        성인 쇄골 중위부 골절의 수술적 치료

        The clavicle plays very important role in the motion of the shoulder girdle and maintenance of power and stability of the arm. The most clavicular shaft fractures can be treated by conservative methods with a high rate of union and low rate of complications. The operative treatment in clavicular fracture is limited in such as wide separation of the fragments with soft tissue interposition, nonunion, or neurovascular involvements. From 1989 to 1991, eighty six consecutive patients with clavicular shaft fractures were treated at Ewha Womans University Hospital. Fifty eight patients were treated conservatively(Group I ) and twenty eight patients operatively(Group II) Authors compared both groups and obtained the following results; 1. The most common cause of injuries was the traffic accident and the most common associated skeletal injury around the shoulder was the rib fracture. 2. The degree of comminution and displacement of the clavicular shaft fracture were more severe in Group lI. 3. The average time to union was 8.7 weeks in Group I and 10.3 weeks in Group II. 4. The average rate of union at 4 months was 93.1% in Group I and 96.4% in Group II. 5. The functional result was good or excellent in over 90% in both Groups according to the Kang's criteria. 6. The rate of complication was 8.6% in Group I (one nonunion, three delayed unions and one malunion) and 3.6% in Group lI(only one case of nonunion) 7. The immobilization period was between 6 to 8 weeks in Group I and within 4 weeks in Group II in most cases.

      • 유산균(L.lactis CBT-8)및 발효 추출물의 Helicobacter pylori 억제 능력

        Helicobacter pylori infection was the cause of chronic gastritis, duodenal ulcer and gastric MALT lymphoma. The eradication rates of H. pylori using antibiotics are around 80%. Lactobacilli have been demonstrated to have in vivo and in vitro inhibitory effects on H. pylori infection. We aimed to evaluate the inhibitory effect of L. lactis CBT-8 and its extract on H. pylori in human stomach. Thirty-five H. pylori-infected volunteers(30-49 yr)were randomized into two groups which were treated with L. lactis CBT-8 and its extract(Group A n=17) or milk containing L. lactis CBT-8(Group B, n=18). They underwent 13C-urea breath tests before and after treatment. The DOB30 of group A(n=14) was decreased 41.0% after treatment(26.0±4.9 before, 15.3±2.1 after treatment)(p<0.05). the DOB30 of group B(n=13) was decreased 22.3% after treatment(22.0±2.2 before, 17.1±1.7 after treatment)(p<0.05). In conclusion, L. lactis CBT-8 was effective in suppressing H. pylori infection in human stomach.

        Association between FGFRs and the susceptibility of digestive and reproductive system cancers in Chinese population

        Fibroblast growth factors(FGFs) and their receptors (FGFRs) modulate a wide range of biological functions, especially tumor genesis. The aim of this study was to investigate the possible association of FGFR expression with the susceptibility of digestive system and reproductive system cancers in Chinese population. In total, 343 patients with digestive or reproductive system cancers were enrolled in this study. The expression levels of four highly-conserved FGFRs including FGFR1, FGFR2, FGFR3 and FGFR4 were measured by immunohistochemistry (IHC). The expression levels of FGFRs were compared between carcinoma and para-carcinoma tissues. FGFR1 expression significantly differed between carcinoma and para-carcinoma tissues in colon and gastric cancers. FGFR2 expression significantly differed between carcinoma and para-carcinoma tissues in esophageal cancer. FGFR3 expression was significantly different between carcinoma and para-carcinoma tissues in liver and gastric cancers. FGFR2 showed the highest expression probability in all the selected cancers and FGFR4 showed the lowest expression probability. FGFR1 and FGFR3 showed comparable moderate expression probabilities. Our findings have demonstrated significant differences regarding FGFR expression levels between carcinoma and para-carcinoma cells in digestive or reproductive system cancer patients. The data also implicated that FGFR2 and FGFR4 could serve as two prominent factors closely related to the susceptibility of digestive and reproductive system cancers.

        Systematic characterization of A-to-I RNA editing hotspots in microRNAs across human cancers

        <P>RNA editing, a widespread post-transcriptional mechanism, has emerged as a new player in cancer biology. Recent studies have reported key roles for individual miRNA editing events, but a comprehensive picture of miRNA editing in human cancers remains largely unexplored. Here, we systematically characterized the miRNA editing profiles of 8595 samples across 20 cancer types from miRNA sequencing data of The Cancer Genome Atlas and identified 19 adenosine-to-inosine (A-to-I) RNA editing hotspots. We independently validated 15 of them by perturbation experiments in several cancer cell lines. These miRNA editing events show extensive correlations with key clinical variables (e.g., tumor subtype, disease stage, and patient survival time) and other molecular drivers. Focusing on the RNA editing hotspot in miR-200b, a key tumor metastasis suppressor, we found that the miR-200b editing level correlates with patient prognosis opposite to the pattern observed for the wild-type miR-200b expression. We further experimentally showed that, in contrast to wild-type miRNA, the edited miR-200b can promote cell invasion and migration through its impaired ability to inhibit <I>ZEB1/ZEB2</I> and acquired concomitant ability to repress new targets, including <I>LIFR</I>, a well-characterized metastasis suppressor. Our study highlights the importance of miRNA editing in gene regulation and suggests its potential as a biomarker for cancer prognosis and therapy.</P>

        Effects of Cu<sup>2+</sup> on alkaline phosphatase from <i>Macrobrachium rosenbergii</i>

