Background: Recombinant IFN-beta products have been approved for the treatment of several diseases. Recently, Inbionet, Co. has successfully developed rhIFN-beta product, Genebeta. This study was performed in a purpose of confirming the biological activity of Genebeta by comparison with another recombinant IFN-beta, Beneserin which was already approved. Methods: In vitro antiporliferative activity, regulation of MHC class I expression, antiviral activity, regulation of natural killer cell cytotoxicity were studied for functional comparison of these two rhIFN-beta products. Results: Proliferation assay data showed that two recombinant IFN-beta inhibited T cell and Daudi cell proliferation to a similar extent. When antiviral activity was compared, there was no significant difference between them. The treatment of these two rhIFN-beta also resulted in a similar level of upregulation of class I expression on the surface of A549 cells. Cytotoxicity experiment against tumor cells revealed that both of rhIFN-beta could enhance NK cell cytotoxicity to comparable level. Conclusion: All comparison data indicated that rhIFN-beta products, Genebeta and Beneserin have comparable potentials as a biological response modifier.
Pulsed Nd:YAG 레이저 조사와 불소 화합물 도포가 초기 우식 법랑질의 표면 경도에 미치는 영향

This study was designed to determine the most effective concentration of fluoride and energy density of laser irradiation for the anticarious effect. For this study, surface hardness in enamel was measured before and after irradiation with pulsed Nd : YAG laser and the topical application of fluoride. Of the permanent mandibular anterior bovine teeth, healthy, carious free ones were used. Three hundred specimens were made. Specimens within 25~45 Vickers hardness numbers were assigned to 20 control and experimental groups : each containing 15 specimens. After forming artificial carious lesions, 10 J/㎠, 20 J/㎠, and 30 J/㎠ energies were irradiated on the enamel surface of each experimental group. Also NaF, NH_4F, Elmex gel(amine fluoride) and APF gel fluoride compounds were applied topically. Next, all the specimens were placed into the pH circulatory procedures for eight days. Vickers hardness numbers were measured using a microhardness tester. Surface changes of the enamel were observed using an scanning electron microscope. The comparative analysis yielded the following results : 1. The reduction of surface hardness of the enamel surface was less in all groups with fluoride application than in the group without fluoride application. 2. The APF gel group with 10 J/㎠ irradiation showed the lowest reduction of surface hardness. 3. The reduction of surface hardness of the enamel surface in the group of laser irradiation without fluoride application not showed any significant difference according to the energy density of the laser. 4. Under the scanning electron microscope, in enamel irradiated with 10 J/㎠ showed appearance similar to acid etching surface. In enamel irradiated with 20 J/㎠, fine enamel crack was detected. In enamel irradiated with 30 J/㎠, severe enamel crack and fusion of enamel were detected. These results suggest that one could obtain the best anticariogenic effects without damage to teeth in the group of application of APF gel after laser irradiation with 10 J/㎠.
Reduced Graphene Oxide Nanosheet for Chemo-photothermal Therapy

<P>The protein-functionalized reduced graphene oxide (rGO) nanosheet is of great interest in stimuli-responsive drug delivery and controlled release applications. We developed doxorubicin (DOX)-loaded bovine serum albumin (BSA)-functionalized rGO (DOX-BSA-rGO) nanosheets. To investigate the reduction of BSA-functionalized GO nanosheets and drug loading efficiency, we used X-ray photoelectron spectroscopy (XPS) and UV-visible spectrophotometer analysis. DOX-BSA-rGO nanosheets exhibited dose-dependent cellular uptake without any cytotoxic effect. We also demonstrated near infrared (NIR)-induced chemo-photothermal therapy of brain tumor cells treated with DOX-BSA-rGO nanosheets. Therefore, this DOX-BSA-rGO nanosheet could be a powerful tool for chemo-photothermal therapy applications.</P>
HS-1200 Overcomes the Resistance Conferred by Bcl-2 in Human Leukemic U937 Cells

