Selection for Duration of Fertility and Mule Duck White Plumage Colour in a Synthetic Strain of Ducks (Anas platyrhynchos)

        H.C. Liu,J.F. Huang,S.R. Lee,H.L. Liu,C.H. Hsieh,C.W. Huang,M.C. Huang,C. Tai,J.P. Poivey,R. Rouvier,Y.S. Cheng 아세아·태평양축산학회 2015 Animal Bioscience Vol.28 No.5

A synthetic strain of ducks (Anas platyrhynchos) was developed by introducing genes for long duration of fertility to be used as mother of mule ducklings and a seven-generation selection experiment was conducted to increase the number of fertile eggs after a single artificial insemination (AI) with pooled Muscovy semen. Reciprocal crossbreeding between Brown Tsaiya LRI-2 (with long duration of fertility) and Pekin L-201 (with white plumage mule ducklings) ducks produced the G0. Then G1 were intercrossed to produce G2 and so on for the following generations. Each female duck was inseminated 3 times, at 26, 29, and 32 weeks of age. The eggs were collected for 14 days from day 2 after AI. Individual data regarding the number of incubated eggs (Ie), the number of fertile eggs at candling at day 7 of incubation (F), the total number of dead embryos (M), the maximum duration of fertility (Dm) and the number of hatched mule ducklings (H) with plumage colour were recorded. The selection criterion was the breeding values of the best linear unbiased prediction animal model for F. The results show high percentage of exhibited heterosis in G2 for traits to improve (19.1% for F and 12.9% for H); F with a value of 5.92 (vs 3.74 in the Pekin L-201) was improved in the G2. Heritabilities were found to be low for Ie (h2 = 0.07±0.03) and M (h2 = 0.07±0.01), moderately low for Dm (h2 = 0.13±0.02), of medium values for H (h2 = 0.20±0.03) and F (h2 = 0.23±0.03). High and favourable genetic correlations existed between F and Dm (rg = 0.93), between F and H (rg = 0.97) and between Dm and H (rg = 0.90). The selection experiment showed a positive trend for phenotypic values of F (6.38 fertile eggs in G10 of synthetic strain vs 5.59 eggs in G4, and 3.74 eggs in Pekin L-201), with correlated response for increasing H (5.73 ducklings in G10 vs 4.86 in G4, and 3.09 ducklings in Pekin L-201) and maximum duration of the fertile period without increasing the embryo mortality rate. The average predicted genetic response for F was 40% of genetic standard deviation per generation of selection. The mule ducklings’ feather colour also was improved. It was concluded that this study provided results for a better understanding of the genetics of the duration of fertility traits in the common female duck bred for mule and that the selection of a synthetic strain was effective method of improvement.

      • FACT complex is required for DNA demethylation at heterochromatin during reproduction in Arabidopsis

        Frost, Jennifer M.,Kim, M. Yvonne,Park, Guen Tae,Hsieh, Ping-Hung,Nakamura, Miyuki,Lin, Samuel J. H.,Yoo, Hyunjin,Choi, Jaemyung,Ikeda, Yoko,Kinoshita, Tetsu,Choi, Yeonhee,Zilberman, Daniel,Fischer, R National Academy of Sciences 2018 PROCEEDINGS OF THE NATIONAL ACADEMY OF SCIENCES OF Vol.115 No.20

