        Biphasic effects of TGFβ1 on BMP9-induced osteogenic differentiation of mesenchymal stem cells

        We have found that the previously uncharacterized bone morphogenetic protein-9 (BMP9) is one of the most osteogenic factors. However, it is unclear if BMP9 cross-talks with TGFβ1 during osteogenic differentiation. Using the recombinant BMP9 adenovirus, we find that low concentration of rhTGFβ1 synergistically induces alkaline phosphatase activity in BMP9-transduced C3H10T1/2 cells and produces more pronounced matrix mineralization. However, higher concentrations of TGFβ1 inhibit BMP9-induced osteogenic activity. Real-time PCR and Western blotting indicate that BMP9 in combination with low dose of TGFβ1 potentiates the expression of later osteogenic markers osteopontin, osteocalcin and collagen type 1 (COL1a2), while higher concentrations of TGFβ1 decrease the expression of osteopontin and osteocalcin but not COL1a2. Cell cycle analysis reveals that TGFβ1 inhibits C3H10T1/2 proliferation in BMP9-induced osteogenesis and restricts the cells in G0/G1 phase. Our findings strongly suggest that TGFβ1 may exert a biphasic effect on BMP9-induced osteogenic differentiation of mesenchymal stem cells. [BMB Reports 2012; 45(9): 509-514]

        LncRNA TMPO-AS1 promotes esophageal squamous cell carcinoma progression by forming biomolecular condensates with FUS and p300 to regulate TMPO transcription

        Esophageal squamous cell carcinoma (ESCC) is one of the most life- and health-threatening malignant diseases worldwide, especially in China. Long noncoding RNAs (lncRNAs) have emerged as important regulators of tumorigenesis and tumor progression. However, the roles and mechanisms of lncRNAs in ESCC require further exploration. Here, in combination with a small interfering RNA (siRNA) library targeting specific lncRNAs, we performed MTS and Transwell assays to screen functional lncRNAs that were overexpressed in ESCC. TMPO-AS1 expression was significantly upregulated in ESCC tumor samples, with higher TMPOAS1 expression positively correlated with shorter overall survival times. In vitro and in vivo functional experiments revealed that TMPO-AS1 promotes the proliferation and metastasis of ESCC cells. Mechanistically, TMPO-AS1 bound to fused in sarcoma (FUS) and recruited p300 to the TMPO promoter, forming biomolecular condensates in situ to activate TMPO transcription in cis by increasing the acetylation of histone H3 lysine 27 (H3K27ac). Targeting TMPO-AS1 led to impaired ESCC tumor growth in a patient-derived xenograft (PDX) model. We found that TMPO-AS1 is required for cell proliferation and metastasis in ESCC by promoting the expression of TMPO, and both TMPO-AS1 and TMPO might be potential biomarkers and therapeutic targets in ESCC.

      • Depiction of concrete structures with seismic separation under faraway fault earthquakes

        One of the most suitable methods in structural design is seismic separator. Lead-Rubber Bearing (LRB) is one of the most well-known separation systems which can be used in different types of structures. This system mitigates the earthquake acceleration prior to transferring to the structure efficiently. However, the performance of this system in concrete structures with different heights have not been evaluated thoroughly yet. This paper aims to evaluate the performance of LRB separation system in concrete structures with different heights. For this purpose, three, 16, and 23 story concrete structures are equipped by LRB and exposed to a far-field earthquake. Next, a time history analysis is conducted on each of the structures. Finally, the performance of the concrete structures is compared with each other in the term of their response to the earthquakes and the formation of plastic hinges. The results of the paper show that the rate of change in acceleration response and the ratio of drift along the height of 8 and 23 stories concrete structures are more than those of the 16-stories, and the use of LRB reduces the formation of plastic joints.

