No effective antiviral therapy is proven yet, but clinical use of ribavirin (RBV) has been tried. We investigated the antiviral effect of RBV against SFTSV in vitro. Methods: To test for cytotoxicity of RBV, Vero cells were treated with different concentrations of RBV (3.90 to 500 μg/mL, two-fold dilution) and analyzed by cell viability MTS assay 48h post-infection. To determine antiviral activity of RBV against SFTSV, Vero cells were infected with SFTSV strain Gangwon/Korea/2012 at 100 TCID50 (50% tissue culture infective dose) per well in a 96-well plate, and RBV was added at the concentrations showing no or minimal cytotoxicity. Viral RNAs were extracted from the culture supernatants and quantifi ed using one-step real-time reverse transcription- PCR to amplify the partial large segment of SFTSV. Statistical analysis was done by one-way ANOVA with Tukey`s post hoc test. Results: Cytotoxicity due to RBV was not observed at RBV concentration =31.3 μg/ mL. Viral RNAs at 24h post-RBV treatment were reduced with increasing RBV concentrations (1-32 μg/mL), compared with those of mock-treated cells (P <0.01, Figure). Half maximal inhibitory concentration (IC50) of RBV was 3.69 μg/mL at 24h post-RBV treatment. Conclusions: Our study shows that RBV has antiviral effect against SFTSV in a dose-dependent manner. Further studies are required to evaluate the effi cacy of RBV in SFTS.

        Discordance in Spirometric Interpretations Based on Korean and Non-Korean Reference Equations

        Objectives: Korean regression models for spirometric reference values are different from those of other ethnic groups. The purpose of this study was to evaluate discordance in spirometric interpretations based on using Korean reference equations versus non-Korean reference equations. Methods: Spirometry was performed on 825 Korean male workers from April 2009 to November 2011. The spirometric patterns and disease severity were evaluated using two Korean equations (Choi’s and Lee’s) and three equations for Caucasians (NHANES III, Morris’s, and Knudson’s), and the results of Choi’s equation were compared with the non-Korean equations. The spirometric patterns were defined as normal, restrictive, and mild and moderate obstructive. Results: The mean differences in the FEV1% and FVC% between the two Korean equations were 2.0 ± 1.3% and 3.5 ± 2.2%, respectively. Morris’s equation had the greatest difference in the FEV1% from Choi’s equation: 32.9 ± 8.5%. Knudson’s equation had the greatest difference in the FVC% from Choi’s equation: 10.5 ± 6.5%. Conclusions: The two Korean equations shared similar characteristics of spirometric interpretation. The spirometric interpretations of Choi’s equation were significantly different from those of Morris’s equation and Knudson’s equation.

      • Cytotoxic and Antitumor constituens from Korean medicinal plants

Page 153-161, 1999. -For the screening of bioactive natural products, the benzene or methanol extracts from 93 medicinal plants of Korea were prepared, and tested for the cytotoxicity against L1210 cells and for the antitumor action (Bae et al., 1992 and 1996). Of 93 extracts tested, 6 samples showed a cytotoxicity in both benzene and methanol extract, 39 samples in benzene and 13 samples in methanol extract. The benzene extract of the root of scutellaria indica L., Carpesium abrotanoides L., Gymnaster koraiensis(Nakai) Kitamura, Pyrola japonica Klenze, Forsythiae Fructus, and Alismatis Rhizoma showed a potent cytotoxic activity. This observation led to isolate active cytotoxic components, some of which demonstrated some antitumor action. In addition, the structure-activity relationship was discussed.

        Breeding of New Ever-bearing Strawberry “Doha” Variety

        “Doha” is a new strawberry (Fragaria x ananassa Duch.) variety, which was released by the Highland Agriculture Research Institute in 2021. The “Doha” variety originates from a 2014 cross between “Saebong No. 3” and “Yeolha,” both of which exhibited excellent ever-bearing characteristics, including continuous flowering and large fruits under long-day and high temperature conditions. This new cultivar was initially named “Saebong No. 13” after examining its characteristics and productivity during summer cultivation between 2015 and 2018. After regional adaptability tests, “Doha” was selected from “Saebong No. 13” as an elite cultivar. The general characteristics of “Doha” include spreading, elliptic leaves, and strong growth. The fruits are long and conical and of a red color. The plant height of “Doha” was similar to that of “Goha,” but the number of leaves was lower. The number of flower clusters of “Doha” was 8.6, which was 2.8 fewer than that of the control variety, “Goha,” with 11.4. The average fruit weight of “Doha” was 13.9 g, which was 4.9 g heavier than that of “Goha.” The fruit hardness of “Doha” was 35.5 g·㎜−2, which was 9.4 g·㎜−2 harder than that of “Goha.” The marketable yield of “Doha” was 26,971 ㎏·㏊−1, 125% more than that of “Goha” with 21,479 ㎏·㏊−1. The findings of this study suggest that “Doha” is a hard fruit and high-yielding variety of ever-bearing strawberries that could increase farming income when distributing to farmers.

