Standard Test Block(STB) for UT(Ultrasonic Testing) is a block approved by authoritative for material, shape and quality. STB is used for characteristic tests, sensitivity calibration and control of the time base range of UT inspection devices. The material, size and chemical components of STB should be strictly controlled to meet the related standards such as ASTM and JIS because it has an effect upon sensitivity, resolution and reproductivity of UT. The STBs which are not approved are sometimes used because the qualified STBs are very expensive. So, the purpose of this study is to survey the characteristics, quality and usability of Non-Standardized Test Blocks. Non-Standardized Test Blacks did not meet the standard requirements in size or chemical components, and ultrasonic characteristics. Therefore if the Non-Standardized Test Blocks are used without being tested, it's likely to cause errors in detecting the location and measuring the size of the defects.

In the course of screening to find a plant material decreasing the activity of triacylglycerol and cholesterol, we identified Tripterygium regelii (TR). The methanol extract of TR leaves (TR-LM) was shown to reduce the intracellular lipid contents consisting of triacylglycerol (TG) and cholesterol in HepG2 cells. TR-LM also downregulated the mRNA and protein expression of the lipogenic genes such as SREBP-1 and its target enzymes. Consequently, TR-LM reduced the TG biosynthesis in HepG2 cells. In addition, TR-LM decreased SREBP2 and its target enzyme HMG-CoA reductase, which is involved in cholesterol synthesis. In this study, we evaluated that TR-LM attenuated cellular lipid contents through the suppression of de novo TG and cholesterol biosynthesis in HepG2 cells. All these taken together, TR-LM could be beneficial in regulating lipid metabolism and useful preventing the hyper-lipidemia and its complications, in that liver is a crucial tissue for the secretion of serum lipids.

Obstructive sleep apnea (OSA) is characterized by repetitive upper airway collapse during sleep, causing chronic intermittent hypoxia (CIH). OSA affects 2-5% of the middle-aged population and is associated with significant morbidity and mortality. In intensive care unit, considerable numbers of patients with acute lung injury (ALI) have underlying OSA. However, little is known regarding the effects of OSA on the clinical course of ALI. This study is aimed to evaluate the impact of CIH on inflammation, fibrosis and related pathway in a murine model of ALI. Seven-to-eight week-old male, C57BL/6J mice were used. The mice were divided into two groups and exposed to room air (RA) or CIH. During 4 weeks, the CIH group was exposed to a 30 s RA/hypoxia (approximately fractional inspired O2 7±0.5%) cycle for 8 hours a day. After 4 weeks, each group was divided into two additional groups with intratracheal lipopolysaccharide (LPS, 0.5 mg/kg) or saline exposure. In CIH group with LPS exposure, lung wet/dry ratio and lung injury score were higher than other three groups. Level of hyproxyproline, TGF-beta, VEGF and collagen deposition were also higher in CIH group with LPS exposure compared to other groups. From the cell differentiation of bronchoalveolar lavage, CIH group with LPS exposure showed significantly higher number of total cell and neutrophil compared with other groups, especially compared with RA group with LPS exposure (p<0.001, respectively). These results suggest that CIH may aggravate inflammation and fibrosis in LPS induced ALI, resulting more damage.

Background: Asthma is characterized by airway inflammation and remodeling. Roflumilast is a selective phosphodiesterase-4 (PDE4) inhibitor that has anti-inflammatory effect in chronic obstructive pulmonary disease (COPD). However, little is known regarding the effect of roflumilast in chronic asthma. This study is aimed to evaluate the effects of roflumilast on airway inflammation and remodelling in a murine model of chronic asthma. Methods: Female BALB/c mice, 6 weeks of age, were used. We developed a mouse model of airway remodeling in which ovalbumin (OVA)-sensitized mice were repeatedly exposed to intranasal OVA administration twice a week for 3 months. Roflumilast was administered orally starting on the 38th and 5 days a week thereafter for 3 months during the intranasal OVA challenge. A lung fibroblast cell line was used in the proliferation assay. Results: Compared with control mice, mice chronically exposed to OVA developed sustained eosinophilic airway inflammation, airway hyperresponsiveness (AHR), and exhibited features of airway remodeling. Administration of roflumilast significantly inhibited eosinophilic inflammation and AHR. The levels of interleukin (IL)-4, IL-5, and IL-13 in the bronchoalveolar lavage (BAL) fluids were significantly lower in the roflumilast group. The level of hydroxyproline was also significantly lower in the roflumilast group. In vitro, roflumilast significantly inhibited stem cell factor (SCF) induced cell proliferation of fibroblasts. Conclusions: These results suggest that roflumilast administration modulates the airway inflammation, AHR, and airway remodeling associated with chronic allergen challenge.

Zanthoxylum ailanthoides (ZA) has been used as folk medicines in East Asian and recently reported to have several bioactivity; however, the studies of ZA on the regulation of triacylglycerol (TG) biosynthesis have not been elucidated yet. In this study, we examined whether the methanol extract of ZA (ZA-M) could reduce oleic acid- (OA-) induced intracellular lipid accumulation and confirmed its mode of action in HepG2 cells. ZA-M was shown to promote the phosphorylation of AMPK and its upstream LKB1, followed by reduction of lipogenic gene expressions. As a result, treatment of ZA-M blocked de novo TG biosynthesis and subsequently mitigated intracellular neutral lipid accumulation in HepG2 cells. ZA-M also inhibited OA-induced production of reactive oxygen species (ROS) and TNF-α, suggesting that ZA-M possess the anti-inflammatory feature in fatty acid over accumulated condition. Taken together, these results suggest that ZA-M attenuates OA-induced lipid accumulation and inflammation through the activation of LKB1/AMPK signaling pathway in HepG2 cells.

The conventional and Bayesian procedures which make use of aggregate time series data have been revieved for estimating transition probability and applied to forecast the planted area of agricultural crops. Also, the predictive performance of various estimators is examined with respect to the mean squared error(MSE) and the chi square value through the x^2 goodness of fit test. The results are consistent with the hypothesis that data are generated from Markov process, which indicate that estimation procedures yield reasonable results.
