        The Effect of HIF-1α siRNA on Growth and Chemosensitivity of Mia-paca Cell Line

        Purpose: Hypoxia-inducible factor-1α (HIF-1α) primarily mediates the hypoxic response. HIF-1α induction by various stimuli contributes to cell proliferation and survival. To investigate the effect of HIF-1α, we used small interfering RNA (siRNA), and expected that cell apoptosis and sensitivity to chemotherapeutic drug increase, when we blocked the HIF-1α gene. Thus we performed in vitro and in vivo experiment to clarify the effect of hypoxia-inducible factor-1α on tumor growth. After selection with geneticin, two new cell lines were made, confirmed via immunoblotting. After treating with gemcitabine, each cell line was assayed to confirm the effect of HIF-1α siRNA using the cell proliferation assay and capase-3 assay. And then in vivo study was performed using female athymic nude mice. After subcutaneously injecting each new cell lines, intraperitoneal gemicitabine chemotherapy was performed for 3 weeks. During that period, we analyzed the difference of tumor growth rate. Results: The tumor growth of HIF-1α siRNA-transfected group was slower than that of the control group both in vitro and in vivo experiment. Conclusion: The suppression of HIF-1α results in decrease of cell proliferation and increase of chemosensitivity of pancreatic cancer cell line.       • Linkage among Local Residents around the Library

        The purpose of the paper is to introduce friends of the libraries in the world, activities and current status of Seodaemun Friends of the Library. The friends groups began from public and national libraries in Germany and France a century ago or from early 1900s. Currently, the groups have been distributed to several countries including the United States with 3,000 groups, the UK, Australia, Canada, New Zealand, Germany, Spain, Iceland, Japan, Indonesia and Chile. Seodaemun Friends of the Library was voluntarily founded in 2011 by the director Lily Yang for the purpose of assisting local public library, which is called the Seodamun Library, by local residents. Currently, there are 380 members and each member donates 2,000 KRW a month to donate books, replace old shelves, provide administrative support to install self-supporting signboards, improve welfare for library officials through supporting book retrieval from Hongje Station and stimulate the budget allocation. Also, the friends group formed a gathering to protect a local bookshop with the history of 53 years, showing the power of local solidarity and reading, provided lectures through members’ network and helped revitalizing the library. The group linked local buses and shops to increase the library availability through leaflets for the library and led local buses passing the library to stop for local residents to conveniently visit the library, developing the local culture by the connection of the library and local people.

        Isovitexin Protects Mice from Methicillin- Resistant Staphylococcus aureus-Induced Pneumonia by Targeting Sortase A

        The rise of methicillin-resistant Staphylococcus aureus (MRSA) has resulted in significant morbidity and mortality, and clinical treatment of MRSA infections has become extremely difficult. Sortase A (SrtA), a virulence determinant that anchors numerous virulence-related proteins to the cell wall, is a prime druggable target against S. aureus infection due to its crucial role in the pathogenicity of S. aureus. Here, we demonstrate that isovitexin, an active ingredient derived from a variety of traditional Chinese medicines, can reversibly inhibit SrtA activity in vitro with a low dose (IC<sub>50</sub>=24.72 μg/ml). Fluorescence quenching and molecular simulations proved the interaction between isovitexin and SrtA. Subsequent point mutation experiments further confirmed that the critical amino acid positions for SrtA binding to isovitexin were Ala-92, Ile-182, and Trp-197. In addition, isovitexin treatment dramatically reduced S. aureus invasion of A549 cells. This study shows that treatment with isovitexin could alleviate pathological injury and prolong the life span of mice in an S. aureus pneumonia model. According to our research, isovitexin represents a promising lead molecule for the creation of anti-S. aureus medicines or adjuncts.