Bcl-2 protects tumor cells from the apoptotic effects of various anti-neoplastic agents. Increased expression of Bcl-2 has been associated with a poor response to chemotherapy in various malignancies, including leukemia. Hence, bypassing the resistance conferred by anti-apoptotic factors such as Bcl-2 represents an attractive therapeutic strategy against cancer cells, including leukemic cells. This study was undertaken to examine whether the anticancer drug, cisplatin and the synthetic chenodeoxycholic acid (CDCA) derivative, HS-1200 show anti-tumor activity in U937 and U937/Bcl-2 cells. Viability assays revealed that HS-1200 overcomes the resistance conferred by Bcl-2 in human leukemic U937 cells. Various apoptosis assessment assays further demonstrated that HS-1200 overcomes the resistance conferred by Bcl-2 in human leukemic U937 cells by inducing apoptosis. In addition HS-1200, but not cisplatin, overcomes the anti-apoptotic effects of Bcl-2 in Bcl-2 over-expressing human leukemic cells (U937/Bcl-2 cells). Notably, we observed that the HS-1200-induced formation of mature promyelocytic leukemia (PML) nuclear bodies (NBs) correlates with a suppression of the anti-apoptotic effects of Bcl-2 in human leukemic cells over-expressing this protein (U937/Bcl-2 cells). Furthermore, HS-1200 was found to induce the association between PML and SUMO-1, Daxx, Sp100, p53 or CBP in the aggregated PML-NBs of U937/Bcl-2 cells. Thus, PML protein and the formation of mature PML-NBs could be considered as therapeutic targets that may help to bypass the resistance to apoptosis conferred by Bcl-2. Elucidating the exact mechanism by which PML regulates Bcl-2 will require further work.

Ability of water extract from Prunella vulgaris Labiatae to stimulate immune system and inhibit tumor metastasis in mice was assessed. In experimental lung metastasis, prophylactic intravenous (i.v.) administration of water extract from P. vulgaris significantly inhibited lung metastasis in a dose-dependant manner. Peritoneal macrophages stimulated with P. vulgaris produced various cytokines such as tumor necrosis factor (TNF)-α and interlukin (IL)-12 as well as induced tumoricidal activity. In an assay for natural killer (NK) cell activity, i.v. administration of P. vulgaris significantly augmented NK cytotoxicity. The depletion of NK cells by injection of rabbit anti-asialo GM1 serum abolished the inhibitory effect of P. vulgaris on lung metastasis of colon26-M3.1 cells. These data demonstrate that P. vulgaris activate innate immune system to inhibit the growth of foreign materials including tumor cells in mice.
Preparation of Low-Temperature Fired PZT Thick Films on Si by Screen Printing

Piezoelectric powder with the composition of PbTiO$_3$-PbZrO$_3$-Pb(Mn$\_$1/3/Nb$\_$2/3/)O$_3$ and small particle size of 0.3 $\mu\textrm{m}$ was investigated for low-temperature firing of PZT thick films. PbTiO$_3$-PbZrO$_3$-Pb(Mn$\_$1/3/Nb$\_$2/3)O$_3$ ceramics showed dense microstructure and superior piezoelectric properties, electromechanical coupling factor (k$\_$p/) of 0.501 and piezoelectric constant (d$\_$33/) of 224. The PZT paste was made of the powder and organic vehicles, and screen-printed on Pt(450nm)/YSZ(110nm)/SiO$_2$(300nm)/Si substrates and fired at 800∼900$^{\circ}C$. Any interface reaction between the PZT thick film and the bottom electrode was not observed in the PZT thick films. The PZT thick film fired at 800$^{\circ}C$ showed moderate electrical properties, the remanent polarization(p$\_$r/) of 16.0 ${\mu}$C/$\textrm{cm}^2$, the coercive field(E$\_$c/) of 36.7 ㎸/cm, and dielectric constant ($\varepsilon$$\_$r/) of 531. Low-temperature sinterable piezoelectric composition and high activity of fine particles reduced the sintering temperature of the thick film. This PZT thick film could be utilized for piezoelectric microactuators or microsensors that require Si micromachining technology.
