        Effects of different primers on indirect orthodontic bonding: Shear bond strength, color change, and enamel roughness

        Objective: We aimed to perform in-vitro evaluation to compare 1) shear bond strength (SBS), adhesive remnant index (ARI), and color change between self-etched and acid-etched primers; 2) the SBS, ARI and color change between direct and indirect bonding; and 3) the enamel roughness (ER) between 12-blade bur and aluminum oxide polisher debonding methods. Methods: Seventy bovine incisors were distributed in seven groups: control (no bonding), direct (DTBX), and 5 indirect bonding (ITBX, IZ350, ISONDHI, ISEP, and ITBXp). Transbond XT Primer was used in the DTBX, ITBX, and ITBXp groups, flow resin Z350 in the IZ350 group, Sondhi in the ISONDHI group, and SEP primer in the ISEP group. SBS, ARI, and ER were evaluated. The adhesive remnant was removed using a low-speed tungsten bur in all groups except the ITBXp, in which an aluminum oxide polisher was used. After coffee staining, color evaluations were performed using a spectrophotometer immediately after staining and prior to bonding. Results: ISONDHI and ISEP showed significantly lower SBS (p < 0.01). DTBX had a greater number of teeth with all the adhesive on the enamel (70%), compared with the indirect bonding groups (0–30%). The ER in the ITBX and ITBXp groups was found to be greater because of both clean-up techniques used. Conclusions: Direct and indirect bonding have similar results and all the primers used show satisfactory adhesion strength. Use of burs and polishers increases the ER, but polishers ensure greater integrity of the initial roughness. Resin tags do not change the color of the teeth.[Korean J Orthod 2018;48(4):245-252]

        Identification of novel mutations in L1CAM gene by a DHPLC-based assay

        X-linked hydrocephalus, MASA syndrome, X-linked complicated Spastic Paraplegia Type I, and X-linked partial agenesis of the corpus callosum are rare diseases mainly affecting male population and broadly referred as L1 syndrome, caused by mutations in the L1CAM gene. In the present study 36 boys and a male fetus whose clinical features were consistent with L1 syndrome were analyzed by dHPLC assay and direct sequencing of L1CAM gene. Sequence analysis of the 14 different aberrant dHPLC elution profiles demonstrated that six of them were associated with already reported polymorphisms, four with previously described causative variants while the remaining four represented novel L1CAM mutations. The dHPLC method proposed identified eight (21 %) causative L1CAM mutations in our patients while direct sequencing failed to detect any variation in patients negative to dHPLC analysis. We conclude that the dHPLC assay represents a fast and efficient method for the screening of L1CAM mutations and that L1 syndrome should be considered in the differential diagnosis of intellectual disability in children, especially when other signs such as hydrocephalus or adducted thumbs are present.

      • 경기침체가 연령관리 정책에 미친 영향 : 오스트리아의 비엔나 종합병원 사례

        피면담자에 따르면 일반적으로 연령관리는 관련된 당사자들이 서로 다른 의제를 갖고 있다는 점에서 매우 복잡한 주제다. 경영진은 효율성 증대에 따른 절감을 원하는 반면에, 직원들은 자율 확대를 희망할 수 있다. 비엔나 종합병원 사례 연구는 어떠한 근로자 집단(이 사례에서는 간호사)을 지원하기 위한 일련의 연령관리 대책 내에서 두 가지 의제를 모두 수용할 수 있다는 것을 보여준다. 이러한 상황은 경제적 조건에 의해서 바뀌지는 않았으며, 오히려 생산적 고령화 프로그램이 더욱 설득력을 얻었다. 광범위한 인구학적 변동으로 고령 인구의 노동시장 참여가 절실해진 오스트리아의 상황을 감안할 때, 이는 중요한 의미를 지닌다. 그러나 고령 근로자의 고용을 유지하기 위한 기제들은 한정되어 있으며, 특히 작업 조건이 신체적으로 고되거나 스트레스 강도가 심한 경우에는 더욱 그러하다. 게다가, 대체로 연령관리 기제들은 비록 장기적으로 그 투자 효과가 확보되기는 하지만 단기적으로는 높은 비용을 초래한다. 비엔나 종합병원의 사례 연구에 의하면, 고령 근로자들이 업무 프로세스에 적극 참여할 수 있는 방안을 모색하기 위해 경영진, 직원, 직원 대표기구가 긴밀하게 협력해야 할 필요가 있다. 고령 인력의 생산적 잠재력이 인정되어야 한다. 성공 요인은 조직이 설계한 개입 장치의 핵심에 개별 근로자를 두는 데에 있다. 여전히 남아 있는 효과적 연령관리 저해 요인은 조기퇴직 규정과 같은 국가적 인센티브로서, 이러한 제도는 근로조건이 아무리 좋더라도 퇴사가 근로자에게 더 유리한 선택이 되게 하기 때문이다.

