A total of 113 novel EST-derived simple sequence repeat (EST-SSR) markers were developed in the Pacific oyster (Crassotrea gigas). Polymorphisms of these markers were evaluated in a wild population of 30 individuals. The number of alleles per locus ranged from 2 to 27 with an average of 6.3, and the observed and expected heterozygosities varied from 0 to 0.9667 and from 0.0333 to 0.9701, respectively. Mendelian segregations were tested for 24 of the markers that were polymorphic in one family produced by single-pair mating. Null alleles were discovered at four loci. Nine tests of segregation ratios revealed significant departures from expected Mendelian ratios. As a useful addition to the collection of the microsatellites that are now available for C. gigas, these EST-SSR markers will help the advance in investigation of QTL mapping and genetic diversity in this species.

A new fluorescent probe L fusing the rhodamine B scaffold with 2H-benzo[b][1,4]oxazin-3(4H)-one moiety has been developed and applied as an acidic pH sensor. The ultraviolet (UV) and fluorescence spectra of the probe at different pH values were investigated in B?R buffer/MeOH (v: v = 3: 7). This probe showed increase of both the UV?Vis absorption and fluorescence emission from alkaline/neutral to acidic condition, reaching a strongest intensity at pH 2.13, but the intensities decreased when the pH decreased to 1.83. The color variation along the pH changes proved that the probe L can be used as a low-cost optical sensor for the naked eye detection of proton in the present buffer.

A new quinoline-based acylhydrazone (1) has been synthesized and applied as a fluorescent probe. Probe 1 exhibits high selectivity and sensitivity to $Cu^{2+}$ with fluorescence "ON-OFF" behavior in HEPES buffered (1‰ DMSO, HEPES 20 mM, pH = 7.4) solution. The on-site generated 1-$Cu^{2+}$ complex displays excellent selectivity to sulfide ions with fluorescence "OFF-ON" performance through copper displacement approach.

Shell-and-tube heat exchanger (STHX) is widely used by virtue of its simple structure and high reliability, especially in a space-constrained surface ship. For the STHX of the surface ship, roll, pitch and other motion of the ship will affect the heat transfer performance, resistance characteristics and structural strength of the heat exchanger. Therefore, it is urgent to carry out numerical simulation research on three-dimensional thermal hydraulic characteristics of surface ship STHX under the marine conditions. In this paper, the numerical simulation of marine shell and tube heat exchanger of surface ship was carried out using the porous media model. Firstly, the mathematical physical model and numerical method are validated based on the experimental data of a marine engine cooling water shell and tube heat exchanger. The simulation results are in good agreement with the experimental results. The prediction errors of pressure drop and heat transfer are less than 10% and 1% respectively. The effect of marine conditions on the heat transfer characteristics of the heat exchanger is investigated by introducing the additional force model of marine condition to evaluate the effect of different motion parameters on the heat transfer performance of the heat exchanger. This study could provide a reference for the optimization of marine heat exchanger design.

A Two-DNA Methylation Signature to Improve Prognosis Prediction of Clear Cell Renal Cell Carcinoma

Purpose: Effective biomarkers and models are needed to improve the prognostic prospects of clear cell renal cell carcinoma(ccRCC). The purpose of this work was to identify DNA methylation biomarkers and to evaluate the utility of DNA methylationanalysis for ccRCC prognosis. Materials and Methods: An overview of genome-wide methylation of ccRCC tissues derived from The Cancer Genome Atlas(TCGA) database was download for analysis. DNA methylation signatures were identified using Cox regression methods. The potentialclinical significance of methylation biomarkers acting as a novel prognostic markers was analyzed using the Kaplan–Meiermethod and receiver operating characteristic (ROC) curves. Results: This study analyzed data for 215 patients with information on 23171 DNA methylation sites and identified a two-DNAmethylation signature (cg18034859, cg24199834) with the help of a step-wise multivariable Cox regression model. The area underthe curve of ROCs for the two-DNA methylation signature was 0.819. The study samples were stratified into low- and high-risk classificationsbased on an optimal threshold, and the two groups showed markedly different survival rates. Moreover, the two-DNAmethylation marker was suitable for patients of varying ages, sex, stages (I and IV), and histologic grade (G2). Conclusion: The two-DNA methylation signature was deemed to be a potential novel prognostic biomarker of use in increasingthe accuracy of predicting overall survival of ccRCC patients.