        <▼1>
Significance

The chromatin remodeling activities of the FACT (facilitates chromatin transactions) complex are required for many cellular functions, including transcription, DNA replication, and repair. Here, we demonstrate that the two FACT subunits, SSRP1 and SPT16, are also required for genome-wide DNA demethylation and regulation of gene imprinting during Arabidopsis reproduction. Without FACT, Arabidopsis seeds undergo abnormal development and exhibit aberrant DNA hypermethylation, including at imprinting control region loci. We show that FACT associates with the DEMETER (DME) DNA demethylase, facilitating DNA demethylation at over half of DME’s targets, specifically those which reside in heterochromatin. These results provide insight into upstream events in the DNA demethylation pathway and reveal the importance of chromatin remodeling for DNA demethylation during Arabidopsis reproduction.
</▼1><▼2>
The DEMETER (DME) DNA glycosylase catalyzes genome-wide DNA demethylation and is required for endosperm genomic imprinting and embryo viability. Targets of DME-mediated DNA demethylation reside in small, euchromatic, AT-rich transposons and at the boundaries of large transposons, but how DME interacts with these diverse chromatin states is unknown. The STRUCTURE SPECIFIC RECOGNITION PROTEIN 1 (SSRP1) subunit of the chromatin remodeler FACT (facilitates chromatin transactions), was previously shown to be involved in the DME-dependent regulation of genomic imprinting in Arabidopsis endosperm. Therefore, to investigate the interaction between DME and chromatin, we focused on the activity of the two FACT subunits, SSRP1 and SUPPRESSOR of TY16 (SPT16), during reproduction in Arabidopsis. We found that FACT colocalizes with nuclear DME in vivo, and that DME has two classes of target sites, the first being euchromatic and accessible to DME, but the second, representing over half of DME targets, requiring the action of FACT for DME-mediated DNA demethylation genome-wide. Our results show that the FACT-dependent DME targets are GC-rich heterochromatin domains with high nucleosome occupancy enriched with H3K9me2 and H3K27me1. Further, we demonstrate that heterochromatin-associated linker histone H1 specifically mediates the requirement for FACT at a subset of DME-target loci. Overall, our results demonstrate that FACT is required for DME targeting by facilitating its access to heterochromatin.
</▼2>

CD137 Is Required for M Cell Functional Maturation but Not Lineage Commitment

        Hsieh, E.H.,Fernandez, X.,Wang, J.,Hamer, M.,Calvillo, S.,Croft, M.,Kwon, B.S.,Lo, D.D. American Association of Pathologists and Bacteriol 2010 The American journal of pathology Vol.177 No.2

Mucosal immune surveillance depends on M cells that reside in the epithelium overlying Peyer's patch and nasopharyngeal associated lymphoid tissue to transport particles to underlying lymphocytes. M cell development is associated with B lymphocytes in a basolateral pocket, but the interactions between these cells are poorly understood. In a cell culture model of M cell differentiation, we found lymphotoxin/tumor necrosis factor α induction of CD137 (TNFRSF9) protein on intestinal epithelial cell lines, raising the possibility that CD137 on M cells in vivo might interact with CD137L expressed by B cells. Accordingly, while CD137-deficient mice produced UEA-1+ M cell progenitors in nasopharyngeal associated lymphoid tissue and Peyer's patch epithelium, they showed an abnormal morphology, including the absence of basolateral B cell pockets. More important, CD137-deficient nasopharyngeal associated lymphoid tissue M cells were defective in microparticle transcytosis. Bone marrow irradiation chimeras confirmed that while induction of UEA-1+ putative M cell precursors was not CD137-dependent, full M cell transcytosis function required expression of CD137 by radioresistant stromal cells as well as by bone marrow-derived cells. These results are consistent with a two-step model of M cell differentiation, with initial CD137-independent commitment to the M cell lineage followed by a CD137-CD137L interaction of M cells with CD137-activated B lymphocytes or dendritic cells for functional maturation.

      • SEARCH FOR THE RETURN OF ACTIVITY IN ACTIVE ASTEROID 176P/LINEAR

        Hsieh, Henry H.,Denneau, Larry,Fitzsimmons, Alan,Hainaut, Olivier R.,Ishiguro, Masateru,Jedicke, Robert,Kaluna, Heather M.,Keane, Jacqueline V.,Kleyna, Jan,Lacerda, Pedro,MacLennan, Eric M.,Meech, Kar American Institute of Physics 2014 The Astronomical journal Vol.147 No.4

We present the results of a search for the reactivation of active asteroid 176P/LINEAR during its 2011 perihelion passage using deep optical observations obtained before, during, and after that perihelion passage. Deep composite images of 176P constructed from data obtained between 2011 June and 2011 December show no visible signs of activity, while photometric measurements of the object during this period also show no significant brightness enhancements similar to that observed for 176P between 2005 November and 2005 December when it was previously observed to be active. An azimuthal search for dust emission likewise reveals no evidence for directed emission (i.e., a tail, as was previously observed for 176P), while a one-dimensional surface brightness profile analysis shows no indication of a spherically symmetric coma at any time in 2011. We conclude that 176P did not in fact exhibit activity in 2011, at least not on the level on which it exhibited activity in 2005, and suggest that this could be due to the devolatization or mantling of the active site responsible for its activity in 2005.