      • Microbial Electrochemical Process for Bioremediation of Recalcitrant Contaminants in Soil and Water

        Environmental pollution that has exceeded the self-purification capacity of nature requires artificial support, which incurs a lot of economic cost and labor. For the potential solution, a bioremediation process based on ecofriendly, sustainable, and low-cost microbial electrochemical system could be used. In the study, we introduce a microbial electrochemical bioremediation for contaminated soil and water. For bioremediation of contaminated soil by pesticide, a soil microbial fuel cell which can harvest electricity while promoting biodegradation of a selective pesticide called Atrazine was developed, and the direction for scale-up for field application of the proposed system was discussed. For water, a study for concurrent harvesting electrical energy and recovering useful resource from a mineral rich wastewater was developed with linked process of microbial fuel cell and chemical precipitation. The study aims to explain the potential of microbial electrochemical bioremediation for contaminated environment with generation of renewable energy and removal of recalcitrant pollutant, towards a promising sustainable environmental biotechnology.

        Exon deletions of the phenylalanine hydroxylase gene in Italian hyperphenylalaninemics

        A consistent finding of many studies describing the spectrum of mutant phenylalanine hydroxylase (PAH)alleles underlying hyperphenylalaninemia is the impossibility of achieving a 100% mutation ascertainment rate using conventional gene-scanning methods. These methods include denaturing gradient gel electrophoresis (DGGE), denaturing high performance liquid chromatography (DHPLC), and direct sequencing. In recent years, it has been shown that a significant proportion of undetermined alleles consist of large deletions overlapping one or more exons. These deletions have been difficult to detect in compound heterozygotes using gene-scanning methods due to a masking effect of the non-deleted allele. To date, no systematic search has been carried out for such exon deletions in Italian patients with phenylketonuria or mild hyperphenylalaninemia. We used multiplex ligation-dependent probe amplification (MLPA), comparative multiplex dosage analysis (CMDA), and real-time PCR to search for both large deletions and duplications of the phenylalanine hydroxylase gene in Italian hyperphenylalaninemia patients. Four deletions removing different phenylalanine hydroxylase (PAH) gene exons were identified in 12 patients. Two of these deletions involving exons 4-5-6-7-8(systematic name c.353-?_912 + ?del) and exon 6(systematic name c.510-?_706 + ?del) have not been reported previously. In this study, we show that exon deletion of the PAH gene accounts for 1.7% of all mutant PAH alleles in Italian hyperphenylalaninemics.

        Colorimetric and Chlorophyll Fluorescence Assays for Fast Detection and Selection of Transgenic Events of Cotton, Cowpea, Soybean and Common Bean Expressing the Atahas Gene

        The detection of the presence and expression of transgenes in genetically modified plants is a key step in the process ofselecting promising lines. We adapted two methods developed for detecting tolerance to the herbicide imazapyr for selection oftransgenic lines expressing the mutated acetohydroxyacid synthase enzyme (AHAS) from Arabidopsis thaliana (Atahas gene). Thiswas achieved using transgenic events from cotton, cowpea, soybean and common bean, which have previously been transformed toexpress the mutated Atahas gene. In the first method, a colorimetric assay was developed that detects acetoin, an intermediate in thebiosynthetic pathway of branched chain amino acids, which is accumulated in the presence of cyclopropanedicarboxylic acid (CPCA),an inhibitor of ketoacid reductoisomerase (KARI). In the presence of the herbicide, it was possible to distinguish non-transgenic fromtransgenic plants. Qualitative analysis of acetoin formed during the AHAS inhibition allowed to indirectly determine the Atahastransgene expression. The second method measured the kinetics of chlorophyll fluorescence emission. Leaf discs pre-treated withimazapyr for 24 hours were evaluated using the modulated fluorimeter for maximum quantum efficiency of Photosystem II (PSII)(Fv/Fm) and relative electron transport rate (ETR). Results showed that almost all species analyzed presented a marked decrease inFv/Fm after treatment with imazapyr. In addition, the ETR was significantly reduced in transgenic plants treated with the herbicide. Collectively, our results showed that it is possible to identify transgenic plants expressing Atahas gene and infer their levels of toleranceto imazapyr at a very early stage after transformation.

        Transanal Irrigation for Neurogenic Bowel Dysfunction in Multiple Sclerosis: A Retrospective Study

        Background/AimsSixty-eight percent of multiple sclerosis (MS) patients suffer from neurogenic bowel dysfunction (NBD). Transanal irrigation (TAI) is part of the therapeutic strategy. This retrospective study aims to assess the efficacy of TAI in MS population. MethodsTwenty-eight MS patients who underwent TAI after a learning period were included. We collected several demographic data: MS disease characteristics, treatments, urinary and bowel dysfunction characteristics, urodynamic parameters, results of the NBD score, the Urinary Symptom Profile (USP) score, and the Patient Global Impression of Severity score, completed by patients before the learning and during the follow-up consultation. We defined 4 specific groups depending on the NBD score severity: very minor, minor, moderate, and severe. ResultsMean follow-up was 124 days, 85.0% were initially constipated and 36% had fecal incontinence. After TAI, improvement of NBD score was higher in initial Moderate NBD score group with 75.0% of patients decreasing their NBD score into lower severity categories. Few modifications were observed for baseline Very minor and Severe NBD score groups with 60.0% and 87.5% of patients staying in the same category. Statistical improvement of USP voiding dysfunction score was observed (95% CI, –6.13-–1.19; P = 0.005) without improvement of overactive bladder USP sub-score. ConclusionsTAI is effective in NBD, especially in MS patients with initial Moderate NBD score. Improvement of voiding dysfunction following TAI confirms the pelvic organ cross-talk and the need to systematically consider and treat bowel dysfunction in MS to also improve urinary symptoms.
