        DISPERSION OF CARBON NANOFIBERS IN AQUEOUS SOLUTION

        Carbon nano¯bers (CNFs) were puri¯ed by the method of heat treating, and stable homogeneous suspensions of heat-treated CNFs were prepared by using methylcellulose (MC) as a surfactant. The heat-treated CNFs were characterized by di®erential thermal analysis (DTA) and thermogravimetry analysis (TGA), and the sonication-driven dispersion of CNFs in aqueous solution was investigated by measuring ultraviolet absorbency (UV absorbency), zeta potential, adsorption isotherm of MC on CNFs and surface tension of CNF suspensions. In these CNF suspensions, the zeta potentials of CNFs progressively increase with increasing MC concentration, and change from ?15.4?0 mV. The surface tension of CNF suspensions decrease from 38.87mN/m to 36.54mN/m. The adsorption isotherm of MC on CNFs shows an adsorption platform (namely \L" style), and the adsorption isotherm reaches the saturation plateau at MC concentration of about 0.4 g/L. All results show that the optimum concentration ratio of MC to CNFs is 2:1 for dispersion.

      • ODV-Associated Proteins of the <i>Pieris rapae</i> Granulovirus

        <P><I>Alphabaculovirus</I> (lepidopteran-specific nucleopolyhedroviruses, NPV) and <I>Betabaculovirus</I> (granuloviruses, GV) are two main genera of the family Baculoviridae. The virion proteomes of <I>Alphabaculovirus</I> have been well studied; however, the <I>Betabaculovirus</I> virion compositions remain unclear. <I>Pieris rapae</I> granulovirus (PrGV) can kill larvae of <I>P. rapae</I>, a worldwide and important pest of mustard family crops. In this study, the occlusion-derived virus (ODV)-associated proteins of PrGV were identified using three mass spectrometry (MS) approaches. The MS analyses demonstrated that 47 proteins were present in PrGV-ODV. Of the 47 PrGV-ODV proteins, 33 have homologues identified previously in other baculovirus ODV/BVs, whereas 14 (P10, Pr21, Pr29, Pr35, Pr42, Pr54, P45/48, Pr83, Pr84, Pr89, Pr92, Pr111, Pr114 and FGF3) were newly identified ODV proteins. Seven of the 14 newly identified ODV proteins are specific to <I>Betabaculovirus</I>, including Pr35, Pr42, Pr54, Pr83, Pr84, Pr111 and Pr114. Furthermore, the data derived from these MS approaches were validated by immunoblotting analysis using antisera prepared from 11 randomly selected recombinant PrGV-ODV proteins (including 5 <I>Betabaculovirus</I>-unique proteins). Comparison analyses revealed the similar and different compositions between <I>Betabaculovirus</I> and <I>Alphabaculovirus</I> virions, which deepen our understanding of the baculovirus virion structure and provide helpful information on <I>Betabaculovirus</I>–host interaction studies.</P><P>We used three mass spectrometry (MS) approaches to identify the occlusion-derived virus (ODV)-associated proteins of the <I>Pieris rapae</I> Granulovirus.       • High Expression of Forkhead Box Protein C2 is Related to Poor Prognosis in Human Gliomas

        Background: Increasing evidence has indicated that high Forkhead box protein C2 (FOXC2) level is closely associated with the development, progression, and poor prognosis of a variety of tumors. However, the relationship between FOXC2 and the progression of human gliomas remains to be clarified. The aim of present study was to assess FOXC2 expression and to explore its contribution in human gliomas. Materials and Methods: Realtime quantitative PCR was performed to examine FOXC2 expression in 85 pairs of fresh frozen glioma tissues and corresponding non-neoplastic brain tissues. Associations of FOXC2 expression with clinicopathological factors and prognosis of glioma patients were statistically analyzed. Results: The relative mRNA expression of FOXC2 was significantly higher in glioma tissues than the corresponding non-neoplastic brain tissues (p<0.001). In addition, high FOXC2 expression was significantly associated with advanced pathological grade (P=0.005) and the low Karnofsky performance score (KPS) (p=0.003), correlating with poor survival (p<0.001). Furthermore, multivariate Cox regression analysis showed that high FOXC2 expression was an independent predictor of overall survival (p=0.006). Conclusions: FOXC2 may act as an oncogenic gene and represent a potential regulator of aggressive development and a candidate prognostic marker in human gliomas.