        Morphology Controlled Synthesis of Octahedral Covalent Imine Frameworks Through Acid Modulated Aldehyde-Amine Polycondensation

        The past decades have witnessed significant advance in the synthesis of covalent organic frameworks (COFs), however, their formation and morphology revolution mechanism have been rarely reported. Here, through an acid modulated dynamic covalent chemistry approach, pure and uniform micro-octahedral covalent imine frameworks were obtained for the first time. Formation mechanism based on the stacking of initially formed two-dimensional nanoplates followed by surface smoothing enabled by the dynamic nature of imine bonding was proposed. Furthermore, we revealed for the first time that nonstoichiometric method can be applied to the synthesis of covalent organic frameworks. Thus we provide novel strategy for the morphology control COFs which will surely facilitate their application in energy-related area.

        Transparent Lamellar Porous Material and Its Greatly Reduced Dielectric Constant

        One trend for low dielectric materials is to reach low dielectric constant values at as low porosity as possible. In this work, a lamellar porous material was prepared by spin-coating of poly(vinyl alcohol) (PVA)/manganese dioxide (MnO2) nanosheet composited film, followed by cross-linking of PVA and removing nanosheets. FTIR, XRD and TGA measurement results demonstrate that the templates were almost completely removed. SEM image shows that the etched PVA film has a lamellar porous structure. Dielectric test results indicate that at the porosity of only 17.5%, the dielectric constant of porous PVA is reduced to approximately half that of neat cross-linked PVA. The serial model shows a good consistence with experimental dielectric constant value. This explains well the high efficiency of lamellar porous structure in reducing dielectric constant.

        Abnormal Vacuolization of the Tapetum During the Tetrad Stage is Associated with Male Sterility in the Recessive Genic Male Sterile Brassica napus L. Line 9012A

        In the recessive genic male sterile line 9012A of Brassica napus, pollen development is affected during the tetrad stage. According to the light and electron microscopy analysis of tapetal cells and tetrads, the sterile tapetal cells swelled with expanded vacuoles at the early tetrad stage and finally filled the center of the locules where a majority of tetrads encased with the thick callose wall collapsed and degraded. We suggested that an absence of callase, which is a wall-degrading enzyme stored in the vacuoles of tapetal cells before secretion, resulted in the failure of tetrad separation. Moreover, transmission electron microscopy analysis showed that the secretory tapetal cells were not observed in sterile anthers, which indicated that the transition of the tapetum from the parietal type to the secretory type was probably aberrant. In plants, degeneration of the tapetum is thought to be the result of programmed cell death (PCD). PCD of tapetal cells was investigated by terminal deoxynucleotidyl transferasemediated dUTP nick-end labeling assay and signals indicative of deoxyribonucleic acid fragmentation were detected much earlier in sterile anther than in fertile anther. This suggests that tapetal breakdown does not occur by the normal procession of PCD and might be following an alternative mechanism of unscheduled apoptosis in line 9012A. This research supports the hypothesis that premature PCD is associated with male sterility in B. napus.

        Filled Skutterudites: from Single to Multiple Filling

        This paper shortly reviews our recent work on filled skutterudites, which are considered to be one of the most promising thermoelectric (TE) materials due to their excellent power factors and relatively low thermal conductivities. The filled skutterudite system also provides a platform for studying void filling physics/chemistry in compounds with intrinsic lattice voids. By using ab initio calculations and thermodynamic analysis, our group has made progresses in understanding the filling fraction limit (FFL) for single fillers in CoSb3, and ultra-high FFLs in a few alkali-metal-filled CoSb3 have been predicted and then been confirmed experimentally. FFLs in multiple-element-filled CoSb3 are also investigated and anonymous filling behavior is found in a few specific systems. The calculated and measured FFLs, in both single and multiple-filled CoSb3 systems, show good accordance so far. The thermal transport properties can be understood qualitatively by a phonon resonance scattering model, and it seems that a scaling rule may exist between the lattice thermal resistivity and the resonance frequency of filler atoms in filled system. Even though a few things become clear now, there are still many unsolved issues that call for further work.

