        A binary matrix for improved detection of phosphopeptides in matrix-assisted laser desorption/ionization mass spectrometry

        <P>Application of matrix-assisted laser-desorption/ionization mass spectrometry (MALDI MS) to analysis and characterization of phosphopeptides in peptide mixtures may have a limitation, because of the lower ionizing efficiency of phosphopeptides than nonphosphorylated peptides in MALDI MS. In this work, a binary matrix that consists of two conventional matrices of 3-hydroxypicolinic acid (3-HPA) and α-cyano-4-hydroxycinnamic acid (CCA) was tested for phosphopeptide analysis. 3-HPA and CCA were found to be hot matrices, and 3-HPA not as good as CCA and 2,5-dihydroxybenzoic acid (DHB) for peptide analysis. However, the presence of 3-HPA in the CCA solution with a volume ratio of 1:1 could significantly enhance ion signals for phosphopeptides in both positive-ion and negative-ion detection modes compared with the use of pure CCA or DHB, the most common phosphopeptide matrices. Higher signal intensities of phosphopeptides could be obtained with lower laser power using the binary matrix. Neutral loss of the phosphate group (−80 Da) and phosphoric acid (−98 Da) from the phosphorylated-residue-containing peptide ions with the binary matrix was decreased compared with CCA alone. In addition, since the crystal shape prepared with the binary matrix was more homogeneous than that prepared with DHB, searching for ‘sweet’ spots can be avoided. The sensitivity to detect singly or doubly phosphorylated peptides in peptide mixtures was higher than that obtained with pure CCA and as good as that obtained using DHB. We also used the binary matrix to detect the in-solution tryptic digest of the crude casein extracted from commercially available low fat milk sample, and found six phosphopeptides to match the digestion products of casein, based on mass-to-charge values and LIFT TOF-TOF spectra.         Interaction of internal forces of interior beam-column joints of reinforced concrete frames under seismic action

        This paper presents detailed analysis of the internal forces of interior beam-column joints of reinforced concrete (RC) frames under seismic action, identifies critical joint sections, proposes consistent definitions of average joint shear stress and average joint shear strain, derives formulas for calculating average joint shear and joint torque, and reports simplified analysis of the effects of joint shear and torque on the flexural strengths of critical joint sections. Numerical results of internal joint forces and flexural strengthsof critical joint sections are presented for a pair of concentric and eccentric interior connections extracted from a seismically designed RC frame. The results indicate that effects of joint shear and torque may reduce the column-to-beam flexural strength ratios to below unity and lead to “joint-yielding mechanism” for seismically designed interior connections. The information presented in this paper aims to provide some new insight into the seismic behavior of interior beam-column joints and form a preliminary basis for analyzing the complicated interaction of internal joint forces.

        Hepatic Sinusoidal Obstruction Syndrome Caused by Herbal Medicine: CT and MRI Features

        Objective: To describe the CT and MRI features of hepatic sinusoidal obstruction syndrome (HSOS) caused by herbal medicine Gynura segetum. Materials and Methods: The CT and MRI features of 16 consecutive Gynura segetum induced HSOS cases (12 men, 4 women) were analyzed. Eight patients had CT; three patients had MRI, and the remaining five patients had both CT and MRI examinations. Based on their clinical presentations and outcomes, the patients were classified into three categories:mild, moderate, and severe. The severity of the disease was also evaluated radiologically based on the abnormal hepatic patchy enhancement in post-contrast CT or MRI images. Results: Ascites, patchy liver enhancement, and main right hepatic vein narrowing or occlusion were present in all 16 cases. Hepatomegaly and gallbladder wall thickening were present in 14 cases (87.5%, 14/16). Periportal high intensity on T2-weighted images was present in 6 cases (75%, 6/8). Normal liver parenchymal enhancement surrounding the mainhepatic vein forming a clover-like sign was observed in 4 cases (25%, 4/16). The extent of patchy liver enhancement was statistically associated with clinical severity classification (kappa = 0.565). Conclusion: Ascites, patchy liver enhancement, and the main hepatic veins narrowing were the most frequent signs of herbal medicine induced HSOS. The grade of abnormal patchy liver enhancement was associated with the clinical severity.

