock (bandha), gestures (mudra), relaxation, Aum chanting, yoga nidra and meditation with particular reference the management of PE.