      • Blood pressure–targeted stepwise resuscitation for hemorrhagic shock in rats

        BACKGROUND: Generation of reactive oxygen species (ROS) is an important mechanism of ischemia-reperfusion injury. Abrupt reoxygenation compared with slow reoxygenation has been known to increase ROS generation. Thus, slow and stepwise reperfusion can reduce ROS generation and subsequent ischemia-reperfusion injury. This study investigated the effect of slow reperfusion by blood pressure–targeted stepwise resuscitation (PSR) in hemorrhagic shock. METHODS: Pressure-controlled hemorrhagic shock was induced in male Sprague-Dawley rats for 1 hour. Rats were then allocated to one of three groups (no-resuscitation group, n = 14; PSR group, n = 15; rapid normalization of blood pressure (RR) group, n = 15). Survival time and hemodynamic changes were recorded and compared. Blood samples and liver tissue were harvested after 6 hours of resuscitation in surviving rats. RESULTS: All of the rats in the no-resuscitation group were expired before the end of the 6-hour observation period. Survival times were significantly longer in the PSR group than in the RR group (survival rates, 11 of 15 vs. 5 of 15, log rank p = 0.032). Plasma amino alanine transferase, histologic liver injury, and ROS generation in the liver tissue were significantly lower in the PSR group than in the RR group (all findings significant, p < 0.05). In addition, PSR significantly decreased plasma nitric oxide, liver interleukin 1&bgr;, and liver interleukin 6 compared with rapid resuscitation in addition to augmenting Akt survival pathways (all p < 0.05). CONCLUSION: Slow reperfusion by PSR decreased mortality, ROS generation, and liver injury in rats undergoing hemorrhagic shock. Stepwise resuscitation also decreased inflammatory cytokine production and augmented Akt survival pathways.

      • Mesocorticolimbic hyperactivity of deprived smokers and brain imaging

        We determined the association of neuronal circuitry with the desire to smoke by acquiring and analyzing functional MRI data. The data were acquired in both abstained and subsequently satiated (by ‘natural’ cigarette smoking) heavy smokers and also in demographically and intellectually matched nonsmokers. During the acquisition, participants were viewing alternating smoking and nonsmoking images that were interleaved by fixation images. From the results, the activities in the mesocorticolimbic pathway including the orbitofrontal cortex, parahippocampus, hippocampus, and midbrain were significantly negatively correlated with carbon monoxide (CO) levels. In contrast, the activities in the motor area and the posterior cingulate cortex plus precuneus were significantly positively correlated with the CO levels. This is the first study to show that mesocorticolimbic and midbrain activities are strongly associated with CO levels, and therefore, possibly with smoking desire levels because of the strong correlation between CO levels and blood nicotine levels.

        Is the Agricultural Work a Risk Factor for Koreans Elderly Spinal Sagittal Imbalance?

        Objective : A primary degenerative sagittal imbalance has been considered because of unique lifestyles such as the prolonged crouched posture during agricultural work and performing activities of daily living on the floor. Previous papers have reported that sagittal imbalance disease is often seen distinctly in the farming districts of “oriental” countries such as Korea and Japan. However, this finding was only evaluated with the use of X-ray, and other factors such as magnetic resonance imaging (MRI), muscle volume, compression fracture, and laboratory results were not considered. Thus, using these, we evaluate the agricultural work-associated factors for Korean elderly spinal sagittal imbalance. Methods : We recruited 103 Korean participants who had a sagittal vertical axis (SVA) of >5 cm in this Korean Elderly Sagittal Imbalance Cohort Study. The following were evaluated : radiological parameters, MRI, compression fracture, vitamin D, parathyroid hormone, C-terminal telopeptide, osteocalcin, bone mineral density and muscle fatty change, muscle volume, and health-related quality of life from patients’ survey. Moreover, in this survey, the farmers’ annual working hours were investigated. Subsequently, we analyzed the associated factors for spinal sagittal imbalance depending on occupation. Results : A total of 46 participants were farmers, and the others were housewives, sellers, and office workers. The farmer group had more SVA (141 vs. 99 mm, p=0.001) and pelvic tilt (31° vs. 24°, p=0.004) and lesser lumbar lordosis (20° vs. 30°, p=0.009) and thoracic kyphosis (24° vs. 33°, p=0.03) than non-farmer group. A significantly positive correlation was noted between the working hour and SVA in the farmer group (p=0.014). The visual analogue scale score for back pain (8.26 vs. 6.96, p=0.008) and Oswestry Disability Index (23.5 vs. 19.1, p=0.003) in the farmer group were higher than that in the non-farmer group, but the Short Form-36 score was not significantly different between the two groups. The Mini-Mental State Exam score was significantly lower in the farmer group than in the non-farmer group (24.85 vs. 26.98, p=0.002). Conclusion : The farmer group had more sagittal imbalance and back pain in proportion to the working hours even though the muscle and bone factors and general laboratory condition were not significantly different between the two groups. These results supported that the long hours spent in the crouched posture while performing agricultural work were a risk factor for severe sagittal imbalance.

        Yellow-colored mats in Jeju Island lava tubes

        The Geomunoreum Lava Tube System, declared as a UNESCO World Heritage Site, has a unique natural ecosystem. The information available about this ecosystem, which contains lava caves with secondary carbonate speleothems, is sparse. Hence, extensive research is warranted for establishing a conservation standard. We commenced microbial research on the system and have been studying the microorganisms coating the lava tube wall to acquire fundamental information for understanding the lava cave ecology of Jeju Island. Samples were collected from yellow-colored walls in six caves that are part of the system-the Bengdwi, Utsanjeon, Bukoreum, Manjang, Gimnyeong, and Yongcheon caves. This study focused on yellow walls as it is the most easily distinguished color. According to previous studies, the color of cave walls is attributed to microorganisms or their components. To determine whether the yellow mats from the Jeju lava tube walls are caused by microorganisms, we examined samples at the microscopic scale, by staining mats and analyzing bacterial isolates from glitter particles. As a result, we found that the yellow walls of lava tubes are comprised of microbial mats.
