        Effect of Hypothyroidism on the Growth of Black Bengal Goats

        Hypothyroidism was induced in three age groups (8 months, 1 year and 2 years) of Black Bengal goats by administering thiourea subcutaneously for 30 days. From fourth day onwards the animals were observed to be in hypothyroid state. For 8 months age group, a control group of similar age was taken. For the 1 year and 2 years age groups there was no control group but the values obtained on the pre-treatment day were considered as control values. For confirming the establishment of hypothyroidism, peripheral plasma triiodothyronine $(T_3)$ and thyroxine $(T_4)$ were estimated in all the animals before and during the treatment period by radioimunoassay. In the thiourea treated 8 months age group the $T_3$ and $T_4$ levels were significantly (p<0.001) lower than those in the control group during the treatment period. In all the thiourea treated groups the $T_3$ and $T_4$ levels observed during the treatment period were significantly (p<0.01) lower than the pre-treatment levels. A nonsignificant loss in the body weight was observed in all the thiourea treated groups whereas there was a significant growth in the control group of animals. The study indicates that hypothyroidism has a negative effect on growth in goats.

        Photoluminescence, thermoluminescence and evaluation of some parameters of Dy<sup>3+</sup> activated Sr<sub>5</sub>(PO<sub>4</sub>)<sub>3</sub>F phosphor synthesized by sol-gel method

        In this study, we have synthesized Dy<SUP>3+</SUP> activated Sr<SUB>5</SUB>(PO<SUB>4</SUB>)<SUB>3</SUB>F (S-FAP) phosphors by sol-gel synthesis method. The synthesized phosphors were characterized by X-ray diffraction pattern (XRD), scanning electron microscopic (SEM), photoluminescence (PL) and thermoluminescence (TL) for structural, morphological and luminescent properties. Dy<SUP>3+</SUP> activated Sr<SUB>5</SUB>(PO<SUB>4</SUB>)<SUB>3</SUB>F phosphor shows its characteristic PL emission at 481 nm and 574 nm due to <SUP>4</SUP>F<SUB>9/2</SUB> → <SUP>6</SUP>H<SUB>15/2</SUB> and <SUP>4</SUP>F<SUB>9/2</SUB> → <SUP>6</SUP>H<SUB>13/2</SUB> transitions, respectively. TL characteristics of Dy<SUP>3+</SUP> doped Sr<SUB>5</SUB>(PO<SUB>4</SUB>)<SUB>3</SUB>F phosphors were taken after irradiation by <SUP>60</SUP>Co gamma exposure. Two separate TL peaks at 126 <SUP>o</SUP>C and 279 <SUP>o</SUP>C were observed in case of Dy<SUP>3+</SUP> doped phosphor. Sr<SUB>5</SUB>(PO<SUB>4</SUB>)<SUB>3</SUB>F:Dy<SUP>3+</SUP> phosphor was irradiated within a wide range of exposure of 50 Gy to 7 kGy doses. Linearity was found up to 2 kGy and thereafter TL response saturates. Fading study was also carried out over the duration of six weeks for Sr<SUB>5</SUB>(PO<SUB>4</SUB>)<SUB>3</SUB>F phosphor. Trapping parameters were calculated using Chen's peak shape method, initial rise method and various heating rate method. Glow curve is deconvoluted using computerized glow curve deconvolution program. TL sensitivity of Sr<SUB>5</SUB>(PO<SUB>4</SUB>)<SUB>3</SUB>F:Dy<SUP>3+</SUP> phosphor is found to be less by a factor of 2.08 than commercially used CaSO<SUB>4</SUB>:Dy phosphor. In order to identify the defect centers formed upon γ-ray irradiation in Sr<SUB>5</SUB>(PO<SUB>4</SUB>)<SUB>3</SUB>F:Dy<SUP>3+</SUP> phosphor, EPR measurements were carried out on un-irradiated and irradiated phosphors.

        A Key Metabolic Regulator of Bone and Cartilage Health

        Taurine, a cysteine-derived zwitterionic sulfonic acid, is a common ingredient in energy drinks and is naturally found in fish and other seafood. In humans, taurine is produced mainly in the liver, and it can also be obtained from food. In target tissues, such as the retina, heart, and skeletal muscle, it functions as an essential antioxidant, osmolyte, and antiapoptotic agent. Taurine is also involved in energy metabolism and calcium homeostasis. Taurine plays a considerable role in bone growth and development, and high-profile reports have demonstrated the importance of its metabolism for bone health. However, these reports have not been collated for more than 10 years. Therefore, this review focuses on taurine–bone interactions and covers recently discovered aspects of taurine’s effects on osteoblastogenesis, osteoclastogenesis, bone structure, and bone pathologies (e.g., osteoporosis and fracture healing), with due attention to the taurine–cartilage relationship.

