        Adiponectin Regulates Bone Marrow Mesenchymal Stem Cell Niche Through a Unique Signal Transduction Pathway: An Approach for Treating Bone Disease in Diabetes : Adiponectin Regulates BMSC Niche

        <P>Adiponectin (APN) is an adipocyte-secreted adipokine that exerts well-characterized antidiabetic properties. Patients with type 2 diabetes (T2D) are characterized by reduced APN levels in circulation and impaired stem cell and progenitor cell mobilization from the bone marrow for tissue repair and remodeling. In this study, we found that APN regulates the mobilization and recruitment of bone marrow-derived mesenchymal stem cells (BMSCs) to participate in tissue repair and regeneration. APN facilitated BMSCs migrating from the bone marrow into the circulation to regenerate bone by regulating stromal cell-derived factor (SDF)-1 in a mouse bone defect model. More importantly, we found that systemic APN infusion ameliorated diabetic mobilopathy of BMSCs, lowered glucose concentration, and promoted bone regeneration in diet-induced obesity mice. In vitro studies allowed us to identify Smad1/5/8 as a novel signaling mediator of APN receptor (AdipoR)-1 in BMSCs and osteoblasts. APN stimulation of MC3T3-E1 osteoblastic cells led to Smad1/5/8 phosphorylation and nuclear localization and increased SDF1 mRNA expression. Although APN-mediated phosphorylation of Smad1/5/8 occurred independently from adaptor protein, phosphotyrosine interaction, pleckstrin homology domain, and leucine zipper containing 1, it correlated with the disassembly of protein kinase casein kinase 2 and AdipoR1 in immunoprecipitation experiments. Taken together, this study identified APN as a regulator of BMSCs migration in response to bone injury. Therefore, our findings suggest APN signaling could be a potential therapeutic target to improve bone regeneration and homeostasis, especially in obese and T2D patients.</P>

        Macroscopic and Mesoscopic Characteristics of a Small-Span Metro Tunnel in the Development of a Disaster Under Load

        A similarity model with a volumetric similarity ratio of 1:100 and a granular flow model for the tunnel were designed. By comparing macroscopic and mesoscopic information (such as fracture process, strain evolution, stress transfer, crack propagation, and stress distribution) of the tunnel models under load, the failure mechanism of the metro tunnel under load was investigated. The result showed that: 1) under the loading path, the instability area of the tunnel is mainly distributed in the straight wall on both sides. When the load is 1.5 MPa, a large number of cracks on both sides of the straight wall run through, resulting in the initial failure of the rock mass; 2) the surface rock mass of arch bottom is under tensile stress and the deep rock mass is under pressure stress, therefore, the fracture does not develop continuously. The surface of straight wall produces continuous development crack under the action of tensile stress; 3) the arch bottom first responds during the stress redistribution of the small-span tunnel; the top and middle parts of the side walls of the running tunnel with greatest potential for damage respond most; 4) in the process of stress redistribution, the peak stress of the deep measuring points of the straight wall is greater than that of the free surface; 5) at the initial stage of loading, tensile cracks account for a high proportion of all cracks found. When the load is 1.5 MPa, the proportion of shear cracks increases to 28%, and when the load is 1.6 Mpa, the proportion of shear cracks increases to 31%. Finally, the tensile-shear effect triggers the failure of the tunnel.

        Exploration of the Mesoscopic Failure Process of Granite Based on a PDE Method

        The meso-structure of rock influences its macro-mechanical properties. In traditional digital image processing, the filter operator causes fuzzy edges. There is a lack of image processing methods which can conveniently, and quickly, obtain a continuous, smooth, and closed curve with a clear boundary. In this study, a Chan-Vese (C-V) module of the partial differential equation (PDE) method was used to realise the three-phase segmentation of granite. Then, the AUTOLISP language was adopted to allow automatic line connection of the discrete boundary points in the digital image. The meso-scale model was implemented into numerical calculations to explore the evolution of a meso-crack. The crack originated from the stress concentration resulting from the uncoordinated deformation of the non-homogeneous materials under a Brazilian splitting test. The crack first appeared inside the materials with weak mechanical properties along the main diagonal of specimens under uniaxial compression. A stress concentration first appeared on the quartz-mica contact surface under triaxial compression. The tensile stress enveloped the weaker mica and feldspar as the load increased.