Cross-Coupled Narrow-Band YBCO Filters with Spurious Suppression by Using a Tap-Connection Technique

        L. M. Wang,Chung-I G. Hsu,D. S. Hsieh,L. H. Chen,M. L. Chu,S. M. Chiou 한국물리학회 2006 THE JOURNAL OF THE KOREAN PHYSICAL SOCIETY Vol.48 No.5I

Narrow-band microstrip cross-coupled band-pass filters based on a quadruplet geometry were designed for wireless-communication applications. High-Tc superconducting (HTS) filters were fabricated by patterning YBa2Cu3Oy (YBCO) films double-side deposited on 20-mm-square and 0.5- mm-thick LaAlO3 substrates. The filters with skew-symmetric feed were designed with a center frequency at 1.9 GHz and a bandwidth of 20 MHz. The inductive coupling between the non-adjacent resonators produced a pair of transmission zeros in the frequency response. Also observed was an additional pair of transmission zeros that enhance the out-of-band rejection characteristics. The realized YBCO filter showed a center frequency at 1.925 GHz with a bandwidth of 20 MHz and a minimum insertion loss of about 0.74 dB. Furthermore, a tap-connection technique was applied to cross-coupled HTS band-pass filters so as to suppress the second and/or the third passbands, which were spurious and undesirable. To improve the out-of-band performance of the band-pass filters, we tapped the input/output microstrip lines at certain locations. In this research, we confirmed that by tapping somewhere along the input/output microstrip lines, a /4 open stub or a /2 short stub, where refers to the guided wavelength at 2fo or 3fo, may effectively suppress harmonics at 2fo and/or 3fo by more than 25 dB. The simulated and the experimental results are discussed.

Applications of Laser-Fabricated Plasma Structures in Laser-Wakefield Accelerators, X-ray Lasers and Plasma Nonlinear Optics

        C.T. Hsieh,M.W. Lin,C.L. Chang,Y.C. Ho,S.Y. Chen,J. Wang,M.C. Chou,J.Y. Lin,C.H. Pai,P.H. Lin,L.C. Tai,S.H. Chen,G.Y. Tsaur,C.C. Kuo,T.Y. Chien 한국물리학회 2008 THE JOURNAL OF THE KOREAN PHYSICAL SOCIETY Vol.53 No.6

A general method for fabricating transient plasma structures with high-intensity laser pulses is developed to gain fine control over laser-plasma interactions. These structures have been used as programmable photonic devices in the development of laser-wakefield accelerators, soft X-ray lasers and plasma nonlinear optics driven by multi-terawatt laser pulses. Plasma ramps are used to control electron injection in laser-wakefield accelerators, plasma waveguides are used to enhance the efficiency of soft X-ray lasers by orders of magnitude and periodic plasma structures are used to achieve quasi-phase matching in relativistic harmonic generation. By scanning the interaction length with the same plasma-fabrication method, tomographic measurements are carried out to resolve the injection/acceleration process in laser-wakefield accelerators and amplification processes in X-ray lasers and relativistic harmonic generation. A theoretical analysis and a computer simulation are also carried out to provide insightful pictures of these processes. These research works show that by controlling plasma structures with optical fabrication methods, laser-plasma interaction can be engineered to expand and enrich the frontier of high-field physics.

Temperature Dependence of the Stokes Shift in Tensile InGaN/GaN MQWs with Advanced Buer Layers

        D. R. Hang,M. M. C. Chou,M. H. Hsieh,M. Heuken 한국물리학회 2008 THE JOURNAL OF THE KOREAN PHYSICAL SOCIETY Vol.53 No.3

The influence of buer layers on the optical absorption and the emission properties of tensile InGaN/GaN multiple quantum wells (MQWs) has been studied. Here, sets of buffer structures consisting of low-temperature AlN and high-temperature AlN/AlGaN/GaN stacks have been employed to compensate for the detrimental eects brought about by the large lattice mismatch and by the difference in thermal expansion coefficients when silicon substrates are used. We found the biaxial tensile strain could be reduced and that the internal quantum efficiency could be considerably improved. Moreover, the Stokes shift could be reduced and its temperature variation became stabilized. The underlying mechanism is mainly due to the quantum confined Stark effect while the contribution from the localization effect is relatively small in our tensile InGaN/GaN MQWs.

