        Histopathologic investigation of the effects of prostaglandin E2 administered by different methods on tooth movement and bone metabolism

        Objective: The aim of this study was to investigate and compare the in vivo effects of prostaglandin E2 (PGE2) administered by different methods on orthodontic tooth movement and bone metabolism macroscopically, histopatologically, and biochemically. Methods: Forty-five young adult New Zealand rabbits were randomly divided into 3 experimental groups (n = 10/group), 1 positive control group (n = 10), and 1 negative control group (n = 5). The experimental rabbits were fitted with springs exerting 20-g reciprocal force on the maxillary incisors and PGE2 (10 μg/mL) was administered by the intravenous, submucosal, or intraligamentous route after appliance insertion and on days 1, 3, 7, and 14 thereafter. All rabbits were sacrificed on day 21 and their premaxillae were resected for histologic evaluation. Results: Tooth movement was observed in the experimental and positive control groups, but the intraligamentous PGE2 group had the highest values of all analyzed parameters, including serum calcium and phosphorus levels and osteoclastic and osteoblastic populations (p < 0.001). Conclusions: Submucosal and intraligamentous PGE2 administration significantly increases orthodontic tooth movement and bone metabolism, but the intraligamentous route seems to be more effective.

        Genome-wide identification and comperative analysis of RuBisCo genes/proteins in Asteracea

        Asteraceae, the largest known plant family in the world, contains economically important species including ornamental plants. The most abundant enzyme on Earth, Ribulose-1,5-bisphosphate (RuBisCo), catalyzes the first stepof carbon assimilation in photosynthesis. In this study, we conducted sequence, physicochemical, phylogenetic, and three-dimensional comparative analyses of 22 RuBisCo proteins in the Asteraceae using various bioinformatic tools. The alignment results showed that all residues corresponding to the region between 48-179 amino acids were conserved in all species, except for just seven residues: G79, I80, S88, F90, F93, I135, and S136. The sequence lengths of the RuBisCo large subunits were between 163 and 477 amino acids, with an average length of 255 amino acids. Their molecular weights (Mw) ranged from 18341.8 to 52895.1 Da, with an average Mw of 26294.5 Da. We found that the most abundant amino acid residue was Gly, which comprised 9.88% of these RuBisCo proteins. Ala (8.17%) and Leu (8.53%) were also both abundant. The extinction coefficient of the RuBisCo proteins at 280 nm ranged from 27515 to 69830 M-1 cm-1. The instability index values ranged from 24.41 to 40.65, and only one protein (from Gymnarrhena micrantha) was unstable in vitro. The aliphatic index values ranged from 72.56 to 81.13, while the GRAVY values ranged from -0.394 to -0.179. A total of ten motifs were identified in the sequences of the RuBisCo proteins. Phylogenetic analysis revealed that the 22 RuBisCo proteins formed two main clades. A RAMPAGEanalysis revealed that 95.0-98.4% of residues were located in the favored region in 22 RuBisCo proteins. Sequence alignment and 3D analysis revealed that a catalytically important Lys residue was conserved among all the RuBisCoenzymes of Asteraceae. The results of this study provide insights into the fundamental characteristics of the RuBisCo proteins of the Asteraceae.

        Synthesis, Analgesic, and Anti-Inflammatory Activities of [6-(3,5-Dimethyl-4-Chloropyrazole- 1-yl)-3(2H)-Pyridazinon-2-yl]Acetamides

        A series of structurally diverse amide derivatives of [6-(3,5-dimethyl-4-chloro-pyrazole-1-yl)- 3(2H)-pyridazinone-2-yl]acetic acid were prepared and tested for their in vivo analgesic and anti-inflammatory activity by using p-benzoquinone-induced writhing test and carrageenaninduced hind paw edema model, respectively. The analgesic and anti-inflammatory activity of the compounds, 7c, 7d and 7k were found to be equipotent to aspirin (as an analgesic) and indometacin (as an anti-inflammatory drug), respectively. The other amide derivatives generally resulted in lower activity on comparision with reference compounds.

      • Experimental investigation of impact behaviour of shear deficient RC beam to column connection

        Reinforced concrete (RC) structures may be subjected to sudden dynamic impact loads such as explosions occurring for different reasons, the collision of masses driven by rockfall, flood, landslide, and avalanche effect structural members, the crash of vehicles to the highway and seaway structures. Many analytical, numerical, and experimental studies focused on the behavior of RC structural elements such as columns, beams, and slabs under sudden dynamic impact loads. However, there is no comprehensive study on the behavior of the RC column-beam connections under the effect of sudden dynamic impact loads. For this purpose, an experimental study was performed to investigate the behavior of RC column-beam connections under the effect of low-velocity impact loads. Sixteen RC beam-column connections with a scale of 1/3 were manufactured and tested under impact load using the drop-weight test setup. The concrete compressive strength, shear reinforcement spacing in the beam, and input impact energy applied to test specimens were taken as experimental variables. The time histories of impact load acting on test specimens, accelerations, and displacements measured from the test specimens were recorded in experiments. Besides, shear and bending crack widths were measured. The effect of experimental variables on the impact behavior of RC beam-column connections has been determined and interpreted in detail. Besides, a finite element model has been established for verification and comparison of the experimental results by using ABAQUS software. It has been demonstrated that concrete strength, shear reinforcement ratio, and impact energy significantly affect the impact behavior of RC column-beam connections.

