        기계경비업무 감독명령 시행의 정책과제

        이 연구에서는 '기계경비 112신고 기준'인 감독명령 제2013-1호를 분석하고 정책적 과제를 제시하였다. 이 감독명령은 기계경비업체의 출동대응의무 등 독자적인 범죄예방 능력 확보와 통계기반 구축을 위해 확인신고를 원칙으로 하고 긴급신고의 대상을 특정하는 선별신고 제도의 시행을 규정하고 있다. 선별신고제도의 목적을 달성하기 위한 정책적 과제로 첫째, 통계기반의 구축을 위한 오경보와 오신고, 비상버튼과 감지신호에 대한 용어의 정립과 기계경비원을 대체할 적절한 용어의 개발이 필요하다. 둘째, 기계경비업체는 이 감독명령의 112신고 기준을 준수하기 위한 대응체제를 구축하여야 한다. 셋째, 경찰은 선별신고제도의 긴급신고 대상 설정에 대한 재검토가 있어야 한다. 넷째, 이 감독명령을 엄정하게 시행하기 위한 행정규제와 감독의 강화가 요구된다. In this research, The Police Order No. 2013-1, which is 'Electronic Security 112 Report Standards,' was analyzed and the relevant policy tasks were presented. The policy tasks to achieve the purpose of the selective report system are as follows. First, in order to construct the statistics base, the establishment of the terms related to false alarm and false call, emergency button, and sensing signal and the development of the appropriate term to replace the electronic security guard are needed. Second, the electronic security companies should build the response system to abide by the 112 report standards of the police order. Third, the police should reexamine the establishment of the emergency report objects of the selective report system. Fourth, the reinforcement of the administrative regulation and supervision to enforce the police order strictly is required.

        On the size effect of interfacial fracture between concrete and fiber reinforced polymer

        <P><B>Abstract</B></P> <P>In this study, the size effect of the nominal interfacial strength between concrete and fiber reinforced polymer (FRP) is investigated by integrating fracture tests and computational simulations. Three-point bending fracture tests are performed with geometrically similar specimens to demonstrate the size effect, and to measure fracture parameters for mode-I. Based on the measured fracture parameters, the crack-growth behavior is predicted using the finite-element-based cohesive zone model. Computational results predict accurately the experimental results of the load versus crack mouth opening displacement (CMOD) curves. Furthermore, both the computational and experimental results illustrate that the nominal interfacial strength decreases with increases in the specimen size, i.e., the size effect of the interfacial fracture. Because of the size effect, fracture parameters, such as the fracture energy, should be measured and evaluated to predict the interfacial fracture between concrete and FRP.</P>

        기계경비업무 감독명령 시행의 정책과제

        Antimicrobial photodynamic therapy with Ligularia fischeri against methicillin-resistant Staphylococcus aureus infection in Caenorhabditis elegans model

        The high prevalence of methicillin-resistant Staphylococcus aureus (MRSA) infection threatens the effectiveness of current clinical settings. Antimicrobial photodynamic therapy (APDT) is a promising alternative to antibiotics for treating infections due to its low resistance. This study aimed to evaluate the antibacterial properties of APDT with L. fischeri extract (LFE) against MRSA and various skin and oral pathogens in vitro and its photopharmaceutical actions in Caenorhabditis elegans. The antimicrobial activities of APDT with LFE against pathogens were evaluated using plate counting method. The chemical profile was characterized using high-performance liquid chromatography and spectrophotometry. The growth rate assay, lifespan assay, and bacterial attachment on worms were performed to assess the therapeutics effects in C. elegans. The swab method was used for the detection of pathogens on the micropig skin surface. The APDT treatment with L. fischeri extract (LFE, 20 µg/mL) and red light (intensity of 120 W/m2) reduced 4.3–4.9 log (colony forming unit/mL) of Staphylococcus aureus, MRSA, Cutibacterium acnes, Streptococcus mutans; and 2.4 log (CFU/mL) of Candida albicans. Chemical analysis revealed that LFE enriched three active photosensitizers. APDT reduced bacterial populations on worms, recovered growth retardation, and improved lifespan in MRSA-infected C. elegans without causing severe side effects. The surface eradication of MRSA after exposure to LFE with red light was demonstrated on micropig skin. These findings highlight the significance of L. fischeri as a natural resource for the safe phototreatment of MRSA infection in the biomedical and cosmeceutical industries.

