      • 茶 침출조건에 따른 Catechin, 무기질 및 Vitamin C 함량 변화

        본 연구에서는 실제 상용하고 있는 각 차음료가 온도, 시간 및 추출 횟수에 따른 성분의 함량의 차이를 관찰코져 시판되고 있는 한국산 녹차, 우롱차 및 홍차를 대상으로 추출방법을 달리한 후 茶浸出液중의 주요성분인 catechin, vitamin C 및 여러가지 무기질의 함량을 측정하여 다음과 같은 결과를 얻었다. 1. 차침출온도에 따른 침출액중의 catechin과 vitamin C 함량의 차이를 볼 때 온도가 높을수록 많이 추출되었다. 2. 같은 온도라는 침출시간에 따른 차이를 관찰한 결과 시간이 길수록 침출액중의 catechin과 vitamin C의 함량이 증가되었다. 3. 녹차는 우려낸 횟수에 따른 catechin의 함량 차이는 처음에 비하여 2회 추출시약 51%, 3회추출시는 71% 감소하였다. 4. 차의 각 무기원소에 따른 함량을 비교시 Mg은 우롱차가 가장 높고 K, Mn은 녹차가, Zn, Cu 그리고 Fe는 홍차가 가장 높게 나타났다. 5. 녹차, 우롱차의 경우 대체로 무기질의 침출정도가 온도가 높을수록 오히려 감소하는 경향을 나타내었고 홍차는 Mg, K을 제외하고 모두 증가하였다. 6. 각 침출액중의 대부분의 무기질 함량은 추출횟수가 증가할수록 감소하였다. 7. 시료로 사용한 녹차, 우롱차, 홍차의 vitamin C 함량은 녹차, 우롱차, 홍차의 순으로 높게 나타났다. 8. 녹차를 추출한 횟수에 따른 vitamin C 함량은 1회에 비해 2회 추출시 약 36%, 3회 추출시 약 62% 감소되었다. 9. 5g의 녹차를 80℃에서 2분간 침출한 액중에서는 catechin은 100ml당 0.34g, Mg은 2.229mg, Ca은 0.365mg, K은 39.77mg, Zn은 0.037mg, Cu은 0.010mg, Fe는 0.001mg, Mn은 0.581mg이며, vitamin C는 9.65mg이 함유되어 있었다. This study was to compare the ingredients which are contained in usual Korean teas in-depednence on leaching temperature, time and number. The main contents i.e. catechin, vi-tamin C and other minerals extracted from Korean commercial green tea, oolong tea and black tea were measured. The resluts are as follows; According to extracting temperature, the higher temperature made catechin and vitamin C extracted more. For the same leaching temperature, the longer time the more catechin and vitamin C. In case of extracting number, the contents of catechin was decreased a-bout 50% and 70% at second time and third, respectively in comparasion to the first time. For minerals from teas, management(Mg) was most in oolong tea, potassium(K)and manganease(Mn) and in green tea and iron(FEe) was in black tea respectively. Most min-erals from greentea and oolong tea were extracted less as leaching temperature was in-creased. But in black tea minerals other than Mg and K were more at increased tempera-ture. Most minerals in extractions were decreased as leaching number was increased. The content of vitamin C in the experimental samples(tea) was most in green tea, next in oo-long tea and third in black tea. In comparasion with the first extraction vitamin C was de-creased about 35% and 60% at 2nd and 3rd leach, respectively.

        Fractionation of barley straw with dilute sulfuric acid for improving hemicellulose recovery

        Dilute acid fractionation of barley straw improves dissolving hemicellulose fraction of the straw, while leaving the cellulose more reactive and accessible to enzyme as a strategy of pretreatment. To characterize the fractionation process, the effects of the acid concentration, temperature and reaction time on the hemicellulose removal as well as on the formation of by-products (furfural, 5-hydroxymethylfurfural and acetic acid) were investigated. The optimum fractionation conditions of barley straw were 1% (w/v) concentration of sulfuric acid, 158 oC of reaction temperature and 15 min of reaction time. Under the optimum conditions, 87% of xylan was hydrolyzed and recovered in liquid hydrolyzate, which was 7% higher than that of the predicted yield. The hydrolyzate contained glucose 2.44 g/L, arabinose 1.70 g/L, xylose 13.41 g/L, acetic acid 1.55 g/L, levulinic acid 0.03 g/L, 5-HMF 0.03 g/L and furfural 0.75 g/L.