Effects of mushroom waster medium and stalk residues on the growth performance and oxidative status in broilers

        Hsieh, Y.C.,Lin, W.C.,Chuang, W.Y.,Chen, M.H.,Chang, S.C.,Lee, T.T. Asian Australasian Association of Animal Productio 2021 Animal Bioscience Vol.34 No.2

Objective: The study developed mushroom stalk residues as feed additives in the broiler diet for improving the growth performance and immunity of broilers as well as to increase the value of mushroom stalk residues. Methods: In total, 300 ROSS 308 broilers were randomly allocated into fifteen pens with five dietary treatments: i) control, basal diet; ii) CMWM, supplemented with 1% Cordyceps militaris waster medium (CM); iii) CMPE, supplemented with 0.5% CM+0.5% Pleurotus eryngii stalk residue (PE); iv) CMPS, supplemented with 0.5% CM+0.5% Pleurotus sajorcaju stalk residue (PS); v) CMFV, supplemented with 0.5% CM+0.5% Fammulina velutipes stalk residue (FV). Results: The chemical analysis results showed that CM extracts, PE extracts, PS extracts, and FV extracts contain functional components such as polysaccharides and phenols and have both 2, 2-diphenyl-1-picryl-hydrazyl-hydrate scavenging and Ferrous scavenging capacities. The group CMWM saw increased body weight gain and feed conversion rate and the promotion of jejunum villus growth, but there is no significant difference in the intestinal bacteria phase. Antioxidant genes in the nuclear factor (erythroid-derived 2)-like 2 (Nrf2)- antioxidant responsive element pathway among the groups are significantly higher than that of the control group, especially in group CMWM. Conclusion: The mushroom stalk residues have antioxidant functional components, can improve the intestinal health and body weight gain of chickens, and can activate the antioxidant pathway of Nrf2 to increase the heme oxygenase-1 expression. The treatment with 1% CM was the most promising as a feed additive.

The Clinical Correlations between Diabetes, Cigarette Smoking and Obesity on Intervertebral Degenerative Disc Disease of the Lumbar Spine

        Ande M. Jakoi,Gurpal Pannu,Anthony D’Oro,Zorica Buser,Martin H. Pham,Neil N. Patel,Patrick C. Hsieh,John C. Liu,Frank L. Acosta,Raymond Hah,Jeffrey C. Wang 대한척추외과학회 2017 Asian Spine Journal Vol.11 No.3

Study Design: Retrospective analysis of a nationwide private insurance database. Chi-square analysis and linear regression models were utilized for outcome measures. Purpose: The purpose of this study was to investigate any relationship between lumbar degenerative disc disease, diabetes, obesity and smoking tobacco. Overview of Literature: Diabetes, obesity, and smoking tobacco are comorbid conditions known to individually have effect on degenerative disc disease. Most studies have only been on a small populous scale. No study has yet to investigate the combination of these conditions within a large patient cohort nor have they reviewed the combination of these conditions on degenerative disc disease. Methods: A retrospective analysis of insurance billing codes within the nationwide Humana insurance database was performed, using PearlDiver software (PearlDiver, Inc., Fort Wayne, IN, USA), to identify trends among patients diagnosed with lumbar disc degenerative disease with and without the associated comorbidities of obesity, diabetes, and/or smoking tobacco. Patients billed for a comorbidity diagnosis on the same patient record as the lumbar disc degenerative disease diagnosis were compared over time to patients billed for lumbar disc degenerative disease without a comorbidity. There were no sources of funding for this manuscript and no conflicts of interest. Results: The total number and prevalence of patients (per 10,000) within the database diagnosed with lumbar disc degenerative disease increased by 241.4% and 130.3%, respectively. The subsets of patients within this population who were concurrently diagnosed with either obesity, diabetes, tobacco use, or a combination thereof, was significantly higher than patients diagnosed with lumbar disc degenerative disease alone (p <0.05 for all). The number of patients diagnosed with lumbar disc degenerative disease and smoking rose significantly more than patients diagnosed with lumbar disc degenerative disease and either diabetes or obesity (p <0.05). The number of patients diagnosed with lumbar disc degenerative disease, smoking and obesity rose significantly more than the number of patients diagnosed with lumbar disc degenerative disease and any other comorbidity alone or combination of comorbidities (p <0.05). Conclusions: Diabetes, obesity and cigarette smoking each are significantly associated with an increased diagnosis of lumbar degenerative disc disease. The combination of smoking and obesity had a synergistic effect on increased rates of lumbar degenerative disc disease. Patient education and preventative care is a vital goal in prevention of degenerative disc disease within the general population.
