        DISPERSION OF CARBON NANOFIBERS IN AQUEOUS SOLUTION

        Carbon nano¯bers (CNFs) were puri¯ed by the method of heat treating, and stable homogeneous suspensions of heat-treated CNFs were prepared by using methylcellulose (MC) as a surfactant. The heat-treated CNFs were characterized by di®erential thermal analysis (DTA) and thermogravimetry analysis (TGA), and the sonication-driven dispersion of CNFs in aqueous solution was investigated by measuring ultraviolet absorbency (UV absorbency), zeta potential, adsorption isotherm of MC on CNFs and surface tension of CNF suspensions. In these CNF suspensions, the zeta potentials of CNFs progressively increase with increasing MC concentration, and change from ?15.4?0 mV. The surface tension of CNF suspensions decrease from 38.87mN/m to 36.54mN/m. The adsorption isotherm of MC on CNFs shows an adsorption platform (namely \L" style), and the adsorption isotherm reaches the saturation plateau at MC concentration of about 0.4 g/L. All results show that the optimum concentration ratio of MC to CNFs is 2:1 for dispersion.

        Using weighted gene co-expression network analysis (WGCNA) to identify the hub genes related to hypoxic adaptation in yak (Bos grunniens)

        Background As a mammal living at the highest altitude in the world, the yak has strong adaptability to the harsh natural environment (such as low temperature, scarce food, especially low oxygen) of Qinghai-Tibet Plateau (QTP) after a long process of natural selection. Objective Here, we used Weighted Correlation Network Analysis (WGCNA), a systematic biology method, to identify hypoxic adaptation-related modules and hub genes. The research of the adaptability of yak against hypoxia is of great signifcance to identify the genetic characteristics and yak breeding. Methods Based on the transcriptome sequencing data (PRJNA362606), the R package DESeq2 and WGCNA were conducted to analyze diferentially expressed genes (DEGs) and construct the gene co-expression network. The module hub genes were identifed and characterized by the correlation of gene and trait, module membership (kME). In addition, GO and KEGG enrichment analyses were used to explore the functions of hub genes. Results Our results revealed that 1098, 1429, and 1645 DEGs were identifed in muscle, spleen, and lung, respectively. Besides, a total of 13 gene co-expression modules were detected, of which two hypoxic adaptation-related modules (saddlebrown and turquoise) were found. We identifed 39 and 150 hub genes in these two modules. Functional enrichment analyses showed that 12 GO terms and 18 KEGG pathways were enriched in the saddlebrown module while 85 GO terms and 22 KEGG pathways were enriched in the turquoise module. The signifcant pathways related to hypoxia adaptation include FoxO signaling pathway, Thermogenesis pathway, and Retrograde endocannabinoid signaling pathway, etc. Conclusions In this study, we obtained two hypoxia-related specifc modules and identifed hub genes based on the connectivity by constructing a weighted gene co-expression network. Function enrichment analysis of two modules revealed mitochondrion is the most important organelle for hypoxia adaptation. Moreover, the insulin-related pathways and thermogenic-related pathways played a major role. The results of this study provide theoretical guidance for further understanding the molecular mechanism of yak adaptation to hypoxia.

      • Study on Corner Reflectors Identification in Highway Deformation Monitoring

        CRInSAR is a newly developed technique to monitor ground deformation. In CRInSAR algorithm, the identification of Corner Reflectors in SAR images is necessary. Due to the uncertainty of traditional identification method, a new method based on the intensity and correlation coefficient of each CR candidates in SAR images is presented. The method has been successfully used to determine the locations of 11 CRs installed along a highway in six SAR images over the study area. The results show that the identification accuracy of the new method is about 1 pixel. It is effective and reliable especially in the area with lots of lightspots around the CR points. The method proposed can play important role in the highway deformation monitoring within CRInSAR algorithm.

      • Comparison of phenolics compounds between healthy and viroid-infected grapes in Korea

        Grapes are widely consumed fruits in Korea. It has been used for the products such as raisins, wine, juice, vinegar and sweet spreads. However, grapes are subject to viroid infection, causing malformed berries and reducing fruit quality and yield. In this study, effect of Grapevine yellow speckle viroid 1 (GYSVd1) on the phenolics in grape skin of three different cultivars including ‘Campbell Early’, ‘Kyoho’, and ‘Shine muscat’ were investigated by liquid chromatography method. Nineteen phenolic compounds in grape skin of three cultivars were detected using HPLC-PDA-MS/MS. Maximum wavelength of anthocyanins were determined at 520 nm while the others were quantified at 350 nm. The separation was achieved by using a mobile phase containing 10% formic acid in water (A) and 10% formic acid in acetonitrile (B) with a gradient elution (Tmin/B: T0/7; T20/20; T45/30; T60/60) at a flow rate of 0.7 mL/min. As a result, considerable decreases of anthocyanins in ‘Campbell’ and ‘Kyoho’ cultivars were observed in viroid infection group (p<0.05). Additionally, content of flavonols in viroid samples of all three cultivars increased signifacantly (p<0.05) and there were no significant change in phenolic acid content. Because of the viroid infection, flavonols seems to be more synthesized, which could act as the ‘antibody’ of grapes. However, reduction of anthocyanins will lead to low quality of the berries as well as products made from viroid-infected grapes.