        Comparison of bioethanol production of simultaneous saccharification & fermentation and separation hydrolysis & fermentation from cellulose-rich barley straw

        Cellulose rich barley straw, which has a glucan content of 62.5%, followed by dilute acid pretreatment,was converted to bioethanol by simultaneous saccharification and fermentation (SSF). The optimum fractionation conditions for barley straw were an acid concentration of 1% (w/v), a reaction temperature of 158 oC and a reaction time of 15 min. The maximum saccharification of glucan in the fractionated barley straw was 70.8% in 72 h at 60 FPU/gglucan,while the maximum digestibility of the untreated straw was only 18.9%. With 6% content WIS (water insoluble solid) for the fractionated barley straw, 70.5 and 83.2% of the saccharification yield were in SHF and SSF (representing with glucose equivalent), respectively, and a final ethanol concentration of 18.46 g/L was obtained under the optimized SSF conditions: 34 oC with 15 FPU/g-glucan enzyme loading and 1 g dry yeast cells/L. The results demonstrate that the SSF process is more effective than SHF for bioethanol production by around 18%.

        Lignocellulosic biomass is the most abundant organic material on earth and also promising raw material for bioenergy production. Agricultural residues in the process of bio-oil extraction, is an abundant and low-cost lignocellulosic material. The technology for conversion of lignocellulosic biomass resources to fuels and chemicals, such as ethanol, has been under development for decades. One of the well-studied technologies that are currently being commercialized is to use a dilute acid-catalyzed pretreatment followed by enzymatic hydrolysis and fermentation to produce ethanol. In this work, the dilute-acid hydrolysis of agricultural residues was optimized through the utilization of statistical experimental design. Evaluation criteria for optimization of the pretreatment conditions were based on high xylose recovery and low inhibitor contents in the hydrolyzates. The purpose of this study was to gain a more accurate understanding of the quantities of acid required for effective hydrolysis and the reactivity trade-offs with reaction time and temperature that will enable overall process optimization.

        농업부산물 고추대 (Pepper Stem)을 이용한 묽은 황산 자일로즈 분별공정의 최적화

        본 연구에서는 반응표면분석법 (RSM)을 따른 고추 대의 묽은 황산을 이용한 자일로즈 분별 공정을 통해 고추대 시료로부터 최적의 자일로즈 수율을 얻을 수 있는 반응 조건을 예측할 수 있었다. 실제 실험 결과 최적 반응은 온도 147℃, 산 농도 2.8% H2SO4, 반응시간 21 min. 임을 알 수 있었고 최적 조건에서의 실험결과는 예측 값 78% 보다 약 7% 낮은 71% 자일로즈 수율을 얻을 수 있었다. SEM 과 XRD분석을 통해 전처리 전후의 시료의 이미지 분석을 하여 산 전처리 공정을 통해 섬유소 기질 외 다른 성분인 반 섬유소가 분해 되어 추출액상으로 자일로즈로 용해 되었음을 가시적으로 확인 할 수 있었고 전처리 된 고추대의 결정화 도가 전처리 전보다 증가했음을 확인 할 수 있었다. 이로써 비결정성 물질인 반섬유소와 기타 성분들이 묽은 황산을 이용한 자일로즈 분별공정에 의해 분리 되었음을 확인하였다. Response surface methodology (RSM) was used for optimization the fraction conditions of xylose from pepper stem with dilute sulfuric acid. The independent variables were acid concentration in the range of 1.134 to 2.866%, reaction temperatures in the range of 142.68 to 177.32℃, and hydrolysis time in the range of 6.34 to 23.66 min. were studied. The dependent variables were xylose yield from pepper stem, and the production of by-products, for example, furfural, acetic aicd, HMF etc. Experimental results had a good match with statistical result. The maximum xylose yield obtained in this experiment was 71% concentration.

        Nuclear translocation of anamorsin during drug-induced dopaminergic neurodegeneration in culture and in rat brain.