        Wind farm resonance characteristics analysis based on harmonic impedance measurement method

        Recently, subsynchronous oscillation (SSO) has been captured in direct-drive permanent magnetic synchronous generator (PMSG) based wind farms in many areas without series compensation in nearby power grids. However, there are fewer reports related to this new type of SSO. In addition, an analysis of the new SSO based on existing methods is computationally cumbersome, and the control effect of the converter was generally simplified in previous studies, which lead to serious errors. This paper proposes a method for analyzing the resonance characteristics of wind farms based on impedance measurement to simplify calculations and improve accuracy. The correctness and effectiveness of the proposed method are validated through the IEEE first benchmark model. A time-domain simulation model of a direct-drive PMSG is established and analyzed by harmonic impedance measurement. The results show that SSO exists when direct-drive PMSGs are connected to a weak AC system. The converter of the direct-drive PMSG exhibits a small negative value at the oscillation mode frequency. This means that the converter behaves like a capacitor with negative impedance. The capacitive impedance of the converter forms a resonant loop with the inductance of the AC grid and causes unstable SSO.

        Serial Frozen Fecal Microbiota Transplantation in the Treatment of Chronic Intestinal Pseudo-obstruction: A Preliminary Study

        Background/Aims Chronic intestinal pseudo-obstruction (CIPO) is a serious, life-threatening motility disorder that is often related to bacterial overgrowth. Fecal microbiota transplantation (FMT) results in restoration of the normal intestinal microbial community structure. We investigated the efficacy of FMT in the treatment of CIPO patients. Methods Nine patients (age 18-53 years) with CIPO were enrolled in this prospective, open-label study. Patients received FMT for 6 consecutive days through nasojejunal (NJ) tubes and were followed up for 8 weeks after treatment. We evaluated the rate of clinical improvement and remission, feeding tolerance of enteral nutrition, and CT imaging scores of intestinal obstructions. Lactulose hydrogen breath tests were performed before FMT and 8 weeks after FMT to evaluate for the presence small intestinal bacterial overgrowth (SIBO). Results FMT significantly alleviated bloating symptoms, and symptoms of pain were relieved 2 weeks after FMT. Enteral nutrition administered through a NJ tube after FMT was well-tolerated by 66.7% (6/9) of patients. CT scores of intestinal obstructions were significantly reduced after FMT (P = 0.014). SIBO was eliminated in 71.0% (5/7) of patients. Conclusions This pilot study demonstrated the safety of using FMT. FMT may relieve symptoms in selected patients with CIPO. FMT may also improve patient tolerance of enteral nutrition delivered via a NJ tube. (J Neurogastroenterol Motil 2017;23:289-297)

        Impact of ejaculation upon effect of acupuncture on chronic prostatitis/chronic pelvic pain syndrome: Secondary analysis of a randomized controlled trial

        Background: Acupuncture can improve chronic prostatitis/chronic pelvic pain syndrome (CP/CPPS). Ejaculation frequencies might impact the conditions of CP/CPPS. The present study aimed to explore the impact of different ejaculation frequencies on the effect of acupuncture among men with CP/CPPS. Methods: This was a secondary analysis of the data from a multicenter, randomized, clinical trial. Eligible participants were patients with moderate to severe CP/CPPS, who had taken 8-week acupuncture treatment, and followed until week 32. Participants fell into the category of 0–3, 4–7, or at least 8 according to their monthly ejaculation frequencies reported at baseline. The primary outcome was the proportion of responders, defined as men who reported at least 6 points reduction from baseline in the National Institute of Health-Chronic Prostatitis Symptom Index (NIHsingle bondCPSI) total score at weeks 8 and 32. Results: 214 participants were included in this secondary analysis, of whom 42 reported a monthly ejaculation frequency of 0–3, 89 reported a frequency of 4–7, and 83 reported a frequency of at least 8. At week 8, 52.20% participants with an ejaculation frequency of 0–3 responded to the acupuncture treatment, 65.38% participants with a frequency of 4–7 responded, and 63.09% participants with a frequency of at least 8 responded. At week 32, 56.14%, 59.57%, and 68.36% participants responded in the three groups, respectively. No significant differences were observed between three groups (all P>0.05). Conclusion: Acupuncture can improve symptoms of CP/CPPS, regardless of ejaculation frequencies. Ejaculation frequencies may not affect the efficacy of acupuncture on CP/CPPS among Chinese men.