        Genome-wide identification and analysis of long noncoding RNAs in longissimus muscle tissue from Kazakh cattle and Xinjiang brown cattle

        Objective: In recent years, long noncoding RNAs (lncRNAs) have been identified in many species, and some of them have been shown to play important roles in muscle development and myogenesis. However, the differences in lncRNAs between Kazakh cattle and Xinjiang brown cattle remain undefined; therefore, we aimed to confirm whether lncRNAs are differentially expressed in the longissimus dorsi between these two types of cattle and whether differentially expressed lncRNAs regulate muscle differentiation. Methods: We used RNA-seq technology to identify lncRNAs in longissimus muscles from these cattle. The expression of lncRNAs were analyzed using StringTie (1.3.1) in terms of the fragments per kilobase of transcript per million mapped reads values of the encoding genes. The differential expression of the transcripts in the two samples were analyzed using the DESeq R software package. The resulting false discovery rate was controlled by the Benjamini and Hochberg's approach. KOBAS software was utilized to measure the expression of different genes in Kyoto encyclopedia of genes and genomes pathways. We randomly selected eight lncRNA genes and validated them by quantitative reverse transcription polymerase chain reaction (RT-qPCR). Results: We found that 182 lncRNA transcripts, including 102 upregulated and 80 downregulated transcripts, were differentially expressed between Kazakh cattle and Xinjiang brown cattle. The results of RT-qPCR were consistent with the sequencing results. Enrichment analysis and functional annotation of the target genes revealed that the differentially expressed lncRNAs were associated with the mitogen-activated protein kinase, Ras, and phosphatidylinositol 3-kinase (PI3k)/Akt signaling pathways. We also constructed a lncRNA/mRNA coexpression network for the PI3k/Akt signaling pathway. Conclusion: Our study provides insights into cattle muscle-associated lncRNAs and will contribute to a more thorough understanding of the molecular mechanism underlying muscle growth and development in cattle.

        Nonlinear endomicroscopy using a double-clad fiber coupler.

        <P>A double-clad fiber coupler is developed to be used in two-photon-excited fluorescence endomicroscopy to replace a dichroic mirror and separate the fluorescence signal from the excitation laser beam. With the double-clad fiber coupler, the endomicroscope becomes more compact, easier to be aligned, and more stable in alignment. The double-clad fiber coupler can transmit 62% of the excitation laser beam through the core. The fluorescence collection efficiency of the double-clad fiber coupler is 34%, which is, to the best of our knowledge, the highest fluorescence collection efficiency achieved by couplers used in two-photon-excited fluorescence endomicroscopes. As a result, the contrast of endomicroscopy imaging is enhanced.</P>

        Effect of fangchinoline on root resorption during rat orthodontic tooth movement

        Objective: To evaluate the short-term effect of fangchinoline, an anti-inflammatory drug widely used in Asia, on root resorption that is associated with orthodontic tooth movement. Methods: Twenty-four Wistar rats were randomly divided into 6 groups. Mesial forces of 0, 50, or 100 g were applied to the maxillary first molar of the rats in each group for 14 days by activating nickel-titanium closed-coil springs. One-half of the rats receiving each of these treatments also received injections of 200 ${\mu}L$ fangchinoline every 2 days. Finally, movement of the maxillary first molars was measured using digitized radiographs. The molars were extracted and the surfaces of the root resorption craters were recorded using a scanning electron microscope. The distance the molars moved and resorption-area ratio was measured, and results were analyzed using 2-way ANOVA tests. Results: There were no statistical differences in the distances the first molars moved under 50 or 100 g force, regardless of treatment with fangchinoline. However, the resorption area ratios were significantly smaller in those rats that were treated with both tension and fangchinoline than in those rats treated by tension alone. Conclusions: Fangchinoline reduced the resorption area ratio in rats and is therefore an important means of alleviating root resorption.

        Source and Negative Effects of Macro-Inclusions in Titanium Stabilized Ultra Low Carbon Interstitial Free (Ti-IF) Steel

        The source and negative effects of macro-inclusions in Ti-IF steel were studied by sliming extraction, ultrasonic detection, depth corrosion and SEM/EDS. High SiO2 content inclusions, and Al2O3 and MgO· CaO inclusions were observed after sliming. Ultrasonic results showed that defects in the surfboard with sizes 0.5 mm-3mm were mainly distributed in the inner arc and outer arc sides close to 1/8 of the slab’s surface. The number of defects in the inner arc side was 1.5 to 2 times of that in the outer arc side. Defects in the surfboard and cold-rolled plant were formed due to inclusions which were similar to that observed in the sliming sample.