        <P>Anamorsin, also called cytokine-induced apoptosis inhibitor 1 (CIAPIN1), was recently identified to confer resistance to apoptosis induced by growth factor deprivation and to be indispensible for hematopoiesis. Recently, it was demonstrated that anamorsin is also widely distributed in both fetal and adult tissues. In this study, we evaluated the tissue distribution of anamorsin in the central nervous system (CNS) during development. In situ hybridization and immunoblot analyses revealed that anamorsin mRNA and protein were both highly and widely expressed in various regions of the CNS, including the cerebral cortex, hippocampus, midbrain, cerebellum, medulla, and spinal cord. Based on these findings, we examined its cellular localization during drug-induced neurodegeneration in MN9D dopaminergic cells. Both immunocytochemical localization and immunoblot analyses indicated that cytosolic anamorsin was translocated into the nucleus in a time-dependent manner following treatment with a reactive oxygen species (ROS)-inducing drug, 6-hydroxydopamine (6-OHDA). Treatment of cells with the apoptosis-inducing reagent, staurosporine, did not appear to cause translocation of anamorsin into the nucleus. When cells were treated with the nuclear export inhibitor, Leptomycin B, alone or with 6-OHDA, nuclear anamorsin levels increased, indicating that nuclear influx and efflux of anamorsin are regulated by 6-OHDA treatment. In rat brain injected with 6-OHDA, nuclear translocation of anamorsin was identified in certain tyrosine hydroxylase (TH)-positive neurons as well as TH-negative cells. Furthermore, treatment of MN9D cells with hydrogen peroxide or ROS-inducing trace metals caused nuclear translocation of anamorsin. Taken together, our data indicate that nuclear translocation of anamorsin is a ROS-dependent event and may participate in the regulation of transcription of critical molecules during dopaminergic neurodegeneration.</P>

        낙엽상 천포창 환자에서 발생한 노르웨이 옴

        Norwegian scabies can be defined as a generalized, severe scabies infestation which usually affects immunocompromised patients. The diagnosis is commonly missed, and can lead to mismanagement. We describe a patient with Norwegian scabies affecting the whole body. The patient had suffered with pemphigus foliaceus for several years and had an immunosuppressed status due to long-term use of oral corticosteroids. He was admitted to our hospital. At first, we made a diagnosis of exfoliative dermatitis secondary to pemphigus foliaceus. However, no improvement of the condition was seen after an increased dosage of oral corticosteroids, azathioprine, and mycophenolate mofetil. We then identified adult scabietic mites, eggs and scybala upon microscopic examination of skin scales. He was treated with an application of gamma benzene hexachloride cream. We recommend that the diagnosis of scabies be considered in immunocompromised patients with thick skin scales and associated night pruritus. (Korean J Dermatol 2006;44(11):1345~1348)

        인진쑥 배지에서 배양한 노루궁뎅이버섯 균사체 추출물(HEAC)의 알코올 대사촉진 활성

        인진쑥 배지에서 배양한 노루궁뎅이버섯(HEAC), 노루궁뎅이버섯과 인진쑥 추출물들의 음주 후 체내 알코올 농도의 감소 효과를 확인하였으며, 또한 혈중 알코올 탈수소효소 및 아세트알데히드 탈수소효소의 활성을 조사하여 알코올 분해 활성을 조사하였다. 체내 알코올 농도 측정결과 HEAC의 경우 170분, 노루궁뎅이 버섯 추출물은 210 분후에 혈중 알코올이 검출되지 않았다. 또한, HEAC는 알코올 탈수소효소 활성에서 대조군 보다 시간에 따라 154%, 노루궁뎅이버섯 추출물은 148%로 비교적 높은 활성을 나타내었다. 아세트알데히드 탈수소활성에서는 HEAC와 노루궁뎅이버섯 추출붙이 대조구에 비해 104~111%이상의 효소활성이 유지되는 것을 확인하였으며 이들 추출물들은 알코올 분해 촉진작용이 매우 우수함을 알았다. Alcohol concentration in the blood was effectively decreased by extracts from Hericium erinaceum hypha cultivated with Artermisia capillaris medium(HEAC), Hericium erinaceum hypha and Artermisia capillaris after dirnking. Also, the activities of alcohol dehydrogenase and acetaldehyde dehydrogenase in the blood was studied. As a result of testing an alcohol concentration in the blood, the alcohol in the blood was not detected after 170 min. in case of HEAC and after 210 min. in case of Hericium erinaceum. Compared to control, each activities of alcohol dehydrogenase of HEAC and Hericium erinaceum hypha was showed up to 154% and 148% respectively. The activities of the acetaldehyde dehydrogenase of both extracts from HEAC and Hericium erinaceum was maintained in the range of 104 to 110% compared to control. In conclusion, such extracts represent significant effect to facilitate decomposition of alcohol.
