The saprophytic fungus Trichoderma reesei has long been used as a model to study microbial degradation of lignocellulosic biomass. The major cellulolytic enzymes of T. reesei are the cellobiohydrolases CBH1 and CBH2, which constitute more than 70% of total proteins secreted by the fungus. However, their physiological functions and effects on enzymatic hydrolysis of cellulose substrates are not sufficiently elucidated. Here, the cellobiohydrolase-encoding genes cbh1 and cbh2 were deleted, individually or combinatively, by using an auxotrophic marker-recycling technique in T. reesei. When cultured on media with different soluble carbon sources, all three deletion strains (Δcbh1, Δcbh2, and Δcbh1Δcbh2) exhibited no dramatic variation in morphological phenotypes, but their growth rates increased apparently when cultured on soluble cellulase-inducing carbon sources. In addition, Δcbh1 showed dramatically reduced growth and Δcbh1Δcbh2 could hardly grew on microcrystalline cellulose (MCC), whereas all strains grew equally on sodium carboxymethyl cellulose (CMC-Na), suggesting that the influence of the CBHs on growth was carbon source-dependent. Moreover, five representative cellulose substrates were used to analyse the influence of the absence of CBHs on saccharification efficiency. CBH1 deficiency significantly affected the enzymatic hydrolysis rates of various cellulose substrates, where acid pre-treated corn stover (PCS) was influenced the least. CBH2 deficiency reduced the hydrolysis of MCC, PCS, and acid pre-treated and delignified corncob but improved the hydrolysis ability of filter paper. These results demonstrate the specific contributions of CBHs to the hydrolysis of different types of biomass, which could facilitate the development of tailor-made strains with highly efficient hydrolysis enzymes for certain biomass types in the biofuel industry.

Position-pose Control of a Pneumatic 3-UPU Robot Based on Immersion and Invariance

For pneumatic servo systems, the piston movement, time-varying parameters, and modeling uncertainties make high-precision position control difficult. A third-order mathematical model was established for a pneumatic 3-UPU (universal-prismatic-universal) robot system to provide a roughly accurate reference model for the control algorithm. Subsequently, a high-precision position-pose control algorithm based on immersion and invariance (I&I) was developed, where the leakage flow in the cylinder was incorporated as an interference term in the state equation, and the state equation was expanded. In addition, through a reasonable design of the compensation function and adaptive rate, the invariance and attraction of the error flow pattern were realized. The disturbance error was estimated in real time based on the adaptive law. Through the sliding surface, the control rate was designed, and the position-pose control of the robot was realized. The results showed that the I&I controller exhibits a strong robustness with a steady-state control accuracy of approximately 0.3 mm and a dynamic (0.2 Hz) tracking mean-squared error less than 6.4 mm.
Double-Lead Content Search Scheme for Producer Mobility in Named Data Networking

In Named Data Networking (NDN), the processing of the packet is based on the data rather than the communication endpoint. It’s very suitable for the core demand of content sharing on the internet today. However, there are still some problems to be dealt with, including mobility support of the producer. By analyzing existing solutions, we put forward a solution finding MCS (Mobile Content Source) passively by HR(Home Repository) and leading interest packets to find MCS actively in the local scope at the same time to speed up to search MCS and to support NDN producer mobility. Triangle routing problem in single HR and low efficiency problem can be based on this solution. Experimental results show that our scheme does better in reducing delay with low handoff cost than other typical solutions.

Impacts of Relationship value on Loyalty of Online Group-Buying Customer in China With the development of e-commerce industry, online group-buying pattern has been one of the most popular online shopping patterns in China. Although the online group-buying platform has a large number of consumers, the consumer loyalty to specific online group-buying platform is low. For this phenomenon, scholars usually studied from the perspective of factors which influenced consumer behavior intentions. However, few studies focus on the component and measuring of relational benefits in the process of group-buying. Based on the perspective of consumers, this paper studied the relational benefits between consumers and online group-buying platform, and the perceived benefits of consumers who are in different relationship life cycle.This paper used SPSS to test the reliability and validity of data and carried some discussions about research model through factor analysis and regression analysis. The results indicated that relational benefits were significantly related to customer loyalty, that relationship quality and relationship life cycle was considered as mediator and moderator, respectively. Further, we made some management recommendations for operators in the online group-buying platform.
Incremental extended finite element method for thermal cracking of mass concrete at early ages

Thermal cracks are cracks that commonly form at early ages in mass concrete. During the concrete pouring process, the elastic modulus changes continuously. This requires the time domain to be divided into several steps in order to solve for the temperature, stress, and displacement of the concrete. Numerical simulations of thermal crack propagation in concrete are more difficult at early ages. To solve this problem, this study divides crack propagation in concrete at early ages into two cases: the case in which cracks do not propagate but the elastic modulus of the concrete changes and the case in which cracks propagate at a certain time. This paper provides computational models for these two cases by integrating the characteristics of the extended finite element algorithm, compiles the corresponding computational programs, and verifies the accuracy of the proposed model using numerical comparisons. The model presented in this paper has the advantages of high computational accuracy and stable results in resolving thermal cracking and its propagation in concrete at early ages.
