Molecular analysis of quasispecies of <i>Kyuri green mottle mosaic virus</i>

<P><B>Abstract</B></P><P>The population diversity of progeny viruses of <I>Kyuri green mottle mosaic virus</I> (KGMMV) in consecutive serial passages in two systemic hosts, zucchini squash and cucumber plants, established from genetically identical viral RNA, was examined in this study. An initial plant was inoculated with in vitro transcripts from a full-length cDNA clone of KGMMV. The initial viral population (passage 0) was transferred five times in parallel populations in the same hosts species for analysis of progenies of KGMMV. The percentage of mutations of progeny viruses fluctuated slightly, as expected, during the serial passage, and these results did not correlated with the mutation frequency calculated as the total number of mutation observed in all the clones sequenced for a given viral population were divided by the total number of bases sequenced for the population. The mutation frequencies represented similar distributions over the course of serial passages in the two systemic host plants. Seventeen unique mutations were detected from a total of 40 clones (19,120 bases) sequenced, indicating a relatively small overall mutation rate of 17 nucleotide substitutions. The majority of observed mutations in the viral populations consisted of substitutions: 61.60 and 64.08% of the mutations in cucumber and zucchini populations, respectively. The types of transitions and silent mutations indicated that progenies of KGMMV reached stabilized selection during the host passages and maintained viral quasispecies in systemic hosts.</P>

The main objective of this study is to investigate an impact of organizational culture on supply chain risk and strategic sourcing. It also examines the relationship between organizational culture and strategic sourcing. By collecting 159 survey responses from supply, executives and managers of U.S. manufacturing firms, it provides empirical evidence that organizational culture and strategic sourcing mitigate supply chain risks. Organizational culture also makes a positive impact on the implementation of strategic sourcing. This study emphasizes the importance of cultural impacts to supply executives and managers in supply chain risk management.

<P>Summary</P><P>The bacterium <I>Burkholderia glumae</I> causes rice grain rot by producing toxoflavin, whose expression is regulated by quorum sensing (QS). We report a major deviation from the current paradigm for the regulation of bacterial polar flagellum genes. The <I>N</I>-octanoyl homoserine lactone (C8-HSL)-deficient mutant of <I>B. glumae</I> is aflagellate and has lost the ability to swim and swarm at 37°C. Mutagenesis of the bacterium with the mini-Tn<I>5rescue</I> identified an IclR-type transcriptional regulator, called QsmR, which is important for flagellum formation. TofR, which is a cognate C8-HSL receptor, activated <I>qsmR</I> expression by binding directly to the <I>qsmR</I> promoter region. From the flagellum gene cluster, we identified <I>flhDC</I> homologues that are directly activated by QsmR. FlhDC in turn activates the expression of genes involved in flagellum biosynthesis, motor functions and chemotaxis in <I>B. glumae</I>. Non-motile <I>qsmR</I>, <I>fliA</I> and <I>flhDC</I> mutants produced toxoflavin, but lost pathogenicity for rice. The unexpected discovery of FlhDC in a polarly flagellate bacterium suggests that exceptions to the typical regulatory mechanisms of flagellum genes exist in Gram-negative bacteria. The finding that functional flagella play critical roles in the pathogenicity of <I>B. glumae</I> suggests that either QS or flagellum formation constitutes a good target for the control of rice grain rot.</P>

<P>A new stereoisomer of a tetrahydrofuranoid lignan, acerifuranoid A (1), and two new oleanane-type triterpenoids, aceriphyllic acids J and K (2 and 3), were isolated from the roots of Aceriphyllum rossii. Their structures were elucidated on the basis of spectroscopic analyses and chemical evidence. PTP1B inhibitory activity of kaurane diterpenes isolated from <i>Siegesbeckia glabrescens</i>

