Association of 20 potential ATP2B1‐interacting genes with blood pressure in Koreans

Plasma membrane calcium‐transporting ATPase 1 (ATP2B1)is associated significantly with blood pressure in Caucasians and Asians. ATP2B1 regulates calcium homeostasis and belongs to the P‐type calcium pump family; several studies have identified diverse proteins that bind to ATP2B1. We hypothesized that ATP2B1 regulates blood pressure through ATP2B1‐interacting genes. To this end, 20 potential ATP2B1‐interacting genes were selected, 197 SNPs of which were analyzed for their association of systolic and diastolic blood pressure. These 20 genes were categorized into 2 groups: ATP2B1‐binding genes and ATP2B1‐cleaving calpain family members. Three ATP2B1‐binding genes (CALM1, NOS1, and PDLIM1)were associated with blood pressure, and a SNP in CALM1(rs2401887) generated the strongest association signal (beta=‐3.60±0.92, p=8.9×10^(‐5) for systolic blood pressure and beta=‐1.40±0.62, p=0.02 for diastolic blood pressure). Of the calpain family members, 3 genes (CAPN6, CAPN9, and CAPN10)were associated with blood pressure, and the CAPN10 SNP rs4676348 yielded the strongest association signal (beta=‐0.88±0.27, p=0.001 for systolic blood pressure and beta=‐0.58±0.18, p=0.015 for diastolic blood pressure). Further, the interaction of CALM1 to ATP2B1 was examined using the blood pressure of individuals who carried both variants of CALM1and ATP2B1 genes. Similarly the interaction of CAPN10 to ATP2B1 was also examined. The CALM1 variant (rs2401887)and CAPN10 variants (rs4676348) appear to decrease blood pressure further in addition to the decrease by the variant (rs17249754) of ATP2B1, which suggests that ATP2B1 might regulate blood pressure through the ATP2B1‐interacting genes CALM1 and CAPN10.

The true heartache is a condition of severe heartache corresponding to angina pectoris, as recorded from Hwang Jae Nai Kyung. According to the literatural study of true heartache, some results can be acquired, such as follows. 1. The site of the true Heartache, can be divided into two categories, first, its superficial and conscious area is the chest as same as the other heartache. But its inner lesion is the Heart-Meridian as others occupied at the Pericardium-Meridian in stead of the Heart-Meridian. 2. The etiological classification of true heartache, are Cold-evil, Heat-evil, Wind-evil, Blood stasis etc. But its major factor is Cold-evil, more than anything else. 3. The symptomatic signs of true heartache, consist of cyanotic change from hands and feet to phalanges; severe heartache pale complexion with cold breathing and its extreme state can manifestate unceased sweating called as Yang exhaustion.

The current study examined the effects of freeze-dried mulberry fruit on disaccharidase activity in the small intestine and the lowering of blood glucose in streptozotocin (STZ)-induced diabetic rats. Sprague-Dawley male rats were randomly assigned to one normal and three streptozotocin (STZ)-induced diabetic groups. The diabetic groups were fed a mulberry fruit-free diet (DM-group), 0.3% mulberry fruit diet (DM-F group) or 0.6% mulberry fruit diet (DM-2F group). After they were fed the experimental diets for three weeks, diabetes was induced with an intraperitoneal injection of streptozotocin 50 mg/kg b.w before sacrificing 9 days later using the same experimental treatments. Analyses of anthocyanins, flavonoid and 1-deoxynojirimycin (DNJ) of lyophilized mulberry fruit were carried out and the major anthocyanins were rutin (142.5 mg), isoquercitrin (10.3 mg), quercetin (5.8 mg), morin (1.6 mg) dihydroquercetin (3.83 mg), cy-3-O-glucopyranoside (230.45 mg) and cy-3-O-rutinoside (131.5 mg) on the basis of 100 g dry weight. Total DNJ content was 2.39 mg/g dry weight of lyophilized mulberry fruit. Blood glucose level decreased in the diabetic mts fed the mulberry fruit supplement. The content of the liver glycogen increased in the diabetic mts fed the mulberry fruit supplement. Disaccharidase activity in the proximal part of the intestine, such as that of maltase, sucrase and lactase in the mulberry fruit supplementation groups, were lower than that of the DM group. These results suggest that mulberry fruit possess a suppressive effect on hyperglycemia, possibly by inhibiting the activity of disaccharidase in the small intestine of rats.
Solid Phase Crystallization Kinetics of Amorphous Silicon at High Temperatures

Solid phase crystallization (SPC) of amorphous silicon is usually conducted at around 600°C since it is used In the application of flat panel display using thermally susceptible glass substrate. In this study we conducted SPC experiments at temperatures higher than 600°C using silicon wafers. Crystallization rate becomes dramatically rapid at higher temperatures since SPC kinetics is controlled by nucleation with high value of activation energy. We report SPC kinetics of high temperatures compared to that of low temperatures.
Gentamicin induced nitric oxide-related oxidative damages on vestibular afferents in the guinea pig