        Exploration of temperature effect on videogrammetric technique for displacement monitoring

        There has been a sustained interest towards the non-contact structural displacement measurement by means of videogrammetric technique. On the way forward, one of the major concerns is the spurious image drift induced by temperature variation. This study therefore carries out an investigation into the temperature effect of videogrammetric technique, focusing on the exploration of the mechanism behind the temperature effect and the elimination of the temperature-caused measurement error. 2D videogrammetric measurement tests under monotonic or cyclic temperature variation are first performed. Features of measurement error and the casual relationship between temperature variation and measurement error are then studied. The variation of the temperature of digital camera is identified as the main cause of measurement error. An excellent linear relationship between them is revealed. After that, camera parameters are extracted from the mapping between world coordinates and pixels coordinates of the calibration targets. The coordinates of principle point and focal lengths show variations well correlated with temperature variation. The measurement error is thought to be an outcome mainly attributed to the variation of the coordinates of principle point. An approach for eliminating temperature-caused measurement error is finally proposed. Correlation models between camera parameters and temperature are formulated. Thereby, camera parameters under different temperature conditions can be predicted and the camera projective matrix can be updated accordingly. By reconstructing the world coordinates with the updated camera projective matrix, the temperature-caused measurement error is eliminated. A satisfactory performance has been achieved by the proposed approach in eliminating the temperature-caused measurement error.

        Systematic Review and Meta-Analysis of Antibiotic-Impregnated Shunt Catheters on Anti-Infective Effect of Hydrocephalus Shunt

        Objective : Shunt infection is a common complication while treating hydrocephalus. The antibiotic-impregnated shunt catheter (AISC) was designed to reduce shunt infection rate. A meta-analysis was conducted to study the effectiveness of AISCs in reduction of shunt infection in terms of age, follow-up time and high-risk patient population. Methods : This study reviewed literature from three databases including PubMed, EMBASE, and Cochrane Library (from 2000 to March 2019). Clinical studies from controlled trials for shunt operation were included in this analysis. A subgroup analysis was performed based on the patient's age, follow-up time and high-risk population. The fixed effect in RevMan 5.3 software (Cochrane Collaboration) was used for this meta-analysis. Results : This study included 19 controlled clinical trials including 10105 operations. The analysis demonstrated that AISC could reduce the infection rate in shunt surgery compared to standard shunt catheter (non-AISC) from 8.13% to 4.09% (odds ratio [OR], 0.48; 95% confidence interval [CI], 0.40-0.58; p=0.01; I2=46%). Subgroup analysis of different age groups showed that AISC had significant antimicrobial effects in all three groups (adult, infant, and adolescent). Follow-up time analysis showed that AISC was effective in preventing early shunt infections (within 6 months after implant). AISC is more effective in high-risk population (OR, 0.24;95% CI, 0.14-0.40; p=0.60; I2=0%) than in general patient population. Conclusion : The results of meta-analysis indicated that AISC is an effective method for reducing shunt infection. We recommend that AISC should be considered for use in infants and high-risk groups. For adult patients, the choice for AISC could be determined based on the treatment cost.

Numerical results of shear, bending moment and torque in joints induced by seismic action are presented for a pair of concentric and eccentric exterior connections extracted from a seismically designed RC frame, and two sections located at the levels of beam bottom and top reinforcement, respectively, are identified as the critical joint sections for evaluating seismic joint behavior. A simplified analysis of the effects of joint shear and torque on the flexural strengths of the critical joint sections is made for the two connections extracted from the frame, and the results indicate that joint shear and torque induced by a strong earthquake may lead to “joint-hinging” mechanism of seismically designed RC frames.

        Injuries Analysis of Rear Row Occupants Exposed to Vehicle’s Frontal Oblique Collision

        This paper discusses the rear left-hand occupant’s injury during frontal oblique collision with PRESYS software. Based on the simulations among different frontal oblique angles (10 ~ 50 °) and occupant constraint system models, the injury of rear left-hand dummy are analyzed at an initial collision speed of 50 km/h. It is found that the impact on the occupant’s dynamic responses (acceleration and shear forces) by the seat belt is significant and depends upon the oblique angle. With the given vehicle and collision speed, it is observed that the seat belt can effectively reduce the occupant’s head acceleration and the neck longitudinal (Fx) shear force if the collision angle is between 10 ° and 20 °. However, the seat belt increases the occupant’s head resultant acceleration when the collision angle is between 35 ° and 40 °. In addition, when the collision angle is between 10 ° and 50 °, the seat belt can also effectively reduce the occupant\'s chest acceleration. At most collision angles, the seat belt can effectively reduce the overall damage of rear row left occupant in frontal oblique collision, but the WIC (Weighted Injury Criterion) value of the rear row dummy with seat belt is still very large at the collision angle below 30 °. Therefore, the restraint system of the rear row occupants needs to be improved.