        Source and Negative Effects of Macro-Inclusions in Titanium Stabilized Ultra Low Carbon Interstitial Free (Ti-IF) Steel

        The source and negative effects of macro-inclusions in Ti-IF steel were studied by sliming extraction, ultrasonic detection, depth corrosion and SEM/EDS. High SiO2 content inclusions, and Al2O3 and MgO· CaO inclusions were observed after sliming. Ultrasonic results showed that defects in the surfboard with sizes 0.5 mm-3mm were mainly distributed in the inner arc and outer arc sides close to 1/8 of the slab’s surface. The number of defects in the inner arc side was 1.5 to 2 times of that in the outer arc side. Defects in the surfboard and cold-rolled plant were formed due to inclusions which were similar to that observed in the sliming sample.

        Ordered Morphologies on the Binary Blend of Diblock Copolymers Film Induced by Nanoparticles

        In this study, we investigate the ordered morphologies on the binary blend of diblock copolymers film induced by the mobile wettable nanoparticles on the substrate. The film forms highly ordered microphase and macrophase morphologies. We constructed the phase diagram, which suggests that the ordered-striped macrophase structures are due to the competition equilibrium of the wetting strength and the chemical potential amplitude upon the nanoparticles. The morphologies of the binary blend film form the striped pattern of different periods with the changing oscillation period of the chemical potential. The short-range interaction between monomers only alters the microphase structures. We analyzed the variation in the interfacial energy, the wetting energy and the particles/substrate's coupling energy on different morphologies. The results provide a new control mechanism to stabilize the ordered microphase and macrophase structures within the copolymer blend film.

        Ultimate strength behavior of steel-concrete-steel sandwich beams with ultra-lightweight cement composite, Part 1: Experimental and analytical study

        Ultra-lightweight cement composite (ULCC) with a compressive strength of 60 MPa and density of 1450 kg/m³ has been developed and used in the steel-concrete-steel (SCS) sandwich structures. ULCC was adopted as the core material in the SCS sandwich composite beams to reduce the overall structural weight. Headed shear studs working in pairs with overlapped lengths were used to achieve composite action between the core material and steel face plates. Nine quasi-static tests on this type of SCS sandwich composite beams were carried out to evaluate their ultimate strength performances. Different parameters influencing the ultimate strength of the SCS sandwich composite beams were studied and discussed. Design equations were developed to predict the ultimate resistance of the cross section due to pure bending, pure shear and combined action between shear and moment. Effective stiffness of the sandwich composite beam section is also derived to predict the elastic deflection under service load. Finally, the design equations were validated by the test results.

        Genomic identification of microRNA promoters and their cis-acting elements in Populus

        MicroRNAs (miRNAs) are non-coding singlestranded RNAs of appropriately 21 nucleotides in length that negatively regulate post-transcriptional processes in plants and animals. In recent years, while an increasing number of miRNAs has been identified in various species, little is known regarding the specific transcriptional regulation of miRNAs in Populus. In this investigation, 368 miRNAs belonging to 37 miRNA families in Populus were analyzed to identify promoter regions and predict the distribution patterns of core promoter elements [TATA boxes and transcription start sites (TSSs)], as well as main cisacting elements through a bioinformatics approach. A total of 368 precursor miRNA (pre-miRNA) gene loci belonging to 213 unique miRNAs were detected. We identified 229 promoters and further predicted 121 TSSs and TATA boxes for 139 pre-miRNAs. The conserved motifs, base composition and the average free energy profiles in the neighboring regions of TSSs were further analyzed. In addition, 101 types of cis-acting elements, such as anaerobic induction elements, abscisic acid-response elements, and MYB binding sites, were predicted in the predicted promoter regions of the pre-miRNAs. The data obtained in our investigation may aid in the analysis of specific upstream sequences of pre-miRNAs for differential expression and further our understanding of the molecular biology of miRNAs in Populus.

