Interleukine-4 (IL-4) is a cytokine that plays an important role in the immune system and recognized as a biological medicine. Therefore, there is a demand for the production of IL-4 with high performance. The expression of a recombinant IL-4 protein in the prokaryotic system usually results in the formation of an inclusion body. To date, the solution to obtain those active products without the refolding process remains to be established. In this study, we tried to acquire a biologically active recombinant Mus musculus IL-4 (rMmIL-4) using a silkworm-baculovirus expression vector system (silkworm-BEVS). We constructed two recombinant baculoviruses coding rMmIL-4 with the distinct location of affinity purification tags and succeeded in the expression and purification of rMmIL-4 proteins directly without the refolding process. Both purified proteins displayed comparable biological activity to the commercial proteins produced by the E. coli expression system. Besides, we performed screening of silkworm strains to seek optimal hosts for the mass-production of rMmIL-4. Intriguingly, we found that some silkworm strains showed significantly higher secretion levels of rMmIL-4 in silkworm sera. Our study provides meaningful insights into the industrial-scale production of rMmIL-4 with high productivity for pharmaceutical applications in the future.

To investigate the difference in the disinfectant efficiency of ozone microbubbles (O3MB) and ozone millibubbles (O3MMB), the morphological change of the treated Fusarium oxysporum f. sp. melonis spores was observed with scanning and transmission electron microscopies (SEM and TEM). The disinfectant efficiency of O3MB on F. oxysporum f. sp. melonis spores was greater than that of O3MMB. On observation with SEM, it was revealed that morphological change of F. oxysporum f. sp. melonis spores was caused by O3MB and O3MMB, and damage to the spore surfaces by O3MB occurred sooner than that by O3MMB. On observation with TEM, it was furthermore confirmed that F. oxysporum f. sp. melonis spores treated with O3MB induced wavy deformation of cell membrane and the intracellular change different from that with O3MMB. Therefore, the greater disinfection efficiency of O3MB was suggested to be caused due to the function of the MB in addition to the oxidative power of O3.

HIGH ANGULAR RESOLUTION [Fe II] λ1.644 μ SPECTROSCOPY OF YSOS WITH SUBARU TELESCOPE

We present results of the velocity-resolved spectroscopy of the [Fe II] $\lambda$1.644${\mu}m$ emission toward outflow sources with the Subaru Telescope at the angular resolution of 0.apos;16 ${\~}$ 0.apos;5 arcseconds. The observed sources are L1551 IRS 5, DG Tau, HL Tau and RW Aur, which are located in the Taurus-Aurigae Molecular Cloud, one of the closest star forming regions (0.apos;1 = 14 AU). We were able to resolve outflow structure in the vicinity of the sources at a scale of a few tens of AU. The position-velocity diagram of each object shows two velocity components: the high velocity component (HVC: 200 - 400 km $s^{-l}$) and the low velocity component (LVC: 50 - 150 km $s^{-l}$), which are clearly distinct in space and velocity. The HVC may be a highly collimated jet presumed from its narrow velocity width and high velocity. The LVC, on the other hand, may be a widely opened disk wind inferred from its broad velocity width and low velocity. The spectrum taken perpendicular to the L1551 IRS 5 outflow at its base shows that the LVC has a spatially wide subcomponent, supporting the above interpretation. We demonstrated that the [Fe II] 1.644 $\mu$ spectroscopy is a very powerful tool for the studies of fast jets and winds that directly emanate from star-disk systems.

We have investigated in detail the dependence of the electroluminescent characteristics on the thin LiF layer inserted under theelectrode layer of multi-layered PVCz polymer light emitting diode (PLED). Clear dependency has been obtained and reduction inthe operating voltage, improvement in luminance and the luminous and current eciencies have been realized with the optimizationof the thickness. Albumin-Bilirubin Score Predicts Tolerability to Adjuvant S-1 Monotherapy after Curative Gastrectomy

Purpose: Due to adverse events, dose reduction or withdrawal of adjuvant chemotherapy is required for some patients. To identify the predictive factors for tolerability to postoperative adjuvant S-1 monotherapy in gastric cancer (GC) patients, we evaluated the predictive values of blood indicators. Materials and Methods: We analyzed 98 patients with pStage II/III GC who underwent postoperative adjuvant S-1 monotherapy. We retrospectively analyzed correlations between 14 parameters obtained from perioperative routine blood tests to assess their influence on the withdrawal of postoperative adjuvant S-1 monotherapy, within 6 months after discontinuation. Results: Postoperative adjuvant chemotherapy was discontinued in 21 patients (21.4%) within 6 months. Univariable analysis revealed that high preoperative albumin-bilirubin (ALBI) scores had the highest odds ratio (OR) for predicting the failure of adjuvant S-1 chemotherapy (OR, 6.47; 95% confidence interval [CI], 2.08-20.1; cutoff value, -2.696). The high ALBI group had a significantly shorter time to failure of postoperative adjuvant S-1monotherapy (hazard ratio, 3.48; 95% CI, 1.69-7.25; P=0.001). Multivariable analysis identified high preoperative ALBI score as an independent prognostic factor for tolerability (OR, 10.3; 95% CI, 2.33-45.8; P=0.002). Conclusions: Preoperative ALBI shows promise as an indicator associated with the tolerability of adjuvant S-1 monotherapy in patients with pStage II/III GC.