      • Soluble P3HT-Grafted Graphene for Efficient Bilayer−Heterojunction Photovoltaic Devices

        <P>CH<SUB>2</SUB>OH-terminated regioregular poly(3-hexylthiophene) (P3HT) was chemically grafted onto carboxylic groups of graphene oxide (GO) <I>via</I> esterification reaction. The resultant P3HT-grafted GO sheets (G-P3HT) are soluble in common organic solvents, facilitating the structure/property characterization and the device fabrication by solution processing. The covalent linkage and the strong electronic interaction between the P3HT and graphene moieties in G-P3HT were confirmed by spectroscopic analyses and electrochemical measurements. A bilayer photovoltaic device based on the solution-cast G-P3HT/C<SUB>60</SUB> heterostructures showed a 200% increase of the power conversion efficiency (η = 0.61%) with respect to the P3HT/C<SUB>60</SUB> counterpart under AM 1.5 illumination (100 mW/cm<SUP>2</SUP>).</P><P><B>Graphic Abstract</B> <IMG SRC='http://pubs.acs.org/appl/literatum/publisher/achs/journals/content/ancac3/2010/ancac3.2010.4.issue-10/nn101671t/production/images/medium/nn-2010-01671t_0007.gif'></P><P><A href='http://pubs.acs.org/doi/suppl/10.1021/nn101671t'>ACS Electronic Supporting Info</A></P>

        Adsorption of phenol and 1-naphthol onto XC-72 carbon

        XC-72 carbon (XC-72) was characterized by SEM, XPS, N2 adsorption-desorption, particle size distribution analysis and potentiometric acid-base titration. The adsorption of phenol and 1-naphthol on XC-72 was studied as a function of contact time, pH, adsorbent content and temperature. The kinetic adsorption data were described well by the pseudo-second-order model. The adsorption isotherms of phenol were described well by Freundlich model, while the adsorption isotherms of 1-naphthol were fitted well by Langmuir model. The results demonstrated that XC-72 had much higher adsorption capacity for 1-naphthol than for phenol. The adsorption thermodynamic data were calculated from the temperature-dependent adsorption isotherms at T=293, 313 and 333 K, and the results indicated that the adsorption of phenol was an exothermic process, whereas the adsorption of 1-naphthol was an endothermic process. XC-72 is a suitable material for the preconcentration of phenol and 1-naphthol from large volumes of aqueous solutions.

        99mTc-3PRGD2 SPECT/CT Imaging for Diagnosing Lymph Node Metastasis of Primary Malignant Lung Tumors

        Objective: To evaluate 99mtechnetium-three polyethylene glycol spacers-arginine-glycine-aspartic acid (99mTc-3PRGD2) singlephoton emission computed tomography (SPECT)/computed tomography (CT) imaging for diagnosing lymph node metastasis of primary malignant lung neoplasms. Materials and Methods: We prospectively enrolled 26 patients with primary malignant lung tumors who underwent 99mTc- 3PRGD2 SPECT/CT and 18F-fluorodeoxyglucose (18F-FDG) positron emission tomography (PET)/CT imaging. Both imaging methods were analyzed in qualitative (visual dichotomous and 5-point grades for lymph nodes and lung tumors, respectively) and semiquantitative (maximum tissue-to-background radioactive count) manners for the lymph nodes and lung tumors. The performance of the differentiation of lymph nodes with and without metastasis was determined at the per-lymph node station and perpatient levels using histopathological results as the reference standard. Results: Total 42 stations had metastatic lymph nodes and 136 stations had benign lymph nodes. The differences between metastatic and benign lymph nodes in the visual qualitative and semiquantitative analyses of 99mTc-3PRGD2 SPECT/CT and 18F-FDG PET/CT were statistically significant (all P < 0.001). The area under the receiver operating characteristic curve (AUC) in the semi-quantitative analysis of 99mTc-3PRGD2 SPECT/CT was 0.908 (95% confidence interval [CI], 0.851–0.966), and the sensitivity, specificity, positive predictive value, and negative predictive value were 0.86 (36/42), 0.88 (120/136), 0.69 (36/52), and 0.95 (120/126), respectively. Among the 26 patients (including two patients each with two lung tumors), 15 had pathologically confirmed lymph node metastasis. The difference between primary lung lesions in patients with and without lymph node metastasis was statistically significant only in the semi-quantitative analysis of 99mTc-3PRGD2 SPECT/CT (P = 0.007), with an AUC of 0.807 (95% CI, 0.641–0.974). Conclusion: 99mTc-3PRGD2 SPECT/CT imaging may notably perform in the direct diagnosis of lymph node metastasis of primary malignant lung tumors and indirectly predict the presence of lymph node metastasis through uptake in the primary lesions.

        Mechanical Performance of Vegetation-Growing Concrete Reinforced by Palm Fibers under Triaxial Compression

        Fibers have been widely applied to improve the mechanical properties of natural soil and traditional cemented soils, but rarely investigated in substrates for slope eco-protection. Vegetation-growing concrete (VC) substrate is a kind of ecological cemented soil and has very wide popularization and application prospects. In this study, unconsolidated-undrained (UU) triaxial compression tests were conducted on the control specimens of VC substrate containing three cement contents and the palm fiber reinforced specimens manufactured by adding four fiber contents. The control specimens were shown to possess increasing yield strength, ultimate strength, yield strain and shear modulus with cement content, which resulted from the cemented texture improved by the hydrate products as recognized previously. Instead of inextensible reinforcement provided by cement, the palm fibers were found to provide extensible reinforcement, through which the obvious softening behavior of VC substrate could transfer to elasto-plasticity. Attributed to the unique ductility, the fibers added could bring with considerable increase in ductility of reinforced substrate, except for additional increases in yield strength, ultimate strength and yield strain.