        Transcriptome-wide identification of miRNA targets and a TAS3- homologous gene in Populus by degradome sequencing

        Background Degradome sequencing has been applied to identify miRNA-directed mRNA cleavage and understand the biological function of miRNAs and their target genes in plants defense to stress. miRNAs involved in the response to cold stress have been identified in Populus, however, there are few reports about the validated targets of miRNAs in Populus under cold stress. Objectives The primary objective of this investigation was to globally identify and validate the targets of the miRNAs and regulatory components in Populus under cold stress. Methods Populus plantlets grown in vitro were treated with cold (4 °C for 8 h) and total RNA was extracted using Trizol reagent. Approximately 200 μg total RNA was used for the construction of the degradome library, and degradome sequencing was conducted on an Illumina HiSeq 2000. The sequences were mapped to Populus genome using SOAP 2.0 and then were collected for degradome analysis. Additionally, trans-acting siRNA sequences from transacting siRNA gene 3 sequences and mature miRNAs cleaved from precursor miRNAs of Populus were analyzed. 5′ RNA ligase-mediated-RACE (5′-RACE) were further conducted. Results 80 genes were experimentally determined to be the target of 51 unique miRNAs, including three down-regulated miRNAs (pto-miR156k, pto-miR169i-m, and pto-miR394a-5p/b-5p) and two up-regulated miRNAs (pto-miR167a-d and pto-miR167f/g). The specificity and diversity of cleavage sites of miRNA targets were validated through 5′-RACE experiment and the results were similar with that of degradome sequencing, further supporting the empirical cleavage of miRNAs on targets in vivo in Populus. Interestingly, the TAS-homologous gene pto-TAS3 (EF146176.1) was identified and 11 potential ta-siRNAs [D1(+)–D11(+)] and their possible biogenesis sites within the pto-TAS3 transcript sequence were predicted in Populus. In addition, the biosynthesis of miRNA from precursor miRNA (pre-miRNA) was also validated through the detection of a total of 17 pre-miRNAs. Conclusion Our investigation expands the application of degradome sequencing for evaluating miRNA regulatory elements and evidence of the miRNA synthesis process, and provides empirical evidence of bona fide cleavage of target genes by miRNAs in Populus, which might be used for the research of miRNA-mediated regulation mechanism and molecular improvement of resistance to cold stress.

      • Allogeneic Hemopietic Stem Cell Transplants for the Treatment of B Cell Acute Lymphocytic Leukemia

        Objective: Explore the feasibility of allo-hemopietic stem cell transplants in treating patients with B cell acute lymphocytic leukemia. Methods: Between september 2006 and February 2011, fifteen patients with B cell acute lymphocytic leukemia (ALL) were treated by allo-hemopietic stem cell transplants (HSCT). Stem cell sources were peripheral blood. Six patients were conditioned by busulfan (BU) and cyclophosphamide (CY) and nine patients were conditioned with TBI and cyclophosphamide (CY). Graft versus host disease (GVHD) prophylaxis regimen consisted of cyclosporine A (CSA), methotrex ate (MTX) and mycophenolatemofetil (MMF). Results: Patients received a median of $7.98{\times}10^8{\cdot}kg^{-1}$ ($5.36-12.30{\times}10^8{\cdot}kg^{-1}$) mononuclear cells (MNC). The median time of ANC> $0.5{\times}10^9/L$ was day 12 (10-15), and PLT> $20.0{\times}10^9/L$ was day 13 (11-16). Extensive acute GVHD occurred in 6 (40.0%) patients, and extensive chronic GVHD was recorded in 6 (40.0%) patients. Nine patients were alive after 2.5-65 months follow-up. Conclusion: Allogeneic stem cell transplant could be effective in treating patients with B cell acute lymphocytic leukemia.
