T-Helper Type 2 Cells Direct Antigen-Induced Eosinophilic Skin Inflammation in Mice
Osamu Kaminuma,Tomoe Nishimura,Noriko Kitamura,Mayumi Saeki,Takachika Hiroi,Akio Mori 대한천식알레르기학회 2018 Allergy, Asthma & Immunology Research Vol.10 No.1
Eosinophilic inflammation in combination with immunoglobulin E (IgE) production is a characteristic feature of atopic dermatitis. Although activated T-helper type (Th) 2 cells play critical roles in the local accumulation and activation of eosinophils, whether they induce eosinophilic skin inflammation, independent of the IgE-mediated pathway has been unclear. To address the functional role of T cells in allergic skin diseases, we herein transferred Th1/Th2-differentiated or naive DO11.10 T cells into unprimed BALB/c mice. Ovalbumin-specific Th2 cells, as well as eosinophils, accumulated in the skin upon antigen challenge, despite the absence of antigen-specific IgE. Neither antigen-specific Th1 nor naive T cells induced eosinophil accumulation, although Th1 cells by themselves migrated into the skin. Interleukin (IL)-4, IL-5, and eotaxin were specifically produced in the skin of antigen-challenged, Th2 cell-transferred mice, whereas interferon (IFN)-γ and regulated on activation, normal T cell expressed and secreted (RANTES) were preferentially produced in Th1 cells-transferred mice. Production of monocyte chemoattractant protein (MCP)-1 and MCP-3 was enhanced by both Th1 and Th2 cells. The accumulation of eosinophils and Th2 cells in the skin was suppressed by both dexamethasone and FK506, indicating an essential role of Th2 cells in eosinophil recruitment. We conclude that Th2 cells can induce eosinophilic infiltration into the skin in the absence of antigen-specific IgE.
Utilization of Food Sources Before and After the Tsunami in Nuttallia olivacea at Gamo Lagoon, Japan
Woo Seok Shin,Osamu Nishimura 대한환경공학회 2013 Environmental Engineering Research Vol.18 No.4
This study was conducted June 2011 at Gamo Lagoon, after tsunami of March 2011, to estimate food sources and utilization. The results show that the tsunami affected the sediment properties by changing the physical environmental alterations. The fatty acids of the gut content of Nuttallia olivacea mostly comprised the same organic matter found in the sediment. Fatty acids in the tissues showed mainly diatoms, bacteria, and dinoflagellates. That is, most of the food sources (i.e., diatoms, bacteria, dinoflagellates, macroalgae, and terrestrial organic matter) probably pass through the digestive system unharmed; however, terrestrial organic matter, which is refrac-tory to biochemical degradation, indicated a different assimilation trend between the gut content and the tissue. This result suggests that input of labile organic matter from the sediment may control selective metabolism in N. olivacea. From these results, although the physical environment of sediment characteristics by tsunami changed, the food utilization of N. olivacea suggested a better assimilation of selected components from the gut content, irrespective of physical alteration.

Objective: To retrospectively evaluate the frequency and risk factors for developing thrombus in a systemic vein such as the infrarenal inferior vena cava or the iliac vein, in which a balloon-occluded retrograde transvenous obliteration (B-RTO) catheter was indwelled. Materials and Methods: Forty-nine patients who underwent B-RTO for gastric varices were included in this study. The B-RTO procedure was performed from the right femoral vein, and the B-RTO catheter was retained overnight in all patients. Pre- and post-procedural CT scans were retrospectively compared in order to evaluate the development of thrombus in the systemic vein in which the catheter was indwelled. Additionally, several variables were analyzed to assess risk factors for thrombus in a systemic vein. Results: In all 49 patients (100%), B-RTO was technically successful, and in 46 patients (94%), complete thrombosis of the gastric varices was achieved. In 6 patients (12%), thrombus developed in the infrarenal inferior vena cava or the right common-external iliac vein. All thrombi lay longitudinally on the right side of the inferior vena cava or the right iliac vein. One of the aforementioned 6 patients required anticoagulation therapy. No symptoms suggestive of pulmonary embolism were observed. Prothrombin time-international normalized ratio and the addition of 5% ethanolamine oleate iopamidol, on the second day, were related to the development of thrombus. Conclusion: Development of a thrombus in a systemic vein such as the inferior vena cava or iliac vein, caused by indwelling of the B-RTO catheter, is relatively frequent. Physicians should be aware of the possibility of pulmonary embolism due to iliocaval thrombosis. Chemical Properties of Sediment in Nanakita Estuarine Tidal Flat
Woo Seok Shin,Yoshio Aikawa,Osamu Nishimura 대한환경공학회 2012 Environmental Engineering Research Vol.17 No.2
The spatial variation of organic matter sources in tidal flat sediment of the Nanakita River estuary, involving Gamo lagoon on the north-east coast of Honshu Island, Japan, was examined using carbon stable isotopes and fatty acid biomarkers. The spatial variation of total organic carbon (TOC) contents and δ13C values were highly variable in between the stations, such as sandy flat (1.3 mg/g, -21.0‰), sand-muddy flat (2.6 mg/g, -21.9‰), and muddy flat (24.9 mg/g, -25.9‰), respectively. Particularly, at the muddy flat, high TOC content and low δ13C value of the sediments indicated that the surface sediment was composed largely of terrestrial organic matter. Whereas, at the sandy flat and sand-muddy flat, the high ratios of diatom and bacteria biomarkers indicated the high contribution of abundant microorganism along with marine organic matter in sediment composition. From these results, it considered that the amount and origin of transported sedimentary organic matter indicated different characteristics in this study stations.