<P>Protein tyrosine phosphatase 1B (PTP1B) is considered as a therapeutic target for the treatment of diabetes and obesity. In our preliminary screening study, a MeOH extract of the aerial part of <I>Siegesbeckia glabrescens</I> was found to inhibit PTP1B activity at 30 μg/mL. Bioassay‐guided fractionation led to the isolation of two active diterpenes, <I>ent</I>-16&bgr;H,17-isobutyryloxy-kauran-19-oic acid (<B>1</B>) and <I>ent</I>-16&bgr;H,17-acetoxy-18-isobutyryloxy-kauran-19-oic acid (<B>2</B>), along with <I>ent</I>-16&bgr;H,17-hydroxy-kauran-19-oic acid (<B>3</B>). Compounds <B>1</B> and <B>2</B> inhibited the PTP1B activity with IC<SUB>50</SUB> values of 8.7 ± 0.9 and 30.6 ± 2.1 μM, respectively. Kinetic studies suggest that both <B>1</B> and <B>2</B> are non-competitive inhibitors of PTP1B. However, compound <B>3</B> substituted with a hydroxyl group at C-17 in kaurane-type showed no inhibitory effects towards PTP1B.</P>
Studies on urease inhibitors from pyroligneous acids

The objectives of this study were to measure the inhibitory effect of pyroligneous acid on urease activity and isolation of urease inhibitor from pyroligneous acid. The fractions of pyroligneous acid were tested for their urease inhibitory activities ny using jack bean urease, bacillus paseurii urease and soil urease in an enzyme assay. The peak 1 was a more effective inhibitor of jack bean urease, bacillus paseruii urease and soil urease than pyroligneous acid, F1, F1b and peak 2. In addition, the inhibitory effect was investigated on urease activity in soil by fractions of pyroligneous acid. When the fractions of pyroligneous acid were applied at rate of 15,150 and 180 ㎍ g^-1 soil, all of the treatments retarded urea hydrolysis in soil and peak1 was the most effective for retardation of urea hydrolysis in soil. In flooded soil, highest inhibition of urea hydrolysis is obtained with peak 1 followed by peak 2. The concentrations of pyroligneous acid, F1, F1b, peak1 and peak 2 required for 50% inhibition under the conditions of the experiment were 19.8, 13.6, 5.1, 1.53 and 1.62 g/㎖, respectively. Peak 1 was the most powerful urease inhibitor among fractions of the pyroligneous acid. Peak 2, F1b, F1 and pyroligneous acid followed, in terms of decreasing inhibition ability. The kinetics of the inhibition of jack bean urease in the presence of peak 1 was studied at pH 7.0. Peak 1 seemed to inhibit the jack bean urease activity via a noncompetitive pattern when urea was used as a substrate.

Objective : Pituitary adenomas frequently extend into the suprasellar space. After a suprasellar tumor is removed, the superiorly extended arachnoid becomes redundant and sinks down into the intrasellar space which often hiders visualization and accessibility to the hidden space behind the evaginated arachnoid. We introduced arachnoid remodeling by clipping technique, and evaluated its usefulness and safety during TSS. Methods : Total 223 patients who underwent arachnoid remodeling with our new clipping technique were included. Redundant arachnoid was clipped along the dural edge with multiple 2.6-mm titanium clips until the redundant arachnoid membrane no longer blocked the surgical route. To check for possible deterioration of hormonal function by this technique, we assessed anterior pituitary function of 166 patients who underwent arachnoid remodeling by clipping and compared this with those of other 429 control patients. Results : Our technique greatly enhanced the accessibility and visualization of intrasellar and parasellar spaces, both of which are generally hindered by redundant arachnoid during transsphenoidal surgery (TSS). We found no difference in anterior pituitary function between a clip-assisted arachnoid remodeling group and the control group, implying that this technique does not result in hypopituitarism. Conclusion : During TSS for pituitary adenomas with suprasellar extension, arachnoid remodeling by clipping technique is very useful and convenient for the management of the redundant arachnoid membrane to enhance visualization and surgical accessibility.
