        The role of the apoptosis-related protein BCL-B in the regulation of mitophagy in hepatic stellate cells during the regression of liver fibrosis

        The clearance of activated hepatic stellate cells (HSCs) by apoptosis is critical for the reversibility of hepatic fibrosis. Mitochondrial homeostasis is regulated by mitophagy, which is an efficient way of clearing injured mitochondria that plays an important role in apoptosis. However, the role of mitophagy in apoptosis in HSCs and hepatic fibrosis is still unclear. Here, we show that mitophagy is enhanced in parallel with increased apoptosis in hepatic stellate cells during the reversal of hepatic fibrosis. The inhibition of mitophagy suppressed apoptosis in HSCs and aggravated hepatic fibrosis in mice. In contrast, the activation of mitophagy induced apoptosis in HSCs. Furthermore, we confirmed that BCL-B, which is a member of the BCL-2 family, is a regulator mediating mitophagy-related apoptosis. The knockdown of BCL-B resulted in increased apoptosis and mitophagy in HSCs, while the overexpression of BCL-B caused the opposite effects. BCL-B inhibited the phosphorylation of Parkin (a key regulator of mitophagy) and directly bound phospho-Parkin. Altogether, enhanced mitophagy promotes apoptosis in HSCs during the reversal of hepatic fibrosis. BCL-B suppresses mitophagy in HSCs by binding and suppressing phospho-Parkin, thereby inhibiting apoptosis. BCLB- dependent mitophagy is a new pathway for the regulation of apoptosis in HSCs during the regression of hepatic fibrosis

        Rs7574865 polymorphism of STAT4 and risk of anti-tuberculosis drug-induced hepatotoxicity in Chinese Han

        Anti-tuberculosis drug-induced hepatotoxicity (ATDH) is the most frequent, and potentially fatal adverse effect in the treatment of tuberculosis (TB). The rs7574865 polymorphism in the signal transducer and activator of transcription 4 gene (STAT4) was reported to be associated with drug-induced liver injury. However, there was no study aimed to this association in Chinese patients. The aim of this study was to investigate the influence of STAT4 polymorphisms on the susceptibility to ATDH in a Chinese Han population. A total of 280 TB patients with the prescription of anti-TB therapy, of Chinese Han origin, were enrolled. They were followed up for 3 months and demographic, clinical, laboratory and therapeutic data at each visit were collected. Two single nucleotide polymorphisms (SNPs) (rs7574865 and rs7582694) of STAT4 were genotyped with the MassARRAY platform. The associations between SNPs and ATDH risk were analyzed by logistic regression adjusting for confounding factors. A total of 24 patients were diagnosed with ATDH and considered as the case group, and 33 patients were lost to follow-up; the remaining 223 subjects without ATDH were considered as the control group. There was strong linkage disequilibrium (LD) between rs7574865 and rs7582694 ( r2 = 0.928 and D′ = 1). No significant association was found between SNPs or haplotypes of STAT4 and ATDH after correction for confounding factors. This prospective study is the first to investigate the association of genetic polymorphisms of STAT4 and ATDH in Chinese individuals. There was no significant association between the rs7574865 of STAT4 and ATDH in a Chinese Han population.

        BSA‑carbon dots a promising “off–on” fluorescence probe for detecting glyphosate residues in agricultural products

        The widespread and extensive use of glyphosate in agriculture has raised concerns about its potential impact on the quality and safety of agricultural products. Conventional detection methods require long analysis times, making them impractical for the rapid detection of large quantities of samples. Therefore, developing a fast and simple detection system for glyphosate pesticide residues is urgent. In this study, the development of a facile fluorescence probe synthesized using a simple one-pot hydrothermal method for the determination of glyphosate is an important step toward addressing the need for a fast and simple detection system. The present sensor was created using bovine serum albumin (BSA) as a precursor, and the sensor operates by producing an “off–on” fluorescent signal. The bovine albumin-derived BSA-CDs emitted light yellow fluorescence, but this fluorescence was quenched (or suppressed) by the presence of Cu2+ ions. However, the fluorescence can be restored by the presence of glyphosate, which interacts with the Cu2+ ions to form a complex and release the BSACDs from suppression. The functional groups in glyphosate can capture Cu2+ and break the BSA-CDs/Cu2+ combinatorial system. The BSA-CDs/Cu2+ fluorescence quenching system had good selectivity for glyphosate. The detection limit of the BSA-CD/Cu2+ fluorescence sensor was 0.05 μg/mL. This developed method was utilized to successfully detect glyphosate in Chinese wheat. The average recoveries ranged from 98.9 to 100.7%, with a relative standard deviation < 3.0%, showing good prospects for practical applicability.

