        High frequency direct plant regeneration from leaf, internode, and root segments of Eastern Cottonwood (Populus deltoides)

        Simple, reproducible, high frequency, improved plant regeneration protocol in Eastern Cottonwood (Populus deltoides) clones, WIMCO199 and L34, has been reported. Initially, aseptic cultures established from axillary buds of nodal segments from mature plus trees on MS liquid medium supplemented with 0.25 mg l-1 KIN and 0.25 mg l-1 IAA. Nodal and internodal segments were found to be extraprolific over shoot apices during course of aseptic culture establishment, while 0.25 mg l-1 KIN concentration played a stimulatory role in high frequency plant regeneration. Diverse explants, such as various leaf segments, internodes, and roots from in vitro raised cultures, were employed. Direct plant regeneration was at high frequency of 92% in internodes, 88% in leaf segments, and 43% in root segments. This led to the formation of multiple shoot clusters on established culture media with rapid proliferation rates. Many-fold enhanced shoot elongation and growth of the clusters could be achieved on liquid MS medium supplemented with borosilicate glass beads, which offer physical support for proliferating shoots leading to faster growth in comparison to semi-solid agar or direct liquid medium. SEM examination of initial cultures confirmed direct plant regeneration events without intervening calli. In vitro regenerated plants induced roots on half-strength MS medium with 0.15 mg l-1 IAA. Rooted 5- to 6-week-old in vitro regenerated plants were transferred into a transgenic greenhouse in pots containing 1:1 mixture of vermicompost and soil at 27 ± 2C for hardening and acclimatization. 14- to 15-week-old well-established hardened plants were transplanted to the field and grown to maturity. The mature in vitro raised poplar trees exhibited a high survival rate of 85%; 4-year-old healthy trees attained an average height of 8 m and an average trunk diameter of 25 cm and have performed well under field conditions. The regeneration protocol presented here will be very useful for undertaking genetic manipulation, providing a value addition to Eastern Cottonwood propagation in future.         Theoretical and experimental investigation on structural stability, electronic and vibrational properties of polyaniline (PANI)

        Conducting polymers have been concerned the ample interest of scientific community due to its special physical and chemical properties. Among this group of materials, the Polyaniline (PANI) merits special attention due to its high thermal and chemical stability under normal conditions, simple process of polymerization low cost and well-studied electrochemical characteristics. In the present work, Density functional theory (DFT) method has been applied for geometry optimization using B3LYP/6-31G** basis set, implemented on Gaussian 09 software package. The well-known model, time-dependent DFT (TDDFT) calculations at the UB3LYP/6-31G(d) level have been performed to investigate the tunable nature, i.e., doping processes, of polyaniline (PANI). The calculated theoretical data show strong correlation with the recent experimental reports, which validates our computational protocol. The calculated properties are extrapolated to the polymer (PANI) through a second-order polynomial fit. Changes in band gap, conductivity, and resistance of (PANI_n) and (PANI_n) (where n = 1-4 and) were studied. Further we have also synthesized and characterized the Polyaniline. The deviations in the obtained results have clearly shown the promising of Polyaniline as a quantity sensitive material that may be useful for the sensing application.

        Real world clinical outcomes of adjuvant sequential chemoradiation in patients with gallbladder carcinomas with poor performance status

