Proceedings of International Symposium on East-West Medicine, Seoul. 212-230, 1999. -Hepatitis B is caused by hepadnavirus. Hepatitis B virus replicates through 3.5kb pregenomic RNA intermediate which is regulated by core promoter. Pathogenesis of hepatitis B virus has been bilieved the result of host immune response. But recently many studies have reported that high level of viral replication caused by mutation in core promoter might result in severs hepatitis. But these studies were performed in vitro, not in vivo. So there is yet debate about which factor, viral of host factor, is more important in pathogenesis of hepatitis B virus. So we measured real viral replication level in 204 chronic hepatitis B patients by quantifying HBV DNA from sera by our novel PCR-based more sensitive method, and compared these results with ALT level measured from same sera, which indicates liver cell damage. Surprisingly there are no significant correlation between HBV DNA quantity and ALT level. Then we cloned core promoter region. In SSCP, we found that many viral mutants coexist in one patient. Base on SSCP result, we chose main viral core promoter type in each patients, which is thought to determine overall viral replication level in this patient. Main type of core promoter region of each 41 patients were directly sequenced. And with these we measured promoter activity by luciferase assay system and compared promoter activity with on another. We found tha there were some differences in promoter activity according to core promoter sequences. And we constructed replication-competent viral constructs with core promoter from 41 patients and Transfected these into HepG2 cell and measured HBV DNA by southern blot. There were also differences in HBV DNA quantity according to core promoter sequences. On these all results we investigated correlation between the effect of HBV core promoter on viral replication in vitro and HBN DNA quantity, ALT level from sera of each patients. We found there is no significant correlation among them. As a result, we concluded that in determining severity chronic hepatitis B patients, host factors of each patient is more important rather than replicative activity of virus itself.

      • 초등학교 체력검사 제도에 관한 교사의 인식도 연구

        This study is to analyze the actual condition of examination of physical strength for elementary students, understand the problems and present materials for improvement 417 persons are selected optionally for this study, who are 346 elementary teachers, 28 public officials, 33 managers for physical education, and 10 physical education teachers, and their education carieer, level급지and scale of each school and their sexes at also ignored. The research was carried out by questionnaire which asked about 7 domains, which were teacher's opinion about elementary school's examination of physical strength, its contribution, appropriateness of examinatorial items, deletion and addition of items, items' relianc, fair administration, and previous questions for effective management Questions were determined in the background of previous studied, and theory, and also examined by a professional adviser. Answering to questions took 5 steps responsive method and description together. In the course of analysis, no answering was excluded, and the significance of probability frequency distribution was tested by Chi-Square Test(Expected or Equal) to check out answering characteristics, while significance level was over 95%. After all, I carry to the following conclusion. 1. Objects were affirmative about the need of examination of physical stength and the examinatorial grade's reflection to physical education's pactical grade and they also agreed to reflect the grade with 50% below, practice the test in September in common currency. They thought the testing criterion in each item is unsuitable. 2. Physical examintion contributes to understand his(or her) own physical strength, but it doesn't stimulate the recognition of physical activities and motives effectively. Its contribution is low to the tested person's physical strength, improvement of health and physical condition, the qualitative advancement of school work, participations to social physical activities, and betterment of leisure time. 3. The appropriateness of 100m run, raising up upper body and 600m run is high, that of chinning exercise, chin-up with the arms crooked is above normal, but the adquacy of jump from the original place is normal, that of throwing is beneath normal. 4. From their responses, the current number of physical examination's item is adequate, and they recommended items for fexibility, balance as additional items, while pointing 600m long run, chinning excercise(chin-up with the arms crooked) as eliminative items. 5. What has the best reliance in elementary physical examination is raising up upper body and then 100m run, and the most important factor to decrease reliance is bad measuring methods. The reliance of testing facility is low. 6. Objects think that inspectors make fair evaluation, but to minute, standardize the facility and norm is required urgently and the view to tighten supervisory system is dominant. 7. The previous tasks for effective running of elementary physical examination system is to repair facilities and they suggested developing objective measuring methods, replenishing equipments and practice in advance.