        Subfertility in Males: An Important Cause of Bull Disposal in Bovines

        The study had two objectives, namely, to estimate the andrological disorders leading to disposal of Karan Fries (KF), Sahiwal cattle and Murrah buffalo bulls and to study the effect of various factors (species/breeds, season of birth and period of birth) on male reproductive parameters. Records on occurrence of subfertility problems and disposal pattern of bulls maintained at the National Dairy Research Institute herd were collected for 15 years (1991 to 2005). Percentage of bulls producing freezable semen was less in the crossbred cattle (58.46%) as compared to Sahiwal (81.69%) and Murrah bulls (81.05%). Various subfertility traits like poor libido and unacceptable seminal profile were found to be the significant reasons (p<0.01) for culling of the breeding bulls. Inadequate sex drive was the main contributing factor for bull disposal in Sahiwal (22.55%) and Murrah bulls (15.12%) whereas poor semen quality and freezability were most frequently observed in KF bulls (24.29 and 7.29 percent, respectively). Least squares analyses of different male reproductive parameters showed that species/breeds had significant effect (p<0.05) on all traits except for frozen semen production periods (FSPP). Periods of birth were significantly different (p<0.05) for all traits except for semen volume. Age at first semen collection (AFSC), age at first semen freezing (AFSF) and age at disposal (AD) were highest in Murrah, while frozen semen production period (FSPP) and semen production period (SPP) were highest in KF and lowest in Sahiwal. The age at first semen donation and breeding period could be reduced by introducing the bulls to training at an early age. These results revealed a declining trend in AFSC, AFSF, FSPP, SPP and AD, thereby indicating an improvement in reproductive performance over the years. The age at first semen donation in bovines can be reduced by introducing the young male calves to training at an early age, which could increase the dosage of semen obtained from each male.

        Study on Suitable Semen Additives Incorporation into the Extender Stored at Refrigerated Temperature

        The objective of this study was to compare the effect of Butylated Hydroxy Toluene (BHT), Pentoxifylline (PTX) and ${\alpha}$-tocopherol (Vit E) on semen quality parameters of Karan Fries bulls. The fortification of extender by various semen additives improves motility as well as fertility of spermatozoa. Split samples of 24 ejaculates of four Karan Fries bulls were extended in extender with or without various additives such as BHT, PTX and Vit E, and performance was evaluated at an interval of 0, 24, 48 and 72 h at refrigerated temperature (4-$7^{\circ}C$). Results of the present study revealed that addition of BHT, PTX and Vit E in extender improved sperm cell function, such as motility, viability, HOST, and acrosome integrity, as compared to the control during liquid storage up to 48 h of preservation at refrigerated temperature. There was no significant (p<0.05) difference between any of the additives up to 48 h of preservation. Overall, the results showed a significant (p<0.05) deterioration in motility after each storage interval. The results showed a significant deterioration in the acrosome integrity and plasma membrane integrity up to 48 h; subsequently, there was not much degradation of both the semen quality parameters. There was a significant increase in spermatozoal tail and total abnormality after each storage interval at refrigerator temperature (4 to $7^{\circ}C$); however, the head and mid-piece abnormalities were almost unaffected. Tail and total abnormality were least in extender fortified with BHT, PTX and Vit E at different hours of incubation as compared to the control. The addition of 1.5 mM BHT, 3.6 mM PTX and 1 mg/ml Vit E in the semen extender has more beneficial effect in terms of semen quality and preservability of spermatozoa.

        Isolation of an Oocyte Stimulatory Peptide from the Ovarian Follicular Fluid of Water Buffalo (Bubalus bubalis)

        Ovarian follicular fluid contains both stimulatory and inhibitory agents that influence the growth and maturation of oocyte. In the present study, an attempt was made to isolate and study the biological properties of ovarian follicular fluid peptide(s) in buffaloes. Bubaline ovarian follicular was made steroid- and cell-free. A protein fraction was obtained by saturation (30-35% level) of the follicular fluid with ammonium sulfate. The protein fraction was purified with Sephadex-G 50 gel filtration chromatography and a single peak was obtained in the eluant volume, which was lyophilized. SDS-PAGE of the lyophilized fraction revealed a single band and the molecular weight of the peptide was 26.6 kDa. The peptide stimulated the cumulus cell expansion and in vitro maturation rate of oocytes in buffaloes in a dose dependent manner when it was incorporated at different dose levels (0, 10, 25, 50, 100 and 1,000 ng $ml^{-1}$ of maturation medium). The basic culture medium consisted of TCM 199 with Bovine serum albumin (0.3%). The in vitro maturation rates were comparable to those obtained with a positive control medium (TCM 199+20 ng EGF $ml^{-1}$+steer serum (20%)). Further purification and biological assays may throw more light on the nature and functions of this peptide.

