농업생산기반시설물 중 양수장은 전국 7,052개로 그 중 3479개는 20년 이상 운영되고 있어 노후화 현상에 대한 우려가 있다. 노후화된 양수장을 재구축 및 신축하기에는 과다한 비용이 소요된다. 따라서 기존 시설의 성능을 정량적으로 판단하여 유지관리 할 필요성이 있다. 농업생산기반 정비사업계획 설계기준에 의하면 양수장의 유지관리는 펌프의 운전목적을 달성할 수 있도록 각 설비 기능들을 정상으로 유지하는 것을 목적으로 한다. 또한 양수장의 성능을 평가하기 위한 항목으로 토목, 전기, 기계, 건축으로 분류하고 있다. 구조물은 양수장의 펌프를 외부환경으로부터 보호해야하기 때문에 노후화에 대한 판단이 필요하지만 펌프관리에 비해 상대적으로 구조물에 대한 운영관리 체계는 구체적이지 못한 실정이다. 이를 보완하기 위해 VE(Value Engineering)를 도입하고자 한다. VE는 가치를 얻기 위한 목적으로 수행되는 프로젝트의 기능분석을 통해 대안창출을 할 수 있는 체계적인 프로세스를 의미한다. 이를 통해 양수장 유지관리 항목에 대해 기능은 유지하면서 역할을 세분화하고 중요도를 설정해 성능을 정량적 평가 할 수 있다. 따라서 본 연구에서는 VE를 통해 양수장에 대한 기능과 성능을 정량적으로 평가하고 기존의 유지관리 방안에 대한 개선사항을 제시하고자 한다.

This study summarizes the recent cutting-edge approaches for dentin regeneration that still do not offer adequate solutions. Tertiary dentin is formed when odontoblasts are directly affected by various stimuli. Recent preclinical studies have reported that stimulation of the Wnt/β-catenin signaling pathway could facilitate the formation of reparative dentin and thereby aid in the structural and functional development of the tertiary dentin. A range of signaling pathways, including the Wnt/β-catenin pathway, is activated when dental tissues are damaged and the pulp is exposed. The application of small molecules for dentin regeneration has been suggested as a drug repositioning approach. This study reviews the role of Wnt signaling in tooth formation, particularly dentin formation and dentin regeneration. In addition, the application of the drug repositioning strategy to facilitate the development of new drugs for dentin regeneration has been discussed in this study.

<P>The bone morphogenetic protein (BMP) ligand Glass bottom boat (Gbb) acts as a retrograde growth signal at the Drosophila neuromuscular junction (NMJ). Endocytic regulation of presynaptic BMP receptors has been proposed to attenuate retrograde BMP signaling. However, it remains unknown whether the Gbb signal is also regulated by postsynaptic mechanisms. Here, we provide evidence that Drosophila Cdc42-interacting protein 4 (dCIP4) functions postsynaptically to inhibit synaptic growth. dCIP4 is localized postsynaptically at NMJs. dcip4 mutations lead to synaptic overgrowth and increased presynaptic phosphorylated mothers against decapentaplegic (Mad) levels, and these defects are rescued by muscle-specific expression of dCIP4. Biochemical and genetic analyses demonstrate that dCIP4 acts downstream of Cdc42 to activate the postsynaptic Wsp-Arp2/3 pathway. We also show that BMP signaling is necessary for synaptic overgrowth in larvae lacking postsynaptic dcip4 or wsp. Finally, dCIP4 and Wsp inhibit Gbb secretion. Thus, we propose that dCIP4 restrains synaptic growth by inhibiting postsynaptic Gbb secretion through the Wsp-Arp2/3 pathway.</P>