        Adaptive Synchronization on Uncertain Dynamics of High-Order Nonlinear Multi-Agent Systems with Partition of Unity Approach

        An adaptive synchronization of uncertain chaotic system is presented using partition of unity method. First, the uncertainties of chaotic systems are approximate by the linear combination of partition of unity. Subsequently, the sliding mode adaptive controllers are proposed for synchronization of the uncertain chaotic systems. The proposed approach offers a systematic design procedure for adaptive synchronization of a class of uncertain chaotic system in the chaos research literature. We also il-lustrate examples of chaotic systems such as Chen and Lorenz chaotic systems to show the effective-ness of the approach.

        A generalized algorithm for the study of bilinear vibrations of cracked structures

        Structural cracks may cause variations in structural stiffness and thus produce bilinear vibrations to structures. This study examines the dynamic behavior of structures with breathing cracks. A generalized algorithm based on the finite element method and bilinear theory was developed to study the influence of a breathing crack on the vibration characteristic. All the formulae derived in the time domain were applied to estimate the period of the overall bilinear motion cycle, and the contact effect was considered in the calculations by introducing the penetration of the crack surface. Changes in the dynamic characteristics of cracked structures are investigated by assessing the variation of natural frequencies under different crack status in either the open or closed modes. Results in estimation with vibrational behavior variation are significant compared with the experimental results available in the literature as well as other numerical calculations.

        Performing the Political in Lust, Caution

        Examining three dramatic scenes of performance in Ang Lee’s 2007 film Lust, Caution, based on Eileen Chang’s short story of the same name, this essay proposes to read Lee’s film as an epilogue to a long-running narrative highlighting the intersection of performance and politics in twenty-century China. It argues, through a close examination of Lee’s creative use of such elements as popular music, political theater, and leftist cinema from the 1930s, that nationalism and revolution staged an intriguing comeback in Lust, Caution, intensified, rather than negated, by its intricate intertwinement with sexuality and ethics. In particular, the two popular film songs from the 1930s with lyrics penned by Tian Han quoted in the film, and their fascinating afterlives in contemporary popular culture represented by Leehom Wang, highlight the power of performance in shaping a complex range of gender, ethical, and political identities. The intersection of performance, politics, and popularity works magic throughout the film. It enables Lee’s film to go beyond Chang’s story in reinvigorating the political through the performative and the popular.

        A Cosmopolitan Utopia on Poetic Form: A Reading of Gwendolyn Brooks`s “The Anniad”

        “The Anniad,” a long poem in Gwendolyn Brooks’s Pulitzer prize-winning Annie Allen, is widely claimed as a high modernist poetic experiment. Annie Allen (1949), Gwendolyn Brooks’s second collection of poems, won the poet a great fame as the first African American winner of Pulitzer Award for Poetry while throwing her into an awkward controversy over her widely claimed “high-modernism.” The work, composed of three parts, is about Annie, an African American girl, growing into womanhood and growing from an egotistic romantic to a realistic idealist. My position in the analysis of the poem is based upon my disagreement with those critics who believe this poem is meant to cater to the white aesthetic taste while excluding the black readers with its high modernism or is an endeavor to subvert or deconstruct the European masters by grafting the black cultural elements and experience on the European tradition. My contention is that Gwendolyn Brooks tries to make poetry as represented by “The Alliad,” a formal and spiritual negotiation of the African American with the white modernism and the European classics. By integrating classical European poetic forms, white modernist techniques and black vernacular, Brooks develops her integrationist dream into a dream of cosmopolitanism.

        Experimental studies on elastic cooling and pyromagnetic effect of polyvinyl chloride sheets with defects

        Polyvinyl Chloride (PVC) sheets were made into samples with precast defects for uniaxial tensile test. The tests are carried out under room temperature with conditions of same displacement rate but different defect sizes or of same defect but different displacement rates. The local nonuniform temperature field on whole deformation area of specimen is recorded with a thermal infrared imager and the whole coupling magnetic field with thermal changes in experiments is detected and measured by a self-developed sensors system. The experimental results show that, in a complete tensile test process of PVC samples, the temperature reduction phenomenon emerges firstly in its elastic deformation stage (areas) that temperature of specimen is cooler than room temperature. And then in viscoplastic deformation period (areas), the temperature increases sharply to be obviously higher than room temperature due to the thermo-mechanical coupling effection of tensile load and viscoplastic deformation heat. These thermal variations lead a coupling pyromagnetic effect occur and the effect intensity is dependent strongly on the strain rate and/or the size of defects. The temperature prejudgment conditions for materials yield are preliminary discussed based on this effect.

