        Accuracy of cardiac output measurements during off-pump coronary artery bypass grafting: according to the vessel anastomosis sites

        Background: During beating heart surgery, the accuracy of cardiac output (CO) measurement techniques may be influenced by several factors. This study was conducted to analyze the clinical agreement among stat CO mode (SCO), continuous CO mode (CCO), arterial pressure waveform-based CO estimation (APCO), and transesophageal Doppler ultrasound technique (UCCO) according to the vessel anastomosis sites. Methods: This study was prospectively performed in 25 patients who would be undergoing elective OPCAB. Hemodynamic variables were recorded at the following time points: during left anterior descending (LAD) anastomosis at 1 min and 5 min; during obtuse marginal (OM) anastomosis at 1 min and 5 min: and during right coronary artery (RCA) anastomosis at 1 min and 5 min. The variables measured including the SCO, CCO, APCO, and UCCO. Results: CO measurement techniques showed different correlations according to vessel anastomosis site. However, the percent error observed was higher than the value of 30% postulated by the criteria of Critchley and Critchley during all study periods for all CO measurement techniques. Conclusions: In the beating heart procedure, SCO, CCO and APCO showed different correlations according to the vessel anastomosis sites and did not agree with UCCO. CO values from the various measurement techniques should be interpreted with caution during OPCAB.         Preparation and Properties of Polyurethane Modified with Polyorganosiloxane Sealant

        α, ω-Trimethoxysilyl polyurethane modified with polyorganosiloxane (TUMPS) sealant was prepared by adding additives, such as plasticizer, crosslinking agent, and viscosity increasing agent, to α, ω-trimethoxysilyl polyurethane modified with polyorganosiloxane hybrid elastomer at room temperature under nitrogen atmosphere. The TUMPS hybrid elastomer was crosslinked by the sol-gel reaction of methoxy group of TUMPS with hydroxyl group in the building stone and moisture of the air. It was found that TUMPS sealant exhibited more stable rheological and sealant properties than those of TUMPS hybrid elastomer. The shrinkage rate of TUMPS sealant was 5.82-6.0%, and skin over time was about 40 min. Alkaline resistance showed good characteristics for 15 days, slump was about 4.0-4.3㎜, and oil content, as the diameter soaked oil from wetted filter paper was 1-2㎜ after 7 days.

        Conditioned Media of ASC-17D Sertoli Cells Induce G1-Growth Arrest of DU145 Human Prostate Cancer Cells

        We studied the effects of ASC-17D rat Sertoli cell-conditioned media (rSCCM) on the proliferation of the DU145 prostate cancer cells. rSCCM was prepared from ASC-17D cells cultured in DMEM/F-12 serum-free media at a nonpermissive temperature of 40℃, which is the condition for the high expression of clusterin. We found that rSCCM could inhibit the proliferation of DU145 cells by arresting the cell cycle in the G1 phase in a dose-dependent manner. This growth arresting activity was abolished by boiling rSCCM for 5 min. The G1 growth-inhibiting activity of rSCCM was also detected in other prostate-originated cancer cells examined (ie., LNCaP and PC-3) but not in other cells (ASC-17D, HepG2, SK-N-SH, and NIH3T3). Western blot analysis of partially purified growth inhibiting fractions with the clusterin antibody showed that the cytostatic factor in rSCCM was not clusterin. This cytostatic factor was semipurified by DEAE-Sepharose, ammonium sulfate precipitation, and Phenyl-Sepharose column chromatography, and was estimated to have a molecular weight of 88 kDa by Sephacryl S-300 gel filtration.

      • Structural and biochemical basis for the inhibition of cell death by APIP, a methionine salvage enzyme

        <P>APIP, Apaf-1 interacting protein, has been known to inhibit two main types of programmed cell death, apoptosis and pyroptosis, and was recently found to be associated with cancers and inflammatory diseases. Distinct from its inhibitory role in cell death, APIP was also shown to act as a 5-methylthioribulose-1-phosphate dehydratase, or MtnB, in the methionine salvage pathway. Here we report the structural and enzymatic characterization of human APIP as an MtnB enzyme with a <I>K</I><SUB><I>m</I></SUB> of 9.32 μM and a <I>V</I><SUB><I>max</I></SUB> of 1.39 μmol min<SUP>−1</SUP> mg<SUP>−1</SUP>. The crystal structure was determined at 2.0-Å resolution, revealing an overall fold similar to members of the zinc-dependent class II aldolase family. APIP/MtnB exists as a tetramer in solution and exhibits an assembly with <I>C4</I> symmetry in the crystal lattice. The pocket-shaped active site is located at the end of a long cleft between two adjacent subunits. We propose an enzymatic reaction mechanism involving Glu139* as a catalytic acid/base, as supported by enzymatic assay, substrate-docking study, and sequence conservation analysis. We explored the relationship between two distinct functions of APIP/MtnB, cell death inhibition, and methionine salvage, by measuring the ability of enzymatic mutants to inhibit cell death, and determined that APIP/MtnB functions as a cell death inhibitor independently of its MtnB enzyme activity for apoptosis induced by either hypoxia or etoposide, but dependently for caspase-1-induced pyroptosis. Our results establish the structural and biochemical groundwork for future mechanistic studies of the role of APIP/MtnB in modulating cell death and inflammation and in the development of related diseases.</P>

