Insulin has a variety of activities like as glucose uptake, inhibition of lipolysis, glycogen biosynthesis in a number of cells and tissues. Recently insulin has known to exert an antiapoptotic activity in different cell systems. However, it is not clear by which insulin can protect apoptosis cells from apoptosis. Thus, CHO cells expressing the human insulin receptors (CHO-IR) were used to evaluate the importances of different signaling steps activated by insulin in this process. The Ras/Raf-1/MEK/ERK cascade and the serine/threonine protein kinase Akt are two survival pathways that may be activated in response to insulin. We tested the hypothesis that inhibition of farnesylated Ras was casually related to manumycin-induced apoptosis and showed that the response to manumycin was found to be independent of K-Ras function. Moreover, blocking Ras/Raf-1/MEK/ERK cascade by the expression of a transdominant inhibitory mSOSI mutant in CHO-IR cells kept cells to sensitive to the antiapoptotic action of insulin. Insulin-dependent activation of Akt was blocked by 4th treatment with manumycin. This study suggest that the deplection of short-lived farnesylated proteins by manumycin suppresses the antiapoptotic action of insulin at least in part by disrupting Aktactivation but not of the K-Ras/Raf-1/MEK/ERK-dependent cascade.

The objective of this study is to investigate life and fatigue crack propagation in the Reduced Activation Ferritic steel(RAFs) JLF-1. Crack growth rate tests on compact tension specimen of thickness 12mm were conducted by using sinusoidal waveform. Compact tension specimens were used and crack growth rate da/dN and stress intensity factor range ΔK were analyzed concerning to the R value of 0.1, 0.3 and 0.5 In order to find out the value of ΔK, load amplitude constant method was applied by the standard fatigue testing method describes in ASTM E647-95a. The effects of stress ratio and specimen size on the fatigue crack growth propagation for JLF-1 steel were discussed within the Paris law. As the result of this study, fatigue crack growth rate increased with in creasing the load ratio, Consequently, fatigue crack propagation of half size specimen was similar to those of full size specimen at room temperature. Also, there was no difference between base metal and weld metal at room temperature and high temperature.(400℃)

Background/Aims: This study aimed to investigate whether the apolipoprotein (Apo) B/ApoA-I ratio is associated with carotid intima-media thickness (CIMT) in type 2 diabetes mellitus (T2DM) subjects with low density lipoprotein cholesterol (LDL-C) levels less than 100 mg/dL. Methods: This cross-sectional study included 845 subjects aged with T2DM 40 to 75 years who had visited Huh’s Diabetes Center in Seoul, Republic of Korea for CIMT measurement. Traditional fasting lipid profiles, ApoB and ApoA-I levels were examined. CIMT was measured at three points on the far wall of 1 cm long section of the common carotid artery in the proximity of the carotid bulb. The mean value of six measurements from right and left carotid arteries were used as the mean CIMT. In this study, carotid atherosclerosis was defined as having a focal plaque or diffuse thickening of the carotid wall (mean CIMT ≥ 1.0 mm) Results: The prevalence of carotid atherosclerosis increased with ApoB/ApoA-I ratio. The ApoB/ApoA-I ratio, expressed as both quartiles (odds ratio [OR], 2.14; 95% confidence interval [CI], 1.21 to 3.79; p for trend = 0.014) and continuous values (OR, 10.05; 95% CI, 3.26 to 30.97; p < 0.001), was significantly associated with a higher risk for carotid atherosclerosis, regardless of conventional cardiovascular disease risk factors. The optimal ApoB/ApoA-I ratio cutoff value for detecting carotid atherosclerosis was 0.57, based on receiver operating characteristic curve analysis with a sensitivity of 58.0% and a specificity of 55.1%. Conclusions: A high ApoB/ApoA-I ratio was significantly associated with carotid atherosclerosis in T2DM patients with LDL-C levels less than 100 mg/dL.

<P>BACKGROUND/AIMS: The aim of the present study was to investigate the clinicopathological significance of p53, Ki-67, and p16 expression in patients with colorectal adenocarcinoma. METHODOLOGY: We evaluated p53, Ki-67, and p16 expression in 356 patients with primary colorectal adenocarcinoma using an immunohistochemical staining method. The relationships between these protein expressions and clinicopathological factors were statistically analyzed. RESULTS: Positive p53 staining was detected more often in typical adenocarcinoma compared to mucinous adenocarcinoma (49% versus 17%, p = 0.007) and in well or moderately differentiated adenocarcinoma compared to poorly differentiated adenocarcinoma (50% versus 32%, p = 0.030). The level of expression of p53 protein was related to lymph node metastasis (p < 0.001) and the TNM stage of the colorectal adenocarcinoma (p = 0.006). The p53 protein expression was related to an increased tendency of lymphovascular invasion (p = 0.058). However, Ki-67 and p16 expression levels were not associated with any of the clinicopathological variables. The overexpression of p53 was correlated with a higher level of Ki-67 (p = 0.001) and positive staining of p16 (p < 0.001). CONCLUSIONS: Our data suggest that overexpression of p53, which was correlated with Ki-67 and p16 expression, plays a critical role in aggressive tumor behaviors in patients with colorectal adenocarcinoma. However, further long-term followup studies are warranted to evaluate the clinical impacts of p53 in a larger group of patients.</P>

OBJECTIVE:: This study evaluated the predictive value of a number of tissue biomarkers, including proliferating cell nuclear antigen, survivin, thymidine phosphorylase, thymidylate synthase, bax, p53, nuclear factor-kappa B, vascular endothelial growth factor, matrix metalloproteinase-2, matrix metalloproteinase-9, CD133, CD44, and cyclooxygenase-2 with regard to preoperative chemoradiation in rectal cancer. BACKGROUND:: The ability to predict tumor response before treatment may significantly impact the selection of patients for preoperative chemoradiation therapy for rectal cancer. However, no definite predictive marker is known. METHODS:: Pretreatment biopsies from 123 patients who underwent preoperative chemoradiation were included. The mRNA levels of 13 biomarkers were analyzed by reverse transcriptase-polymerase chain reaction, with normalization relative to glyceraldehydes 3-phosphate dehydrogenase. Response to treatment was assessed by a 4-point tumor regression grade scale based on the ratio of fibrosis to residual cancer. RESULTS:: Among the 13 markers, no significant correlations in terms of T downstaging, N downstaging, and tumor-node-metastasis downstaging were observed. On multiple logistic regression analysis, only CD44 expression was found to be significant independent predictive factors for tumor regression grade response [odds ratio, 4.694 (1.155, 17.741), P = 0.030]. CD44 mRNA expression was significantly associated with expressions of the remaining 12 markers (all P < 0.05). Among the 118 patients receiving radical resection, proliferating cell nuclear antigen was the only independent factor to predict pathologic node negative status [odds ratio, 4.328 (1.078, 12.536), P = 0.037]. CONCLUSIONS:: Elevated CD44 mRNA levels in pretreatment biopsies might be predictive of poor tumor regression after preoperative chemoradiation in rectal cancer. Moreover, the proliferating cell nuclear antigen mRNA level might be predictive of nodal regression.
