Protein kinase A-induced phosphorylation at the Thr154 affects stability of DJ-1

<P><B>Abstract</B></P> <P><B>Introduction</B></P> <P>Most cases of Parkinson's disease (PD) are sporadic, but genetic variations have been discovered in PD patients. PARK7/DJ-1 is a known cause of early-onset autosomal-recessive PD and is implicated in neuroprotection against oxidative stress. Although several post-translational modifications of DJ-1 have been proposed, phospho-modification of DJ-1 and its functional consequences have been less studied.</P> <P><B>Methods</B></P> <P>Putative phosphorylation sites of DJ-1 were determined by liquid chromatography-tandem mass spectrometry (LC-MS/MS analysis). Subsequently, phosphorylation site of DJ-1 was confirmed by <I>in vitro</I> kinase assay and cell-based pull-down assay. Impaired dimer formation of phospho-null mutant was measured using DSS crosslinking assay and immunoprecipitation assay. To evaluate physiological consequences of this event, protein stability of DJ-1 WT and DJ-1 phospho-null mutant were compared using cycloheximide chase assay and ubiquitination assay.</P> <P><B>Results</B></P> <P>Here, we showed that DJ-1 directly bound to the catalytic subunit of protein kinase A (PKAcα). We found that PKAcα is responsible for phosphorylation of DJ-1 at the T154 residue. Interestingly, dimerization of DJ-1 was not detected in a DJ-1 T154A mutant. Furthermore, stability of the DJ-1 T154A mutant was dramatically reduced compared with that of wild-type DJ-1. We found that DJ-1 T154A was prone to degradation by the ubiquitin proteasome system (UPS).</P> <P><B>Conclusion</B></P> <P>We identified a novel phosphorylation site of DJ-1. Furthermore, we determined protein kinase A that is responsible for this posttranslational modification. Finally, we demonstrated physiological consequences of this event focusing on dimerization and protein stability of DJ-1.</P> <P><B>Highlights</B></P> <P> <UL> <LI> PKAcα phosphorylates PARK7/DJ1 at the Thr154 residue. </LI> <LI> Disruption of Thr154 phosphorylation is linked to a lower stability. </LI> </UL> </P>

With advance of cellular phone technologies, the use of short message transmission with small or portable electronic devices has been explosively increasing. In spite of such demands there have been not many studies for Korean characters and their symbol combinations (below "Hangul"). While some other studies have been focused on the Hangul vowel, in this paper we proposed a new Hangul input system which presents Hangul consonants with basic symbol components (called as "hweok"). And we have performed performance evaluation with comparison to SK's Chunjiin and LG's Naragul in terms of the number of key buttons, finger moving distance and key strokes. The proposed system can make all of Korean characters with 5 or 6 Hangul hweoks without significant degradation of system performance.

In recent years, as the demands of VBNS(Very high speed Backbone Network Service) and VDSL(Very high-data rate Digital Subscriber Line) increase, the development of kernel parts of optical communication such as PLC(Planar Light Circuit), Coupler, WDM elements increase. The alignment and the attachment technology are very important to fabricate the optical elements for communication. In this paper, the mechanism of rotational stage are studied. with the three different method and the results of them are applied to the design of the system. The performance test of resolution and travel is performed.

Objectives : This study aims to estimate the prevalence of the DSM-IV psychiatric disorders in Korean population using the Korean version of Composite International Diagnostic Interview (K-CIDI). Methods : Subjects were selected by taking multi-stage, cluster samples of 7,867 adult household residents, 18 to 64 years of age, in ten catchment areas. Total 78 trained interviewers administered the K-CIDI to the selected respondents, from June 1 to November30,2001. Results : Total 6,275 respondents completed the interview. Some 33.5% of respondents reported at least one lifetime disorder, 20.6% reported at least one-year disorder, and 16.7% reported at least one-month disorder. The most common lifetime disorders were alcohol abuse/dependence (17.24%), nicotine dependence/withdrawal (11.19%), specific phobia (5.16%), and major de-pressive disorder (4.25%). The lifetime prevalence of substance abuse/dependence (0.25%) and schizophrenia (0.16%) was very low. Nicotine and alcohol use disorder showed very high male/female ratio. Mood disorder and anxiety disorder were more prevalent among female than male. Conclusion : The prevalence of psychiatric disorders was high. In comparison with other studies, remarkable differences in distributions of psychiatric disorders across the areas and times were observed.
Rheological Behavior of Polymer/Layered Silicate Nanocomposites under Uniaxial Extensional Flow