        논문 : 수위-유량 관계곡선을 이용한 수막재배용 지하수 사용량 추정

        수위-유량 관계곡선을 이용한 동절기 수막재배기간의 지하수 사용량 추정 방법을 제시하고, 이를 논산 왕전리 수막재배 지역에 현장 적용하였다. 연구지역의 동절기 수막재배 기간의 농수로수의 구성성분은 주로 지하수와 강우의 직접 유출 성분으로 구성된다. 농수로 수위를 관측하고, 강우에 의한 농수로 수위 상승분을 수위 관측 자료로 부터 제거시키기 위해 기저유출분리법을 적용하였다. 강우에 의한 수위 상승분이 제거된 농수로 수위자료에 수위-유랑 관계를 적용하여 연구지역의 수막재배용 지하수 배출량을 산출하였다. 왕전리 지역의 수막재배용 지하수 사용량은 평균 10,900 m3/d, 420m3/d/ha인 것으로 추정되었다. 추정 지하수 사용량(420 m3/d/ha)과 전국의 수막재배면적(10,746 ha), 그리고 연간 수막재배일수(120 days/year)을 기준한 전국 규모의 수막재배용 지하수 사용량은 5억 4천만 m3에 이른다. 이는 농업용 지하수연간 사용량(약 17 억 m3)의 약 32%에 달한다. A method is proposed to estimate groundwater usage for water curtain cultivation (WCC) using a rating curve, and it is applied to field measurements of groundwater discharge used for WCC in Wangjeon-ri, Nonsan. During the winter season, the hydraulic components of irrigation ditches in the study area consist mainly of direct run-off and groundwater discharged from nearby pumping wells. Changes in stage of the ditches were monitored, and a baseflow separation method was applied to remove increments in stage due to direct run-off. The resulting records of stage were translated to groundwater discharge by applying the-stage-discharge relation. The estimated average groundwater discharge for the WCC in Wangjeon-ri was 10,900 m3/d or 420 m3/d/ha when the estimation is normalized by the total area for WCC facilities of this region. Applying this estimation (420 m3/d/ha) to the entire area of the WCC in Korea (10,746 ha),and considering the number of pumping days for the WCC (120 days/year), the total ground water usage for the WCC nation-wide is estimated to be 0.54 billion m3. This is equivalent to 32% of the total groundwater discharge for agricultural use in Korea (1.7 billon m3).

      • Effects of Amino Acid Addition on Molar Fraction of 3-Hydroxyvalerate in Copolyester of 3-Hydroxybutyrate and 3-Hydroxyvalerate Synthesized by Alcaligenes sp. SH-69

        Addition of the amino acids threonine, isoleucine, and valine into culture medium greatly increased the molar fraction of 3-hydroxyvalerate(3HV) in the copolyester of 3-hydroxybutyrate (3HB) and 3HV produced by Alcaligenes sp. SH-69. In shake flask cultures, addition of threonine, isoleucine, or valine into culture mediumresulted in 53.0, 40.7, or 14.8 mol% 3HV in the copolyester, respectively. The 3HV content in the copolyester was 14 mol% and 27 mol% in jar fermentor cultures containing 10 mM and 20 mM threonine, respectively. Similar molar freactions of 3HV(11-14 mol%) in the copolyester were obtained in experiments carried out with culture media containing 10 mM isoleucine or valine. The 3HV content in the copolyester increased as aminoacid concentration increased. Addition of the amino acids into culture medium at different culture times revealed that final 3HV contents were ismilar irrespective of addition time except for the case when glucose was depleted.

      • Thymosin 관4 expression correlates with lymph node metastasis through hypoxia inducible factor-관 induction in breast cancer.

        <P>Intratumoral hypoxia has been correlated with distant metastatic potential. Two hypoxia inducible factors (HIFs), HIF-1관 and HIF-2관, are induced by hypoxia, and high expression of these proteins has been correlated to angiogenesis and distant metastasis. Thymosin 관4 (T관4) is frequently highly expressed in cancer, and this overexpression correlates with malignant progression. The objective of this study was to investigate the clinical correlation of HIF-관 with T관4 and the intracellular functional roles of T관4 on HIF-관 activation. We examined HIF-1관, HIF-2관 and T관4 expressions in clinical human breast carcinoma (n=70) by immunohistochemistry. We show that high expression of HIF-1관 and HIF-2관 strongly correlates with T관4 expression (P??.0001) and overexpression of T관4 correlates significantly with patients with lymph node metastasis (P<0.05) of human breast cancer. Additionally, we demonstrate that hypoxia up-regulates intracellular T관4 protein, which then affects HIF-관 activity, which is the key in regulating VEGF expression. We confirmed that hypoxia-induced intracellular T관4 and HIF-관 activities were reduced by interference of T관4 expression using T관4 shRNA lentivirus. Vascular epidermal growth factor (VEGF)-A, a well-recognized lymphangiogenic cytokine, was also down-regulated, but VEGF-C and VEGF-D expressions were not affected. These findings suggest that the overexpression of T관4 is strongly associated with HIF-1관 and HIF-2관 expression and is also clinicopathologically involved with lymph node metastatic potential of breast cancer through the modulation of HIF-관activation and induction of VEGF-A. Ultimately, these results highlight T관4 as a potentially therapeutic target in malignant cancers.</P>