Background: Plasma cell myeloma (PCM) is caused by immune dysregulation. We evaluated the expression of immune checkpoint programmed cell death protein-1 (PD-1) on T cell subsets in PCM patients according to disease course and cytogenetic abnormalities. This study aimed to find a target group suitable for therapeutic use of PD-1 blockade in PCM. Methods: A total of 188 bone marrow (BM) samples from 166 PCM patients and 32 controls were prospectively collected between May 2016 and May 2017. PD-1 expression on BM T cell subsets was measured using flow cytometry. Results: At diagnosis, the median PD-1 expression on CD4+ T cells was 24.6%, which did not significantly differ from that in controls. After stem cell transplantation, PD-1 expression on CD4+ T cells was higher than that at diagnosis (P<0.001), regardless of residual disease. PD-1 expression on CD4+ T cells in patients with residual disease after chemotherapy was significantly higher than that at diagnosis (P=0.001) and after complete remission following chemotherapy (P=0.044). PD-1 expression on CD8+ T cells was higher in PCM patients with cytogenetic abnormalities, including monosomy 13, 1q gain, complex karyotype, and hypodiploidy. Conclusions: PD-1 blockade might have therapeutic potential in refractory PCM patients after chemotherapy, especially in those with high- or intermediate-risk cytogenetic abnormalities.

Understanding the sophisticated imprinting pattern is the first challenge for deciphering the molecular mechanism of an imprinted gene. However, the determination of imprinted genes, and their methylation status in pigs, remains inefficient at present. Herein, for the first time, we have made an attempt to illustrate the complex imprinting pattern of porcine GNAS complex locus by an efficient strategy using RNA-seq analysis of parthenogenetic fetuses containing no paternal allele, but two maternal alleles. RNA-seq results showed that Nespas, Gnasxl, and Exon 1A genes expressed at extremely lower levels (p<0.05) in parthenogenetic fetuses compared to the normally fertilized fetuses, suggest these three genes are paternally-expressed. Whereas, the expression levels of Gnas were similar in both groups, suggesting biallelic expression of Gnas. The parental-specific expression of these genes was further confirmed by quantitative real-time PCR. Bisulfite sequencing revealed that the maternally derived Nespas promoter was methylated, but the paternal chromosome was not methylated. Our finding provided full-scale imprinting information on porcine GNAS locus for the first time. Furthermore, our study is useful for identify novel imprinted genes, confirm the previously known imprinted genes, and determine imprinting pattern efficiently.

Axon extension and guidance require a coordinated assembly of F-actin and microtubules and regulated translation in the growth cone. The molecular basis of how the translation of mRNAs encoding guidance proteins could be closely tied to the pace of cytoskeletal assembly is poorly understood. Previous studies showed that Short Stop(Shot) mediates direct interactions between F-actin and microtubules required for motor and sensory axon extension in the Drosophila embryo. Here, we provide biochemical and genetic evidence that Shot functions in neurons together with a novel translation inhibitor, Krasavietz(Kra), to steer longitudinally directed CNS axons away from the midline. Kra can bind directly to the C-terminal domain of Shot. shot and kra mutants have weak robo-like phenotypes and synergistically affect midline avoidance. Mutations in shot and kra dominantly enhance the frequency of midline crossovers in embryos heterozygous for slit. We demonstrate that Kra interacts with the translation machinery and inhibits translation in vitro. These data suggest that Shot-Kra interaction controls the translation of mRNAs encoding proteins necessary for the Slit/Robo response of the growth cone. We propose previously unknown role of Shot as a direct physical link between translational regulation and cytoskeleton reorganization.

Recently, a number of adulterated products, which are advertised as hair-growth enhancer have been emerged among those who suffer hair loss disease. For continuous control of illegal products, in this study, a rapid and sensitive method for simultaneous screening of 12 compounds that enhance hair-growth was established to protect public health by ultrahigh-performance liquid chromatography coupled to quadrupole-orbitrap mass spectrometry (UHPLC-Q-Orbitrap-MS). Fragmentation pathways of them were proposed based on $MS^2$ spectral data obtained using the established method. In this analysis, the LODs and LOQs ranged from 0.05 to 50 ng/mL and from 0.17 to 167 ng/mL, respectively. The square of the linear correlation coefficient ($R^2$) was determined as more than 0.995. The intra- and inter-assay accuracies were respective 88-112 % and 88-115 %. Their precision values were measured within 5 % (intra-day) and 10 % (inter-day). Mean recoveries of target compounds in adulterated products ranged from 84 to 115%. The relative standard deviation of stability was less than 12 % at $4^{\circ}C$ for 48 h. The method was employed to screen 14 dietary supplements advertised to be effective for the treatment of hair loss. Some of the products (~21 %) were proven to contain synthetic drugs that promote hair growth such as triaminodil, minoxidil, and finasteride.
