<P>The immunogenic nonhuman carbohydrate sequences in membrane proteins from porcine kidney were identified and characterized using MALDI-TOF MS and ESI-QTOF-MS. The MALDI profile, investigated by incubation with exoglycosidases, showed a series of about 40 carbohydrates that were identified as high mannose glycans (Man<SUB>3–9</SUB>GlcNAc<SUB>2</SUB>) and complex bi-, tri-, and tetra-antennary glycans with and without core fucose. The antennae of many of the complex glycans were terminated with α-galactose residues, with the numbers of these residues ranging from one up to the number of antennae. Negative ion ESI-MS/MS spectra confirmed the location of the α-galactose residues on the ends of the antennae. This total glycan profile of the membrane proteins from porcine kidney will thus provide important information for the study of molecular interactions between antigenic carbohydrates and proteins in xenotransplantation.</P>
Functional role of serotonin in insulin secretion in a diet-induced insulin-resistant state.

<P>The physiological role of serotonin, or 5-hydroxytryptamine (5-HT), in pancreatic β-cell function was previously elucidated using a pregnant mouse model. During pregnancy, 5-HT increases β-cell proliferation and glucose-stimulated insulin secretion (GSIS) through the Gαq-coupled 5-HT2b receptor (Htr2b) and the 5-HT3 receptor (Htr3), a ligand-gated cation channel, respectively. However, the role of 5-HT in β-cell function in an insulin-resistant state has yet to be elucidated. Here, we characterized the metabolic phenotypes of β-cell-specific Htr2b(-/-) (Htr2b βKO), Htr3a(-/-) (Htr3a knock-out [KO]), and β-cell-specific tryptophan hydroxylase 1 (Tph1)(-/-) (Tph1 βKO) mice on a high-fat diet (HFD). Htr2b βKO, Htr3a KO, and Tph1 βKO mice exhibited normal glucose tolerance on a standard chow diet. After 6 weeks on an HFD, beginning at 4 weeks of age, both Htr3a KO and Tph1 βKO mice developed glucose intolerance, but Htr2b βKO mice remained normoglycemic. Pancreas perfusion assays revealed defective first-phase insulin secretion in Htr3a KO mice. GSIS was impaired in islets isolated from HFD-fed Htr3a KO and Tph1 βKO mice, and 5-HT treatment improved insulin secretion from Tph1 βKO islets but not from Htr3a KO islets. Tph1 and Htr3a gene expression in pancreatic islets was not affected by an HFD, and immunostaining could not detect 5-HT in pancreatic islets from mice fed an HFD. Taken together, these results demonstrate that basal 5-HT levels in β-cells play a role in GSIS through Htr3, which becomes more evident in a diet-induced insulin-resistant state.</P>

Purpose: Controversy exists regarding the role preoperative urodynamic study for asymptomatic closed spinal dysraphism as it has failed to reveal the benefit in surgical decision and expectation of urological outcomes. We explore the relationship between preoperative videourodynamic study and postoperative urological outcomes after toilet training completed, focusing on their capability of spontaneous voiding. Methods: We retrospectively reviewed the data of 181 patients who underwent preventive spinal cord untethering and followed at least till the completion of toilet training. Before untethering, patients underwent preoperative videourodynamic study. Postoperative voiding function was evaluated in 3 phases: (1) till postoperative 6 months, (2) till the completion of toilet training, and (3) 2 years after toilet training. Changing distribution of emptying pattern at each period was described. Also, relevance to preoperative urodynamic parameters on spontaneous voiding and urinary continence after toilet training was assessed. Results: Spinal lipoma and low lying conus were found in 145 (80%) and 128 patients (70.7%), respectively. Spontaneous voiding was found in 125 (69.1%), 164 (90.6%), and 162 patients (89.5%) at postoperative 6 months, till the toilet training, and 2 years after toilet training, respectively. Videourodynamics helped to clarify the presence of vesicourethral synergy. This was correlated with spontaneous voiding at postoperative 6 months and better urinary continence after 2 years of toilet training. Conclusions: Eventual spontaneous voiding was achieved till toilet training in 90% patients following preventive untethering. Those showing preoperative vesicourethral synergy was associated with faster achievement of spontaneous voiding and better urinary continence when they enter elementary school.
Synthesis and Electrical Characterization of Magnetic Bilayer Graphene Intercalate