        Caenorhabditis elegans as a powerful tool in natural product bioactivity research

        In addition to synthetic and semisynthetic compounds, natural products have received much attention as prolific sources of lead compounds with therapeutic effects on various diseases. In the process of screening the bioactivities of potential candidates, an in vivo assay is very important for providing meaningful insight into the efficacy, adverse effects, and modes of action that are relevant to humans. Among the many experimental models, Caenorhabditis elegans is particularly efficient due to its advantages in morphology, behavior, and genetic aspects. This review summarizes some basic and useful techniques commonly used in screening the bioactivities of natural products. Recent studies of naturally occurring extracts as well as bioactive compounds in various areas, namely, anti-aging, anti-neurodegeneration, anti-obesity, anti-infection, and gut health, are reviewed as examples of the applicability of the C. elegans model. Technological developments that incorporate C. elegans in other fields, such as instrumental analysis and emerging methods, are also discussed in this paper.

        <P><B>Objectives</B></P><P>To develop a high-throughput screening system to measure the conversion of testosterone to dihydrotestosterone (DHT) in cultured human prostate cancer cells using turbulent flow chromatography liquid chromatography-triple quadrupole mass spectrometry (TFC-LC-TQMS).</P><P><B>Results</B></P><P>After optimizing the cell reaction system, this method demonstrated a screening capability of 103 samples, including 78 single compounds and 25 extracts, in less than 12 h without manual sample preparation. Consequently, fucoxanthin, phenethyl caffeate, and <I>Curcuma longa</I> L. extract were validated as bioactive chemicals that inhibited DHT production in cultured DU145 cells. In addition, naringenin boosted DHT production in DU145 cells.</P><P><B>Conclusion</B></P><P>The method can facilitate the discovery of bioactive chemicals that modulate the DHT production, and four phytochemicals are potential candidates of nutraceuticals to adjust DHT levels in male hormonal dysfunction.</P><P><B>Electronic supplementary material</B></P><P>The online version of this article (10.1007/s10529-017-2480-5) contains supplementary material, which is available to authorized users.</P>

        <P>Dietary polyacetylenes from various foods have been receiving attention as promising cancer chemopreventive agents. However, until now, the detailed molecular mechanism and the regulatory proteins underlying these effects have not been elucidated. We investigated the effects of gymnasterkoreayne B (GKB), a model dietary polyacetylene from wild vegetables, on the programmed cell death of HCT116 human colorectal cancer cells. GKB inhibited HCT116 cell proliferation by inducing apoptotic cell death. GKB treatment resulted in ROS accumulation, leading to the activation of both intrinsic and extrinsic apoptotic pathway. We also found that FN1, TGFB1, APP, SERPINE1, HSPD1, SOD1, TXN, and ACTN4 may act as secretory signaling molecules during GKB-induced apoptotic cell death using LC–MS/MS identification followed by spectrum counting, statistical calculation, and gene ontology analysis.         Effects of fermented milk treatment on microbial population and metabolomic outcomes in a three-stage semi-continuous culture system

        <P><B>Abstract</B></P> <P>We investigated the impact of a fermented milk product on gut microbiota and their metabolism in 3 different conditions of the colon with a systemic viewpoint. An <I>in vitro</I> semi-continuous anaerobic cultivation was used to assess the colon compartment-specific influence of fermented milk, followed by a multiomics approach combining 16S rDNA amplicon sequencing and nuclear magnetic resonance (NMR) spectroscopy. The microbiome profiling and metabolomic features were significantly different across three colon compartments and after fermented milk treatment. Integrative correlation analysis indicated that the alteration of butyrate-producing microbiota (<I>Veillonella, Roseburia, Lachnospira,</I> and <I>Coprococcus</I>) and some primary metabolites (butyrate, ethanol, lactate, and isobutyrate) in the treatment group had a strong association with the fermented milk microorganisms. Our findings suggested that fermented milk treatment significantly affected microbial population in an <I>in vitro</I> cultivation system as well as the colonic metabolome in different ways in each of colon compartment.</P> <P><B>Highlights</B></P> <P> <UL> <LI> A fermented milk product was treated using an <I>in vitro</I> colonic fermentation. </LI> <LI> Three colon compartments showed an apparent microbial and metabolomic distinction. </LI> <LI> Fermented milk treatment induced an increase in butyrate-producing bacteria. </LI> <LI> There were clear changes in colonic primary metabolome after fermented milk treatment. </LI> </UL> </P> <P><B>Graphical abstract</B></P> <P>[DISPLAY OMISSION]</P>