        Purpose: The purpose of the study is to analyze the overall survival, relapse-free survival, and relapse patterns of adjuvant sequential chemoradiation for gallbladder cancers after curative resection in patients with poor performance status. Materials and Methods: We retrospectively reviewed clinical records of gallbladder patients with pathologic stage T2-4 or node positivity treated with sequential chemoradiation at our institute between January 2015 and January 2019. Sequential chemoradiotherapy protocol consisted of six cycles of gemcitabine 1,000 mg/m2 and oxaliplatin 100 mg/m2 administered every 2 weekly and postoperative radiation therapy (45 Gy in 25 fractions over 5 weeks) by three-dimensional conformal technique. Results: A total of 36 patients were included. The median overall survival and relapse-free survival was 26 months (95% confidence interval [CI], 21.4-30.5) and 21 months (95% CI, 11.8-30.1), respectively. The 2-year overall and relapse-free survival rates were 55.1% (95% CI, 37.9%-72.3%) and 44.7% (95% CI, 27.5%-61.9%), respectively. Locoregional, systemic, and combined recurrence were noted in 2 (5.5%), 14 (38.8%), and 3 (8.3%) patients, respectively. On univariate analysis, tumour grading significantly influenced relapse free survival; nodal stage and overall stage demonstrated a statistically significant influence on overall survival (p < 0.05) with a trend towards significance for lymphovascular invasion. On multivariate analysis, no significant factors were found. Grade 3 and 4 haematological adverse events were observed only in 2 (5.5%) with chemotherapy. No grade 3 and 4 adverse events were observed due to radiation therapy. Conclusion: Sequential chemoradiation is feasible and tolerable with acceptable efficacy in the adjuvant setting in patients unfit for concurrent chemoradiotherapy.

        Episodes of prolonged “trance-like state” in an infant with hypothalamic hamartoma

        Hypothalamic hamartoma (HH) is one of the most common causes of central precocious puberty (CPP) in first few years of life. It can present with either seizures or CPP, although both manifestations coexist in the majority of the children. Gelastic seizures (GS), or laughing spells, are usually the first type of seizures seen in patients with HH. Although a wide variety of seizure types are known to occur in children with HH, GS are most common and consistent seizure type. The clinical presentation of HH may vary with the size and position of the mass, although large tumours typically present with both CPP and seizures. Although CPP can be managed with medical therapy, seizures can be very difficult to treat, even with multiple antiepileptic drugs. Noninvasive gamma knife surgery has been used with some success for the treatment of refractory epilepsy. We present a case of HH with very early onset seizures and CPP. The patient had an atypical form of seizures described by the parents as a "trance-like state" in which the patient had prolonged episodes of unresponsiveness lasting for hours with normal feedings during the episodes. GS occurred late in the course and were refractory to various combinations of antiepileptic drugs. A brain magnetic resonance imaging showed a large sessile HH (>20 mm). Later in the course of the disease, the patient experienced cognitive and behavioural problems. The patient underwent gamma knife surgery at nearly 5 years of age and experienced a modest response in seizure frequency. This case highlights the presentation of HH as a previously unreported seizure morphology described as a prolonged "trance-like state."

        Reactive oxygen species (ROS), highly reactive molecules, are produced by living organisms as a result of normal cellular metabolism and environmental factors, and can damage nucleic acids and proteins, thereby altering their functions. The human body has several mechanisms to counteract oxidative stress by producing antioxidants. A shift in the balance between oxidants and antioxidants in favor of oxidants is termed as "oxidative stress". Paradoxically, there is a large body of research demonstrating the general effect of oxidative stress on signaling pathways, less is known about the initial and direct regulation of signaling molecules by ROS, or what we term the "oxidative interface." This review focuses on the molecular mechanisms through which ROS directly interact with critical signaling molecules to initiate signaling in a broad variety of cellular processes, such as proliferation and survival (MAP kinases and PI3 kinase), ROS homeostasis, and antioxidant gene regulation (Ref-1 and Nrf-2). This review also deals with classification as well as mechanisms of formation of free radicals, examining their beneficial and deleterious effects on cellular activities and focusing on the potential role of antioxidants in preventing and repairing damage caused by oxidative stress. A discussion of the role of phytochemical antioxidants in oxidative stress, disease and the epigenome is included.

