Telomerase activity in pterygeal and normal conjunctival epithelium

All pterygia have similar histologic features of solar degeneration seen in the skin such as acanthosis, keratosis, or hyperkeratosis. Although the pathogenesis of pterygia is still unclear, an association with solar exposure, in particular with UV radiation, has been reported. Telomerase activity has been found to be higher in some degenerative, precancerous, and cancerous skin lesions. We investigated telomerase activity in the epithelium and the stromal tissues of the pterygium. Pterygeal tissues were obtained from 30 patients. Telomerase activity was measured with TRAPeze-ELISA kit. Three of the 28 (10.7%) pterygeal stromal tissues demonstrated positive telomerase activity. Fourteen of the 27(51.9%) epithelial tissues were positive in telomerase activity, whereas telomerase activity was positive in only 3 of 9 normal epithelia (33.3%). Telomerase activity in the pterygium-covered epithelium was increased as compared with that seen in the normal epithelium, but the increase was not statistically significant. In conclusion, telomerase activity was somewhat increased in pterygeal tissues. Telomerase activity may be involved in the pathogenesis of pterygium.

This paper presents a localization system of mobile robot using LRF(laser range finder) in indoor environment. The system is composed of three main procedures: data pre-processing, laser scan data segmentation, searching target object, and localization. Basic level data organization are used in data pre-processing. Breakpoint detection and edge detection are used for laser scan data segmentation. Breakpoint detection is used to determine sequence of measurements. Edge detection decides edge in segmented measurements. Extended IEPF(Iterative Endpoint Fit) algorithm is that used in the part. Each breakpoint and edge is used for searching three target objects.

Mutations in the RPE65 gene, associated with Leber congenital amaurosis, early-onset severe retinal dystrophy, and retinitispigmentosa, gained growing attention since gene therapy for patients with RPE65-associated retinal dystrophy is available inclinical practice. RPE65 gene accounts for a very small proportion of patients with inherited retinal degeneration, especiallyAsian patients. Because RPE65-associated retinal dystrophy shares common clinical characteristics, such as early-onset severenyctalopia, nystagmus, low vision, and progressive visual field constriction, with retinitis pigmentosa by other geneticmutations, appropriate genetic testing is essential to make a correct diagnosis. Also, fundus abnormalities can be minimal inearly childhood, and the phenotype is highly variable depending on the type of mutations in RPE65-associated retinal dystrophy,which makes a diagnostic difficulty. The aim of this paper is to review the epidemiology of RPE65-associated retinaldystrophy, mutation spectrum, genetic diagnosis, clinical characteristics, and voretigene neparvovec, a gene therapy productfor the treatment of RPE65-related retinal dystrophy.

Purpose: To evaluate whether systemically injected bone marrow-derived mesenchymal stem cells (MSCs) can be incorporated into neuroretinal tissues and play an important role in retinal wound healing in the laser-induced retinal trauma model. Methods: Retinotomies were made by applying an Nd:YAG laser to rat retina. On the first day after the injuries, cell suspensions that were obtained from the same line of rat (containing 1 × 106 green fluorescence protein [GFP]-marked bone marrow-derived MSCs) were injected through a tail vein in the experimental group and phosphate buffer solution (PBS) was injected in the same way in the control group. Fundus photographs were taken serially for fundus examination and eyeballs were enucleated for histological studies that were conducted at five and seven weeks after MSC and PBS injection. After the tissues were prepared, the retinotomy sites were observed with routine histological staining and confocal microscopy. Results: Retinal detachment resolved in the experimental group, whereas it progressed in the control group. The retinotomy sites closed partially with identifiable GFP positive cells 5 weeks after MSC injection. At 7 weeks after MSC injection, complete healing without retinal detachment and plentiful GFP positive cells were observed at the transitional zone between damaged and normal retina. Conclusions: Systemically administered GFP-marked MSCs may be incorporated into the neuroretinal tissues and play an important role in the wound modulation of physically damaged retinal tissues.

