Therapeutic Co-targeting of WEE1 and ATM Downregulates PD-L1 Expression in Pancreatic Cancer

Purpose Pancreatic cancer (PC) is one of the most lethal cancers worldwide, but there are currently no effective treatments. The DNA damage response (DDR) is under investigation for the development of novel anti-cancer drugs. Since DNA repair pathway alterations have been found frequently in PC, the purpose of this study was to test the DDR-targeting strategy in PC using WEE1 and ATM inhibitors. Materials and Methods We performed in vitro experiments using a total of ten human PC cell lines to evaluate antitumor effect of AZD1775 (WEE1 inhibitor) alone or combination with AZD0156 (ATM inhibitor). We established Capan-1–mouse model for in vivo experiments to confirm our findings. Results In our research, we found that WEE1 inhibitor (AZD1775) as single agent showed anti-tumor effects in PC cells, however, targeting WEE1 upregulated p-ATM level. Here, we observed that co-targeting of WEE1 and ATM acted synergistically to reduce cell proliferation and migration, and to induce DNA damage in vitro. Notably, inhibition of WEE1 or WEE1/ATM downregulated programmed cell death ligand 1 expression by blocking glycogen synthase kinase-3! serine 9 phosphorylation and decrease of CMTM6 expression. In Capan-1 mouse xenograft model, AZD1775 plus AZD0156 (ATM inhibitor) treatment reduced tumor growth and downregulated tumor expression of programmed cell death ligand 1, CMTM6, CD163, and CXCR2, all of which contribute to tumor immune evasion. Conclusion Dual blockade of WEE1 and ATM might be a potential therapeutic strategy for PC. Taken together, our results support further clinical development of DDR-targeting strategies for PC.

Dendritic cell (DC) activator, thymic stromal lymphopoietin (TSLP) induces DCs to produce Th2-attracting chemokines, causing differentiation of CD4+ and CD8+ T cells into effector cells. The differentiated cells are characterized by a typical pro-allergic phenotype. In this study, we identified four single nucleotide polymorphisms (SNPs) and one variation site (g.-305delT) from the results of scanned human TSLP gene. The SNPs are as following: g.-1914A>G and g.-847C>T in promoter, g.-82C>T in 5` UTR and g.1117C>T in intron 2. We also evaluated the association of genotype and allele frequencies of these SNPs between non-allergic rhinitis controls and allergic rhinitis patients. We further investigated possible correlations between each genotype with serum total IgE levels and peripheral blood eosinophil counts in allergic rhinitis patients. The frequencies of haplotype constructed by these SNPs between these two groups were also compared. Our results demonstrate the two SNPs (g.-1914A>G and g.-847C>T) were associated with susceptibility of allergic rhinitis (P=0.006 and P=0.015 respectively), however, serum total IgE levels were not significantly related with the SNPs.

5-Methoxy-8-(2-hydroxy-3-buthoxy-3-methylbutyloxy)-psoralen (MP) is a medicinal herbal product isolated from Angelica dahurica that inhibits the cyclooxygenase-2 (COX-2)-dependent phase of prostaglandin $D_2 (PGD_2)$ generation in bone marrow-derived mast cells ($IC_{50}$, $23.5\;{\mu}M$). Western blotting with specific anti-COX-2 antibodies showed that the decrease in $PGD_2$ production was accompanied by a decrease in COX-2 protein levels. In addition, this compound consistently inhibited the production of leukotriene $C_4$ in a dose dependent manner, ($IC_{50}$, $2.5\;{\mu}M$). These results demonstrate that MP inhibits both cyclooxygenase-2 and 5- lipoxygenase activity. Furthermore, this compound also inhibited the degranulation reaction $(IC_{50}$, $4.1\;{\mu}M$). Therefore, this compound might provide a basis for novel anti-inflammatory drug development.
The “Fake Marriage” Test in Taiwan: Gender, Sexuality, and Border Control

According to many reports, migrant sex workers often use marriages of convenience to cross national borders in order to avoid laws criminalizing commercial sex in many destination countries. Taiwan is one of the countries developing strategies to prevent this illicit migration, particularly through the application of a fake marriage test. Based on in-depth interviews with eighteen Chinese migrant sex workers and thirteen officers of Taiwan’s National Immigration Agency (NIA), this article argues, first, that the discourse of “national security” has been widely drawn on to justify Taiwan’s rigid border control at the expense of stigmatized Chinese prostitutes who have been scapegoated. Border control is therefore not only racialized or classed but also sexualized, to the extent that all Chinese migrant women are considered potential prostitutes. Second, this article reveals how the exclusion of and hostility toward Chinese sex workers are simultaneously linked with a gender regime that seeks to exclude Chinese spouses who deviate from Taiwanese gender and social norms. The border is therefore a contested site where gender, sexuality, and nationality are interwoven.