        Microfluidic generation of Prussian blue-laden magnetic micro-adsorbents for cesium removal

        <P><B>Abstract</B></P> <P>Here, we designed and synthesized a recoverable multifunctional adsorbent using a microfluidic reaction system and evaluated the removal performance of the smart adsorbent toward radioactive cesium as a model sample. Prussian blue-laden magnetic micro-adsorbents (PB-MNPs-MAs) with uniform morphology and monodispersity were generated via two-step sequential procedures using a glass capillary microfluidic system, followed by chemical co-precipitation with a high production rate. The cesium removal efficacy of the PB-MNPs-MAs was analyzed based on Langmuir and Freundlich isotherms by controlling adsorption parameters such as adsorbent size, initial cesium concentration, and contact time. The adsorption isotherm of the PB-MNPs-MAs was better fitted to the Langmuir model with a maximum cesium adsorption capacity of 58.73 mg g<SUP>−1</SUP>, which was 40% higher than that of macro-adsorbents in a dynamic magnetic field. This result can be attributed to their large specific area, which increased the kinetic rate of cesium adsorption and achieved saturation within 20 min. Additionally, the PB-MNPs-MAs were recovered from wastewater within 5 s under a static magnetic field, indicating their great potential for magnetic actuation.         Nutritional components of Ojuk leaf tea and Ojuk shoot tea prepared from the leaves and shoots of black bamboo (Phyllostachys nigra Munro) by tea manufacturing process were evaluated. In addition, the extraction yield of water soluble components from these teas in the general tea brewing condition (water extraction in 80oC for 10 min.) and the contents of polyphenol and flavonoid were compared with not only the dried raw materials, but also green tea and mate tea. Finally, offline and online scavenging activities against 2,2`-azino-bis(3-ethylbenzothiazoline- 6-sulphonic acid) (ABTS) and di(phenyl)-(2,4,6 trinitrophenyl)iminoazanium free radicals were investigated to evaluate the antioxidant activity and explore the components showing ABTS free radical scavenging activity from tea infusion. These results demonstrated that these teas from black bamboo contain various nutritional components and can be used as traditional tea beneficial to human health.

        The home meal replacement (HMR) market has attracted significant attention due to COVID-19 pandemic and products that utilize the combination of different processed ingredients are now being developed. In this study, Broughton’s ribbed ark Scapharca broughtonii was soaked in seasoned soy sauce with the incorporation of mustard leaf Brassica juncea (M-BRA), which is known to have a unique texture as well as excellent functional properties such as antioxidation, to develop a regional specialty product. The optimal conditions for manufacturing M-BRA from the seasoned soy sauce (the sauce to be added [X<sub>1</sub>] and the soaking time [X<sub>2</sub>]), were examined using response surface methodology (RSM) to analyze the significance of the salinity (Y<sub>1</sub>), amino-N content (Y<sub>2</sub>), and overall acceptance (Y<sub>3</sub>). The coefficient of determination (R<sup>2</sup>) between X<sub>1</sub>-X<sub>2</sub> and Y<sub>1</sub>-Y<sub>3</sub> were close to 1, thereby confirming the suitability of the RSM model. The optimal conditions were seasoned soy sauce addition of 140% and soaking time of 156 min. The M-BRA manufactured under these conditions exhibited superior overall acceptance compared to seasoned commercial soy sauce, which was used as a control. We expect that the market for M-BRA manufactured by combining marine and agricultural materials will expand owing to superior overall acceptance compared with commercial products.

        비린내와 조직감이 개선된 치즈 토핑 반건조 조미 피조개(Scapharca broughtonii)의 개발 및 특성

        Methods for the development of home meal replacement seafood tailored to consumer needs for the advanced use of Broughton’s ribbed ark Scapharca broughtonii (BRA) in Korea are required. In this study, we developed a cheesetopped, semi-dried, and seasoned Broughton’s ribbed ark (S-BRA) tailored for the younger generation with an improved texture and fish odor. The optimization of conditions to improve the texture and fish odor was performed using RSM. The design of the model was appropriate because there was no significant difference (P>0.05) between the predicted and actual values of moisture content, hardness, and overall acceptance, and the optimal preparation conditions were a vinegar content of 2.68%, a soaking time of 62 min, a drying temperature of 60℃, and a time of 162 min. The SBRA manufactured under these optimal conditions exhibited a lower odor intensity compared to the unsoaked and undried control, suggesting that the fish odor of S-BRA has been improved. The moisture content related to the texture of the S-BRA was lower than that of the control, and the hardness was higher. Therefore, the S-BRA developed in this study will appeal to people of all ages, especially the younger generation; their consumption is expected to increase.

        Analysis of Lignans in Acanthopanax sessiliflorus Fruits and Their Fermented Wine by HPLC

        High performance liquid chromatography (HPLC) was used for the determination of lignans, eleutherosides B and E, in Acanthopanax sessiliflorus fruits and their fermented wine. The lignans were quantified by a reversed-phase system using a gradient of H2O and acetonitrile as a mobile phase within 20 min. The analysis was successfully carried out within 20 min. The contents of eleutherosides Band E as main active principles of Acanthopanax species were measured in A. sessiliflorus fruits (1.15 and 8.49 μg/mg, respectively), their fermented wine (0.45 and 1.33 μg/mg, respectively) and wine residues (no detection).