<P><B>Abstract</B></P><P>Gentamicin is a well-known ototoxic aminoglycoside. However, the mechanism underlying this ototoxicity remains unclear. One of the mechanisms which may be responsible for this ototoxicity is excitotoxic damage to hair cells. The overstimulation of the <I>N</I>-methyl-<SMALL>D</SMALL>-aspartate (NMDA) receptors increases the production of nitric oxide (NO), which induces oxidative stress on hair cells. In order to determine the mechanism underlying this excitotoxicity, we treated guinea pigs with gentamicin by placing gentamicin (0.5mg) pellets into a round window niche. After the sacrifice of the animals, which occurred at 3, 7 and 14 days after the treatment, the numbers of hair cells in the animals were counted with a scanning electron microscope. We then performed immunostaining using neuronal nitric oxide synthase (nNOS), inducible NOS (iNOS) and nitrotyrosine antibodies. The number of hair cells in the animals was found to decrease significantly after 7 days. nNOS and iNOS expression levels were observed to have increased 3 days after treatment. Nitrotyrosine was expressed primarily at the calyceal afferents of the type I hair cells 3 days after treatment. Terminal deoxynucleotidyl transferase biotin-dUTP nick end labeling (TUNEL) staining revealed positive hair cells 3 days after treatment. Our results suggest that inner ear treatment with gentamicin may upregulate nNOS and iNOS to induce oxidative stress in the calyceal afferents of type I hair cells, via nitric oxide overproduction.</P>

A human endogenous retroviral family (HERV-W) has been related to multiple sclerosis associated retrovirus (MSRV) and schizophrenia associated retrovirus (SZRV) sequences. The HERV-W family was identified in the cerebrospinal fluids and brains of individuals with schizophrenia. Using cDNA libraries derived from human brains, we performed PCR amplification and identified new 16 HERV-W env elements (9 from human adult brain and 7 from human fetal brain). Those sequences showed a high degree of sequence similarity (89.2-99.6%) with HERV-W env (GenBank accession no. AF072506). Clones HB-1, HB-9 from adult brain and FB-2, FB-5 from fetal brain showed no frameshift and termination codons by deletion/insertion or point mutation. Synonymous and nonsynonymous calculation indicated that these sequences (HB-1, HB-9, FB-2, FB-5) could be associated with an active provirus in human brain tissues. In phylogenetic analysis, clones HB-1, HB-9, FB-2, FB-5 containing putative amino acid sequences showed sister relationship with the HERV-W and W-7-1 derived from human chromosome 7. Taken together, our data suggest that the related genes of the HERV-W env sequences are expressed in human brains and may contribute to an understanding of biological function connected to neuropsychiatric diseases.

Background: Radiation proctitis is a common complication of radiation to lower abdomen and pelvis. Different modalities of treatment are available, however the effi cacy is incomplete. We hypothesized that human placenta and adipose-derived mesenchymal stem cells could have therapeutic potential on radiation proctitis. Methods: Placenta and adipose-derived mesenchymal stem cells were locally injected on distal rectal mucosa of female Sprague-Dawley rats within 24hrs of 25Gy rectal irradiation. 1,2,4 weeks later, they were sacrifi ced and the rectum was removed and blood sample was obtained to evaluate various parameters of infi ammation Results: Transplanted stem cell was identifi ed at rectal mucosa during 4 weeks after irradiation. Severe proctitis was provoked after pelvic irradiation refi ected with extensive infi ammatory cell infi ltration, loss of crypt epithelium and collagen deposition. However, treatment with mesenchymal stem cell restored these pathologic indices. In addition, mesenchymal stem cell had anti-infiammatory effect, anti- apoptotic effect. Regenerative property were increased. These effect was more prominent on adipose-derived mesenchymal stem cell. Conclusions: Placenta and adipose derived mesenchymal stem cell transplantation may be an potential therapeutic strategy to treatment of radiation proctitis.

More than 1,000 limestone caves are extensivelydeveloped in Gangwon-do and Chungcheongbuk-do, Korea. A vari-ety of numerous speleothems are actively growing in these cavestoday. A potential to use speleothems as regional paleoclimateproxy was explored by analyzing soda straw, stalactite and stalag-mite. These proxy recorders that have grown during the past several210Pb. Mostspecimens collected from six limestone caves were found to have anexcess 210Pb, indicating that they are less than about 100 yrs old.This excess 210Pb was employed to estimate the growth rate for atubular “soda straw” that yielded a longitudinal growth rate of1.7 m/yr past 60 years. Coeval decrease in δ13C values of the sodastraw, spanning the time period of about 1930195 AD, mayreflect the carbon isotope ratio of atmospheric CO2 for the sameof recent speleothems along with age control using 210Pb excessmethod from the speleothems in Korean limestone caves promise agreat potential for the reconstruction of climate and environmentalchanges during the past hundred years or so.