        Role of Riboflavin in Induced Resistance against Fusarium Wilt and Charcoal Rot Diseases of Chickpea

        Riboflavin caused induction of systemic resistance inchickpea against Fusarium wilt and charcoal rotdiseases. The dose effect of 0.01 to 20 mM riboflavinshowed that 1.0 mM concentration was sufficient formaximum induction of resistance; higher concentrationdid not increase the effect. At this concentration,riboflavin neither caused cell death of the host plant nordirectly affected the pathogen’s growth. In time courseobservation, it was observed that riboflavin treatedchickpea plants were inducing resistance 2 days aftertreatment and reached its maximum level from 5 to 7days and then decreased. Riboflavin had no effect onsalicylic acid (SA) levels in chickpea, however, riboflavininduced plants found accumulation of phenolsand a greater activities of phenylalanine ammonia lyase(PAL) and pathogenesis related (PR) protein, peroxidasewas observed in induced plant than the control. Riboflavin pre-treated plants challenged with thepathogens exhibited maximum activity of the peroxidases4 days after treatment. Molecular weight of thepurified peroxidase was 42 kDa. From these studies wedemonstrated that riboflavin induced resistance is PRproteinmediated but is independent of salicylic acid.

        Molecular and Morphophysiological Characterization of Superior Cluster Bean (Cymopsis tetragonoloba) Varieties

        Cluster bean (Cymopsis tetragonoloba) belongs to tribe Indigoferae of family Leguminosae. India is the world-leader for cluster bean production as it contributes 80% shares of its total production. Cluster bean (guar) is a cash crop for its application in textile, paper, petroleum, mining, pharmaceuticals, explosives, and food industries. Owing to its immense wealth of variable morphophysiological and industrial qualities there is a strong need for appropriate addressing and well documentation of the germplasm. Efforts are to be made to organize research programs on germplasm characterization, utilization, and molecular characterization. Superior cluster bean varieties were selected on the basis of morphophysiological characters and subjected to DNA-based molecular marker analysis. Eighteen of the best genotypes were chosen for DNA extraction, optimization of PCR conditions, and genetic diversity studies using 37 random primers. A total of 381 random amplification fragments were obtained; number of amplifications ranging from 4 to 22 with an average of 10.29 amplified fragments per primer. Evaluation of RAPD data reveals a magnificent range (0.34-0.76) of genotypic similarity coefficients. The UPGMA dendrogram was constructed based on similarity indices which illustrated discrete clustering of different genotypes into groups. Results recorded a positive correlation amongst varieties vis-à-vis cluster analysis generated by NTSYSpc and morphophysiological characteristics. The degree and distribution of genetic diversity in cluster bean would facilitate an evolutionary relationship between numerous accessions that eventually catalogues genetic resources in a more concerted fashion.       • Comparison of Spectrum of Complications after Pancreaticoduodenectomy in Patients with or without Preoperative Biliary Stents

        Aims: Preoperative biliary drainage (PBD) prior to Pancraticoduodenectomy (PD) continues to be routine in many centres despite sufficient evidence showing PBD to increase perioperative complications. This study was planned to see complications of PD and compare between stented and non-stented. Methods: Total 59 patients were enrolled in a period of one and half year. During surgery bile aspirate was sent routinely for culture sensitivity. Drain fluid amylase and LFT were obtained on day 3, 7 and 10. Morbidity was graded according to the Clavien-Dindo classification. Pancreatic fistula, haemorrhage and DGE were documented as per international guidelins. USG Abdomen was done on POD7. Patients were assessed daily for complications as per ISGPS definition. Results: Fifty Nine patients were enrolled. 21 were stented and 38 were not stented. Median age was 62 and 55 respectively.. DGE in 13 stented and 24 non-stented patients. Pancreatic fistula occurred in 7 (33%) stented and 8(21%) non -stented group. No significant difference was found in two groups.Fever was documented in 5 stented and 4 non-stented . Post PD haemorrhage occured in three patients and no difference between scented and non-stented. Bile culture was sterile in Two (9.5%) stented and 28 (73%) non stented patients. This was significant. Enterococci were most common isolate on stented patients accounting for 33% . E coli were most common among non-stetted . Conclusions: We could not find significant difference in morbidity, hospital stay and operative time between the stented and non-stented groups after Pancreaticoduodenectomy. There was significant difference in bile culture positivity.

From these studies we demonstrated that riboflavin induced resistance is PR-protein mediated but is independent of salicylic acid.