We investigated the rheological behaviors and orientation of three different types of layered silicate composite systems under external flow: microcomposite, intercalated and exfoliated nanocomposites. Rheological measurements under shear and uniaxial extensional flows, two-dimensional, small-angle X-ray scattering and transmission electron microscopy were conducted to investigate the properties, as well as nano- and micro-structural changes, of polymer/layered silicate nanocomposites. The preferred orientation of the silicate layers to the flow direction was observed under uniaxial extensional flow for both intercalated and exfoliated systems, while the strain hardening behavior was observed only in the exfoliated systems. The degree of compatibility between the polymer matrix and clay determined the microstructure of polymer/clay composites, strain hardening behavior and spatial orientation of the clays under extensional flow.

Objective The purpose of this study was to observe the natural course of remained untreated spondylolysis adjacent to previous fusion segments in patients with multi-level lysis and its clinical outcome. Methods Nineteen patients who underwent selective fusion of multi-level spondylolysis (MLS) at a single institute were enrolled. As a matched cohort for comparison, 19 patients who had single-level spondylolysis (SLS) and undergone single-level fusion with similar demographics and preoperative radiologic measurements as the MLS group were included. We evaluated the preoperative, postoperative, and last follow-up angular displacement and sagittal translation on dynamogram radiographs, and axial and radial pain using the visual analogue scale in both groups. We then compared the increment in radiological instability and clinical outcome between the MLS and SLS groups. Results There were no significant differences in patient demographics and preoperative radiological measurements, including disc degeneration, facet degeneration, lumbar lordosis, pelvic incidence, and sacral table angle between both groups. Both groups showed an increase in the average angular displacement and slippage during the final follow-up as compared to preoperative findings, but no significant difference was noticed between them. Both the MLS and SLS groups showed improvement in lower back pain and leg pain from before surgery, but with no statistical significance. Conclusion Selective fusion in patients with multiple spondylolysis can be an alternative surgical option without increasing the risk of adjacent segment degeneration under strict narrow indications. However, a thorough preoperative evaluation is needed to prevent early surgical failure.

NO released by myenteric neurons controls the off contraction induced by electrical field stimulation (EFS) in distal esophageal smooth muscle, but in the presence of nitric oxide synthase (NOS) inhibitor, L-NAME, contraction by EFS occurs at the same time. The authors investigated the intracellular signaling pathways related with G protein and ionic channel EFS-induced contraction using cat esophageal muscles. EFS-induced contractions were significantly suppressed by tetrodotoxin ($1\;{\mu}M$) and atropine ($1\;{\mu}M$). Furthermore, nimodipine inhibited both on and off contractions by EFS in a concentration dependent meaner. The characteristics of 'on' and 'off contraction and the effects of G-proteins, phospholipase, and $K^+$ channel on EFS-induced contraction in smooth muscle were also investigated. Pertussis toxin (PTX, a $G_i$ inactivator) attenuated both EFS-induced contractions. Cholera toxin (CTX, $G_s$ inactivator) also decreased the amplitudes of EFS-induced off and on contractions. However, phospholipase inhibitors did not affect these contractions. Pinacidil (a $K^+$ channel opener) decreased these contractions, and tetraethylammonium (TEA, ${K^+}_{Ca}$ channel blocker) increased them. These results suggest that EFS-induced on and off contractions can be mediated by the activations Gi or Gs proteins, and that L-type $Ca^{2+}$ channel may be activated by G-protein ${\alpha}$ subunits. Furthermore, ${K^+}_{Ca^-}$ channel involve in the depolarization of esophageal smooth muscle. Further studies are required to characterize the physiological regulation of $Ca^{2+}$ channel and to investigate the effects of other $K^+$ channels on EFS-induced on and off contractions.