        <P>The comparison APMP.T-K4 is the regional extension of the CCT-K4: an intercomparison of the realizations of the freezing-points of Al (660.323 °C) and Ag (961.78 °C). The comparison was organized in two loops and four sub-loops with high temperature standard platinum resistance thermometers (HTSPRTs) as transfer thermometers in the freezing-point comparisons. The comparison involved eight APMP NMIs (KRISS, NMIJ, SCL, NMC, CMS, NIMT, SIRIM, NPL), and KRISS and NMIJ acted as linking laboratories to the CCT-K4.</P><P>The transfer HTSPRTs showed a strong drift during the transportation between the NMIs. In the case of the Ag freezing-point comparison, the comparison results were scattered much more than expected. In the APMP meeting held in 2009, the participants agreed that the Ag comparison results would be omitted in the report. It revealed that the measurement results at the Al freezing-point of participants were in agreement with the key comparison reference value of the CCT-K4 within 4 mK except for one laboratory.         <P>During the Space Telescope and Optical Reverberation Mapping Project observations of NGC 5548, the continuum and emission-line variability became decorrelated during the second half of the six-month-long observing campaign. Here we present Swift and Chandra X-ray spectra of NGC 5548 obtained as part of the campaign. The Swift spectra show that excess flux (relative to a power-law continuum) in the soft X-ray band appears before the start of the anomalous emission-line behavior, peaks during the period of the anomaly, and then declines. This is a model-independent result suggesting that the soft excess is related to the anomaly. We divide the Swift data into on-and off-anomaly spectra to characterize the soft excess via spectral fitting. The cause of the spectral differences is likely due to a change in the intrinsic spectrum rather than to variable obscuration or partial covering. The Chandra spectra have lower signal-to-noise ratios, but are consistent with the Swift data. Our preferred model of the soft excess is emission from an optically thick, warm Comptonizing corona, the effective optical depth of which increases during the anomaly. This model simultaneously explains all three observations: the UV emission-line flux decrease, the soft-excess increase, and the emission-line anomaly.</P>

        Effect of Buffalo Follicular Fluid Alone and in Combination with PMSG and M199 on in vitro Buffalo Oocyte Maturation

        The effect of replacement of in vitro maturation medium completely with the buffalo follicular fluid (buFF) on in vitro oocyte maturation of buffalo oocytes was studied. 5 to 8 buffalo cumulus oocyte complexes were cultured in a single drop with each of the eight media studied i.e., M199+steer serum (10% v/v), M199+steer serum (10% v/v)+PMSG, M199+buFF (10% v/v), M199+buFF (10% v/v)+PMSG, M199+buFF (50% v/v), M199+buFF (50% v/v)+ PMSG, buFF (100%) and buFF+PMSG at $39^{\circ}C$ and 5% $CO_2$ in air for 24 h. Supplementation of M199 with Steer serum alone resulted in IVM rate of 35% only. When the above medium was supplemented with PMSG, the maturation rate rallied to 82%. Significant increase in the maturation rates were observed when M199 was supplemented with increasing levels of buFF. A further increase in the maturation rate was also obtained when PMSG was incorporated into the medium of M199 supplemented with buFF. The rate of maturation was to the tune of 91% when oocytes were matured in buFF alone which was increased non significantly on the addition of PMSG. Highest maturation rate (97%) obtained with M199+buFF (50%v/v)+PMSG did not differ significantly from that obtained by either M199+buFF (10%v/v)+PMSG or buFF+PMSG. It is suggested that buFF alone without any supplementation can form the effective in vitro maturation medium for buffalo oocytes.

        Magnetism in Single Metalloorganic Complexes Formed by Atom Manipulation

        <P>The magnetic properties of molecular structures can be tailored by chemical synthesis or bottom-up assembly at the atomic scale. We used scanning tunneling microscopy to study charge and spin transfer in individual complexes of transition metals with the charge acceptor, tetracyanoethylene (TCNE). The complexes were formed on a thin insulator, Cu<SUB>2</SUB>N on Cu(100), by manipulation of individual atoms and molecules. The Cu<SUB>2</SUB>N layer decouples the complexes from Cu electron density, enabling direct imaging of the TCNE molecular orbitals as well as spin-flip inelastic electron tunneling spectroscopy. Results were obtained at low temperature down to 1 K and in magnetic fields up to 7 T in order to resolve splitting of spin states in the complexes. We also performed spin-polarized density functional theory calculations to compare with the experimental data. Our results indicate that charge transfer to TCNE leads to a change in spin magnitude, Kondo resonance, and magnetic anisotropy for the metal atoms.</P><P><B>Graphic Abstract</B> <IMG SRC='http://pubs.acs.org/appl/literatum/publisher/achs/journals/content/nalefd/2014/nalefd.2014.14.issue-3/nl404054v/production/images/medium/nl-2013-04054v_0007.gif'></P><P><A href='http://pubs.acs.org/doi/suppl/10.1021/nl404054v'>ACS Electronic Supporting Info</A></P>