Age-related macular degeneration (AMD) is the most frequent cause of vision loss in the elderly, and there is currently no effective treatment. Recently, many studies have attempted to reduce the risk of conversion to late AMD through lifestyle correction, smoking cessation and exercise. Several studies have also found that taking supplements orally can help prevent disease progression. Nutritional supplements consisting of vitamin C, vitamin E, beta-carotene, zinc, and copper were taken in the Age-Related Eye Disease Study (AREDS) 1, and lutein and zeaxanthin were recommended instead of beta-carotene in AREDS 2. This review focuses on the role of supplements in the prevention of AMD and summarizes the current issues about the most frequently used micronutrients.

Purpose: To evaluate the changes in multifocal electroretinogram (mfERG) and optical coherence tomography (OCT) after intravitreal bevacizumab injection in the treatment of age-related macular degeneration (AMD). Methods: Twenty-one eyes with choroidal neovascularization secondary to AMD were studied before and after intravitreal bevacizumab injection for best corrected visual acuity (BCVA), OCT, and mfERG. Results: The BCVA improved, while central macular thickness and total macular volume in OCT decreased after intravitreal bevacizumab injection (p=0.03, 0.01, and 0.01, respectively). In mfERG, the amplitude of P1, and implicit time of P1 and N1 indicated a statistically significant improvement of retinal response after intravitreal bevacizumab injection. Conclusions: There is a potential role for mfERG in evaluating the effect on retinal function of intravitreal bevacizumab injection.

Purpose: To compare short-term efficacy and safety of intravitreal brolucizumab injection with aflibercept in treatment-naive neovascular age-related macular degeneration (nAMD) patients. Methods: A total of 59 eyes from 59 treatment-naive nAMD patients in three hospitals were retrospectively reviewed. Of which, 27 patients underwent intravitreal brolucizumab injections and 32 received aflibercept. After monthly consecutive three injections, best-corrected visual acuity (BCVA; in logarithm of minimal angle of resolution [logMAR]), central macular thickness (CMT), dry macula achievement rate, and intraocular inflammation (IOI) incidence were compared. Results: After loading-phase treatment, BCVA was significantly increased from 0.48 ± 0.30 logMAR at baseline to 0.33 ± 0.21 logMAR at 3 months in the brolucizumab group (p = 0.002) and 0.40 ± 0.39 logMAR at baseline to 0.33 ± 0.36 logMAR at 3 months in the aflibercept group (p = 0.007). But there was no significant difference in BCVA improvement at 3 months between the two groups. CMT significantly decreased from 429.67 ± 250.59 μm at baseline to 210.67 ± 93.53 μm at 3 months in the brolucizumab group and from 346.69 ± 159.09 μm to 234.52 ± 83.42 μm in the aflibercept group (both p < 0.001). The amount of CMT reduction was significantly greater in the brolucizumab group after 3 months (p = 0.036). In typical AMD eyes, brolucizumab showed similar BCVA improvement but better CMT reduction at 3 months (p = 0.018). Dry macula achievement rate was not significantly different between the two groups. One IOI was observed in the brolucizumab group. Conclusions: Intravitreal injections of brolucizumab and aflibercept showed similar anatomical and functional outcomes. But CMT reduction was greater in the brolucizumab group. One IOI was identified, which was tolerable for topical agents. These results suggest that brolucizumab could be a novel first line treatment option for treating naive nAMD patients.

Purpose: To investigate the changes of electroretinogram (ERG) at baseline and during the first 3 injections of intravitreal bevacizumab in central retinal vein occlusion (CRVO) patients. Methods: Thirteen eyes of 13 CRVO patients who received 3 injections of intravitreal bevacizumab at 6-week intervals and who could be examined for visual acuity, optical coherence tomography and ERG at 4 weeks after each injection were included in the present study. In addition, the ERG results of the unaffected fellow eyes group at 4 weeks after each injection were compared with the CRVO eyes group. Results: Amplitudes of rod b wave, a and b waves of maximal combined response and cone response, oscillatory potentials and 30-Hz flicker significantly decreased in the CRVO eyes group than the fellow eyes group at 4 weeks after each injection (<em>p</em> < 0.001). After injection, amplitudes of all the above-mentioned ERG parameters gradually increased during the 1st and the 2nd injection (<em>p</em> < 0.001). However, there was no significant difference at 4 weeks after the 2nd and 3rd injections (<em>p</em> > 0.05). Conclusions: After intravitreal bevacizumab injection, macular functions significantly improved and amplitudes of all ERG parameters gradually increased in CRVO patients.