<P>We report synthesis and transport properties of the minimal graphite intercalation compound, a ferric chloride (FeCl<SUB>3</SUB>)<SUB><I>n</I></SUB> island monolayer inside bilayer graphene. Chemical doping by the intercalant is simultaneously probed by micro-Raman spectroscopy and Hall measurements. Quantum oscillations of conductivity originate from microscopic domains of intercalated and unintercalated regions. A slight upturn in resistance related to magnetic transition is observed. Evaluation of rice root oxidizing potential using digital image analysis

The aerenchymal transport of oxygen to rice roots has significantly influenced the anaerobic root zone of flooded paddy soils. Therefore, the visualization of redox dynamics may be useful to characterize rice root oxidation potentials and the dynamics of redox-influenced ions in the root zone of paddy soils. In order to investigate the interaction between root oxidation potential and Fe uptake of (a) six different rice cultivars (Oryza sativa L.; Chuchung, Dongjin, Ilmi, Junam, Nampyeong, and Samkwang) were monitored in a flooded paddy soil with the aid of rhizotron experiment throughout the vegetation period, (b) digital images of the root zone were taken at the important growing stages, and (c) rice Fe uptake was characterized simultaneously. The images were processed by image analysis to display the reduction and oxidation areas in the root zones, and the distinct areas which were colorized due to varying soil redox changes were localized and quantified. Oxidized areas were mainly observed in the surrounding active roots and in a distinct layer on the soil surface. The selected rice cultivars have shown significantly different root-oxidized areas at the same rice growing stage. Root-oxidized area was significantly and positively correlated with total Fe content of rice root, but negatively correlated with the inner root Fe content. Rice cultivars having higher root oxidation potential precipitated more Fe on the outer root surface in the form of Fe plaques. In conclusion, digital image analysis is an effective tool for evaluating the oxidizing potential of rice root under anaerobic soil condition.

목적: 회전근 개 질환 환자의 견봉하 점액낭 조직에서, cDNA Array 기법을 이용하여 염증 사이토카인 및 수용체들의 발현 양상을 알아보고자 하였다. 대상 및 방법: 회전근 개 질환 환자 18명과 정상 견봉하 점액낭 조직을 가진 환자 4명의 견봉하 점액낭 조직을 수술 도중 채취한 후, cDNA Array 기법을 이용하여 염증 반응에 관여하는 사이토카인 및 수용체들의 유전자 발현 양상을 두 군에서 비교 분석하였다. 통계학적 분석은 Mann Whitney U test와 t-test를 이용하였다. 결과: 회전근 개 질환 환자군에서 비교군에 비하여 유의한 수준으로 증가된 사이토카인들은 interleukin군(IL-1, 6, 12, 13, 15, 16 및 17)과 수용체들, lymphotoxin, small inducible cytokine군, chemokine receptor군(CCR 4, 6 및 7), 그리고 stromal cell derived factor-1 (SDF-1)이었다(p<0.05). 결론: 회전근 개 질환 환자의 견봉하 점액낭 조직에서 염증 반응에 관여하는 사이토카인의 발현이 유의하게 증가하였다. 이러한 결과는 견봉하 점액낭에서 염증반응이 활발히 진행되고 있음을 나타내며, 이러한 염증반응이 회전근 개 질환 환자에서 견관절 동통을 일으키는 중요한 유발 인자 중의 하나임을 시사한다. Purpose: To examine the expression pattern of inflammatory cytokines/receptors in the subacromial bursa of patients with rotator cuff disease using a cDNA(Complement DNA) Array technique. Materials and Methods: Twenty two human subacromial bursal specimens were obtained intraoperatively from patients during shoulder surgery (18 bursitis, 4 normal bursa). The RNA was isolated from the bursal tissues and the presence of gene expression was analyzed using a cDNA Array technique. The statistical differences between bursitis and the normal bursa specimens were determined using a Mann Whitney U test and Student's t-test. Results: cDNA Array analysis revealed a significant increase in the expression of several cytokine genes and their receptors in patients with subacromial bursitis compared with the controls (p<0.05). These cytokines included the interleukins (IL-1, 6, 12, 13, 15, 16, 17) and their receptors, Iymphotoxin, small inducible cytokines, chemokine receptor (CCR 4, 6, 7) and stromal cell derived factor-1 (SDF-1). Conclusion: This study demonstrated a significant increase in many inflammatory cytokines in the subacromial bursa of patients with rotator cuff disease. This suggests that there is an active inflammatory reaction at the subacromial bursa in rotator cuff disease.