        <P><B>Abstract</B></P> <P>To gain insight into the effect of Cu<SUP>2+</SUP> on the activity and structure of alkaline phosphatase (ALP) from <I>Macrobrachium rosenbergii</I>, the enzyme was purified using ammonium sulfate fractionation, Sephacryl S-200, and DEAE anion exchange chromatography. We studied Cu<SUP>2+</SUP>-mediated inhibition and aggregation of ALP, and found that Cu<SUP>2+</SUP> significantly inactivated ALP activity with an <I>IC</I> <SUB>50</SUB> of 1.47 ± 0.02 mM. We further revealed that Cu<SUP>2+</SUP> reversibly inhibited ALP in a mixed-type manner with <I>K</I> <SUB>i</SUB> = 0.41 ± 0.02 mM. Time-interval kinetics showed that the inhibition followed first-order reaction kinetics. This process was associated with conformational changes and significant transient free-energy change. Spectrofluorometry results showed that Cu<SUP>2+</SUP> induced ALP tertiary structural changes, including the exposure of hydrophobic surfaces that directly induced ALP aggregation. The results provide new information regarding ALP from <I>M. rosenbergii</I>.</P>

        <P>Fundamental understanding;of reactions, of lithium peroxides and superoxides is this for :the development of Li-O-2 batteries. In context, an investigation is reported of the hydrolysis of lithium superoxide, which has recently been synthesized in a Li-O-2 battery. Surprisingly, the hydrolysis of solid LiO2 is significantly different from that of NaO2 and KO2. Unlike KO2 and NaO2, the hydrolysis of LiO2 does not produce H2O2. Similarly, the reactivity of Li2O2 toward water differs from LiO2 in that Li2O2 results in H2O2 as a product. The difference in the LiO2 reactivity with water due to the more exothermic nature of the formation of LiOH and O-2 compared with the corresponding reactions, of NaO2 and KO2. We also show that a titration method used in this study, based on reaction of the discharge product with a Ti(IV)OSO4 solution; provides a useful diagnostic technique to provide information Oh the composition of a,discharge product in a Li-O-2 battery.</P>

        Proteomic analyses for profiling regulated proteins/enzymes by <i>Fucus vesiculosus</i> fucoidan in B16 melanoma cells: A combination of enzyme kinetics functional study

        <P><B>Abstract</B></P> <P>Fucoidans are complex sulfated polysaccharides that have a wide range of biological activities. Previously, we reported the various effects of <I>Fucus vesiculosus</I> fucoidan on tyrosinase and B16 melanoma cells. In this study, to identify fucoidan-targeted proteins in B16 melanoma cells, we performed a proteomics study and integrated enzyme kinetics. We detected 19 candidate proteins dysregulated by fucoidan treatment. Among the probed proteins, the enzyme kinetics of two candidate enzymes, namely lactate dehydrogenase (LDH) as an upregulated protein and superoxide dismutase (SOD) as a downregulated enzyme, were determined. The enzyme kinetics results showed that <I>Fucus vesiculosus</I> fucoidan significantly inhibited LDH catalytic function while it did not affect SOD activity even at a high dose, while only slightly decreased activity (up to 10%) at a low dose. Based on our previous and present observations, fucoidan could inhibit B16 melanoma cells growth via regulating proteins/enzymes expression levels such as LDH and SOD known as cell survival biomarkers. Interestingly, both expression level and enzyme catalytic activity of LDH were regulated by fucoidan, which could directly induce the apoptotic effect on B16 melanoma cells along with SOD downregulation. This study highlights how combining proteomics with enzyme kinetics can yield valuable insights into fucoidan targets.</P>

      • Effect of KIOM-79 against mitochondrial damage induced by streptozotocin in pancreatic beta-cells.

        <P>The present study examined the effects of KIOM-79 on streptozotocin (STZ)-induced mitochondrial oxidative stress in rat pancreatic beta-cells (RINm5F). KIOM-79 is a mixture of plant extracts from parched Puerariae radix, gingered Magnoliae cortex, Glycyrrhizae radix, and Euphorbiae radix. A marked increase in mitochondrial reactive oxygen species (ROS) was observed in STZ induced diabetic cells, which returned to control conditions after KIOM-79 treatment. Mitochondrial manganese superoxide dismutase (Mn SOD) activity and its protein expression were downregulated by STZ treatment but upregulated by KIOM-79 treatment. In addition, KIOM-79 treatment restored the loss of the mitochondrial membrane potential (Deltapsi) produced by STZ treatment. KIOM-79 induced an increase in Bcl-2 and a decrease in phospho Bcl-2 and Bax, which are related to permeability of the mitochondrial membrane. Further, KIOM-79 inhibited the translocation of cytochrome c from the mitochondria to the cytosol and elevated the ATP level, which was reduced by STZ treatment. These results suggest that KIOM-79 exhibits a protective effect through activation of antioxidant defense mechanisms and by attenuation of mitochondrial dysfunction in STZ-induced diabetic cells.</P>

        Obstacle-negotiation performance on challenging terrain for a parallel legwheeled robot

        High obstacle-negotiation performance of quadruped robots for the challenging terrain is strongly demanded in some fields, including payload delivery and disaster relief. In this study, a parallel leg-wheeled robot with the high mobility performance on targeted types of rough terrain is proposed. In contrast to other obstacle-negotiating robots, the strength of this robot is that it has adequate over-obstacle capabilities with a horizontal body in terms of different obstacle shapes, which mainly derives from the multi-DOF (degree of freedom) flexible locomotion of legs and the independent actuated wheels as end effectors. Particularly for a steep terrain where a large inertia robot negotiates it with low efficiency and stability, the proposed step-over gait, combined with static gait and wheeled locomotion, improves the overobstacle efficiency and maintains pose stability. Finally, simulations and experiments verify the performance on negotiating the diverse obstacles with level body over challenging terrain.