        The Investigation of Treatment of Pistachio Processing Industrial Wastewater by Electrochemical Methods in Terms of Chemical Oxygen Demand and Total Phenol Removal

        This work aims to investigate the efficiency of electrocoagulation (EC) of pistachio processing industrial wastewater (PPIW) using the continuous EC process. The tubular reactor made of stainless steel with an internal diameter of 60 mm was used as a cathode electrode. The effect of some parameters was examined on the removal of chemical oxygen demand (COD) and total phenols (TP) removal efficiency. The influences of the initial pH of wastewater (from 4 to 8), flow rate (from 25 to 125 mL/min), current density (from 7 to 21 mA/cm<sup>2</sup>), and supporting electrolyte type (NaCl, NaNO<sub>3</sub>, and Na<sub>2</sub>SO<sub>4</sub>), supporting electrolyte concentration (from 10 to 100 mg/L NaCl) on removal efficiency were investigated to determine the best experimental conditions. The examination of the physico-chemical parameters during the EC treatment showed that the best removal efficiency was obtained under conditions where the flow rate was 25 mL/min (20 min reaction time), the pH value was 5.2, and the current density was 21 mA/cm<sup>2</sup> has set. Under these experimental conditions, COD and TP removal efficiency were found to be 75% and 97%, respectively, while energy consumption was 18.5 kW h/m<sup>3</sup>. The study results show that the EC can be applied to PPIW pre-treatment.

        Reattachment of Rhegmatogenous Retinal Detachment via Fibrin Tissue Adhesive

        Purpose: To manage reattachment of rhegmatogenous retinal detachment using tissue adhesive (fibrin glue) in five patients. Methods: Five eyes of five patients with rhegmatogenous retinal detachment were treated with 23-gauge pars plana vitrectomy and the application of fibrin glue (Tisseel Lyo) into the break under air without laser photocoagulation or cryopexy. Results: The median age of patients was 64 (range, 46–82) years, and the median duration of symptoms was 15 (range, 7–60) days. Three eyes had inferior and two had superior rhegmatogenous retinal detachments. Successful retinal reattachment was achieved in all cases and maintained during the followıng 10 to 24 months. The median best-corrected visual acuity improved from 2 / 100 preoperatively to a Snellen visual acuity of 20 / 50 by 6 months postoperatively. No postoperative complications were detected during the follow-up period. Conclusions: Excellent adhesive effects of fibrin glue with respect to sealing retinal tears with no side effects were seen in our study. With further prospective studies, this technique could be an alternative method to conventional retinopexy.

        Preparation and Characterization of Polyethylene Glycol Functional Hydroxyapatite/Polycaprolactone Electrospun Biomembranes for Bone Tissue Engineering Applications

        Surfaces of previously synthesized Hydroxyapatite particles (HAP) have been modified with polyethylene glycolfunctional silane (PEG-400Si). For the surface modification of HAP, firstly, synthesis of PEG-400Si was performed byurethane reaction of hydroxyl and isocyanate groups. Then, HAP was synthesized by sol-gel method. Afterwards, surfacemodification of HAP was realized with PEG-400Si. SEM, TEM, XRD and FTIR analyses were utilized to characterize themorphology and structural properties of the synthesized and modified particles. Results revealed that the surface of HAP wasmodified successfully and the crystal structure of HAP was not changed after modification. Electrospinning process wasconducted to obtain unmodified and modified HAP incorporated nanofibrous biomembranes and the characteristics andbiological performances of these membranes have been compared to each other. SEM analysis presented that defect-free andround shape nanofibers obtained and the fiber diameter ranged from 230±114 nm to 760±291 nm. In vitro biologicalevaluations revealed that all electrospun nanofibrous biomembranes were nontoxic and the one with PCL/PEG-400Si-HAPexhibited greatest cellular protein expression approximately 1.5 times higher than the PCL biomembrane for 24 h, 48 h and72 h.

        Impact of lymph node ratio on survival in stage IIIC endometrioid endometrial cancer: a Turkish Gynecologic Oncology Group study

        OBJECTIVE: The purpose of this study was to investigate the prognostic value of lymph node ratio (LNR) in women with stage IIIC endometrioid endometrial cancer (EC). METHODS: A multicenter, retrospective department database review was performed to identify patients with stage IIIC pure endometrioid EC at 6 gynecologic oncology centers in Turkey. A total of 207 women were included. LNR, defined as the percentage of positive lymph nodes (LNs) to total nodes recovered, was stratified into 2 groups: LNR1 (≤0.15), and LNR2 (>0.15). Kaplan-Meier method was used to generate survival data. Factors predictive of outcome were analyzed using Cox proportional hazards models. RESULTS: One hundred and one (48.8%) were classified as stage IIIC1 and 106 (51.2%) as stage IIIC2. The median age at diagnosis was 58 (range, 30-82) and the median duration of follow-up was 40 months (range, 1-228 months). There were 167 (80.7%) women with LNR ≤0.15, and 40 (19.3%) women with LNR >0.15. The 5-year progression-free survival (PFS) rates for LNR ≤0.15 and LNR >0.15 were 76.1%, and 58.5%, respectively (p=0.045). An increased LNR was associated with a decrease in 5-year overall survival (OS) from 87.0% for LNR ≤0.15 to 62.3% for LNR >0.15 (p=0.005). LNR >0.15 was found to be an independent prognostic factor for both PFS (hazard ratio [HR]=2.05; 95% confidence interval [CI]=1.07-3.93; p=0.03) and OS (HR=3.35; 95% CI=1.57-7.19; p=0.002). CONCLUSION: LNR seems to be an independent prognostic factor for decreased PFS and OS in stage IIIC pure endometrioid EC.