        Hydraulic node model and parameter identification of a dull recoil brake

        This paper proposes a hydraulic node model for recoil brake. This model considers the compressibility and phase transition of recoil liquid by introducing a density-based piecewise equation of liquid state. For applying the hydraulic node model to a device named dull recoil brake, a least-squares parameter identification method based on the ant lion optimizer is used to determine the resistance coefficients. We measure the pressure responses of the dull recoil brake in firing tests on a 155 mm howitzer at two different working conditions and identify the resistance coefficients according to the pressure responses. Comparison between the simulations and the measurements verifies the hydraulic node model and the parameter identification result. This model shows remarkable performance in predicting the occurrence and magnitude of the hydraulic impacts. According to simulation results, we investigate the mechanism of the delayed retarding force and the secondary liquid impact. We find that the axial ending point of the crescent slots determines the moments when the recoil brake force rises and when the secondary liquid impact occurs.

        OTX1 Contributes to Hepatocellular Carcinoma Progression by Regulation of ERK/MAPK Pathway

        Orthodenticlehomeobox 1 (OTX1) overexpression had previously been associated with the progression of several tumors. The present study aimed to determine the expression and role of OTX1 in human hepatocellular carcinoma (HCC). The expression level of OTX1 was examined by quantitative real-time PCR (qRT-PCR) in 10 samples of HCC and paired adjacent non-cancerous tissues, and by immunohistochemistry (IHC) analysis in 128 HCC samples and matched controls. The relationship between OTX1 expression and the clinicopathological features werealso analyzed. Furthermore, the effects of OTX1 knockdown on cell proliferation and migration were determined in HCC cell lines. Axenograft mouse model was also established to investigate the role of OTX1 in HCC tumor growth. TheqRT-PCR and IHC analyses revealed that OTX1 was significantly elevated in HCC tissues compared with the paired non-cancerous controls. Expression of OTX1 was positively correlated with nodal metastasis status (P = 0.009) and TNM staging (P = 0.001) in HCC tissues. In addition, knockdown of OTX1 by shRNA significantly inhibited the proliferation and migration, and induced cell cycle arrest in S phase in vitro. Tumor growth was markedly inhibited by OTX1 silencing in the xenograft. Moreover, OTX1 silencing was causable for the decreased phosphorylation level of ERK/MAPK signaling. In conclusion, OTX1 contributes to HCC progression possibly by regulation of ERK/MAPK pathway. OTX1 may be a novel target for molecular therapy towards HCC.

        Robust Model Predictive Control for Uncertain Positive Time-delay Systems

        This paper proposes the robust model predictive control of positive time-delay systems with interval andpolytopic uncertainties, respectively. The model predictive control framework consists of linear constraint, linearperformance index, linear Lyapunov function, linear programming algorithm, and cone invariant set. By virtue ofmatrix decomposition technique, robust model predictive controllers of interval and polytopic positive systems withmultiple state delays are designed, respectively. A multi step control strategy is utilized and a cone invariant set isconstructed. Linear programming is used for the corresponding MPC conditions. Finally, a numerical example isgiven to verify the effectiveness of the proposed design.

        EFFECT OF MACROMOLECULAR COUPLING AGENT PMMA-b-PVTES ON STABILIZING NANO-Si3N4

        Poly(methyl methacrylate)-b-poly(vinyltriethoxysilane) (PMMA-b-PVTES) are synthesized using atom transfer radical polymerization (ATRP) and used as macromolecular coupling agents to modify silicon nitride nanoparticles (nano-Si3N4). The chemical composition of copolymers PMMA-b-PVTES and modified nano-Si3N4 are confirmed by various characterization techniques. The modified nano-Si3N4 shows excellent hydrophobic nature, which make nanoparticles (NPs) stably disperse in organic solvent. Transmission electron microscope (TEM), particle size testing, contact angle measuring and sedimentation experiment are employed to examine the dispersion of modified nano-Si3N4 in chloroform. By comparing the effects of copolymers with varied number-average molecular weight (Mn) and block length ratio on stabilizing nano-Si3N4, we discussed the mechanism of macromolecular coupling agent stabilizing NPs. In our experiments, the copolymer PMMA88-b-PVTES17 is found to be the most effective macromolecular coupling agent for stabilizing nano-Si3N4.