        IRE1α protects against osteoarthritis by regulating progranulin-dependent XBP1 splicing and collagen homeostasis

        Osteoarthritis (OA) is a full-joint, multifactorial, degenerative and inflammatory disease that seriously affects the quality of life of patients due to its disabling and pain-causing properties. ER stress has been reported to be closely related to the progression of OA. The inositol-requiring enzyme 1α/X-box-binding protein-1 spliced (IRE1α/XBP1s) pathway, which is highly expressed in the chondrocytes of OA patients, promotes the degradation and refolding of abnormal proteins during ER stress and maintains the stability of the ER environment of chondrocytes, but its function and the underlying mechanisms of how it contributes to the progression of OA remain unclear. This study investigates the role of IRE1α/ERN1 in OA. Specific deficiency of ERN1 in chondrocytes spontaneously resulted in OA-like cartilage destruction and accelerated OA progression in a surgically induced arthritis model. Local delivery of AdERN1 relieved degradation of the cartilage matrix and prevented OA development in an ACLT-mediated model. Mechanistically, progranulin (PGRN), an intracellular chaperone, binds to IRE1α, promoting its phosphorylation and splicing of XBP1u to generate XBP1s. XBP1s protects articular cartilage through TNF-α/ERK1/2 signaling and further maintains collagen homeostasis by regulating type II collagen expression. The chondroprotective effect of IRE1α/ERN1 is dependent on PGRN and XBP1s splicing. ERN1 deficiency accelerated cartilage degeneration in OA by reducing PGRN expression and XBP1s splicing, subsequently decreasing collagen II expression and triggering collagen structural abnormalities and an imbalance in collagen homeostasis. This study provides new insights into OA pathogenesis and the UPR and suggests that IRE1α/ERN1 may serve as a potential target for the treatment of joint degenerative diseases, including OA.

      • Introduction to The Technical Verification Site for Intelligent Manufacturing Taiwan

        Intelligent manufacturing has become a key technology for survival for industry due to the trends on labor force shortage and massive customization in global and domestic market environment. However, most domestic manufacturing companies are SMEs which lack of resource to conduct R&D activities or build intelligent factory. Therefore, an open demo site would be an important infrastructure to promote intelligent manufacturing as well as a testbed for advanced research with international partners on cloud computing, 5G, AI and digital twins. Technologies built in the site includes: (1) Domestic machines: all machine tools, robots, conveyors, storage systems, AGVs are made in Taiwan. More than 60% of these machines are equipped with CNC built by ITRI. (2) Software for intelligent factory: Production management system, IoT, process diagnostics and optimization software are developed by ITRI. These software enable flexible mixed production of parts across various industry sectors. (3) Digital twins and extended reality: Digital twins for processes, machines are developed and deployed in the cloud computing platform with corresponding interaction interfaces constructed based on immersive technology to achieve high fidelity user experience. Advanced research projects based on the infrastructure of the site includes: (1) Intelligent machinery cloud platform with the objective to build unified IoT software and cloud computing platform for Taiwan machinery industry (2) 5G private network and use cases on immersive interaction for process diagnostics and industrial cyber-physical control International research projects based on the site includes: (1) The Canadian Network for Research and Innovation in Machining Technology, (CANRIMT) which is an international project focusing on machining technology. (2) The UMATI information model, initiated by VDW to define unified information model for various machinery based on OPCUA. (3) The 5G for industrial connectivity (5GConni) with the aim to develop use cases to leverage available 5G private network solutions During the construction and promotion of the intelligent manufacturing technology verification site, many industrial partners start to invest on intelligent manufacturing. Therefore we believe that the verification site will continue to act as the key infrastructure for intelligent manufacturing and provide latest solution for the industry.
