        Adenosine A3 Receptor Mediates ERK1/2- and JNK-Dependent TNF-α Production in Toxoplasma gondii-Infected HTR8/SVneo Human Extravillous Trophoblast Cells

        Toxoplasma gondii is an intracellular parasite that causes severe disease when the infection occurs during pregnancy. Adenosine is a purine nucleoside involved in numerous physiological processes; however, the role of adenosine receptors in T. gondii-induced trophoblast cell function has not been investigated until now. The goal of the present study was to evaluate the intracellular signaling pathways regulated by adenosine receptors using a HTR-8/SVneo trophoblast cell model of T. gondii infection. HTR8/SVneo human extravillous trophoblast cells were infected with or without T. gondii and then evaluated for cell morphology, intracellular proliferation of the parasite, adenosine receptor expression, TNF-α production and mitogen-activated protein (MAP) kinase signaling pathways triggered by adenosine A3 receptor (A3AR). HTR8/SVneo cells infected with T. gondii exhibited an altered cytoskeletal changes, an increased infection rate and reduced viability in an infection time-dependent manner. T. gondii significantly promoted increased TNF-α production, A3AR protein levels and p38, ERK1/2 and JNK phosphorylation compared to those observed in uninfected control cells. Moreover, the inhibition of A3AR by A3AR siRNA transfection apparently suppressed the T. gondii infection-mediated upregulation of TNF-α, A3AR production and MAPK activation. In addition, T. gondii-promoted TNF-α secretion was dramatically attenuated by pretreatment with PD098059 or SP600125. These results indicate that A3AR-mediated activation of ERK1/2 and JNK positively regulates TNF-α secretion in T. gondii-infected HTR8/SVneo cells.

        A pilot study of gonadotropin-releasing hormone agonist combined with aromatase inhibitor as fertility-sparing treatment in obese patients with endometrial cancer

        Objective: This study aims to evaluate the effects and pregnancy outcomes of gonadotropin-releasing hormone agonist (GnRH agonist) combined with aromatase inhibitor (AI) in preserving the fertility of obese women with grade 1 endometrial cancer (EC). Methods: This study recruited obese EC patients who wished to preserve their fertility. The treatment regimen consisted of intramuscular GnRH agonist 3.75 mg every 4 weeks and oral AI 2.5 mg daily. The maintenance regimen was the same as the initial treatment regimen. Primary outcomes included response rate, time to complete response (CR), and time to recurrence; pregnancy outcomes included the time to pregnancy, pregnancy rate and live birth rate. Results: Six obese patients with EC were included in this study, with the age (mean±standard deviation [SD]) of 30.5±3.3 years and body mass index (mean±SD) of 35.0±1.4 kg/m2. CR rate was 100%, and time to CR was 3–6 months. None of the patients had recurrence after a median follow-up of 4.0 years (range, 1.3–7.0 years). The most common side effects were menopause-like symptoms. Among these patients, no weight gain was observed during treatment. The pregnancy rate and live birth rate was 50.0% and 75.0%, respectively, with a median time to pregnancy of 2.4 years (range, 1.0–5.5 years). Conclusion: The combination of GnRH agonist and AI demonstrated promising long-term effect in young obese EC patients who wished to preserve their fertility. No weight gain side effects were observed. Further studies with a larger sample size are needed to fully evaluate this novel treatment regimen.

        Effect of Co-doping on structural, magnetic and magnetocaloric properties of La0.67Ca0.13Ba0.2Mn1-xCoxO3 (x = 0, 0.02, 0.04, 0.06, 0.08, 0.1) manganites

        The bulk polycrystalline samples of La0.67Ca0.13Ba0.2Mn1-xCoxO3 (x=0, 0.02, 0.04, 0.06, 0.08, 0.1) have been prepared by conventional solid-state reaction method. X-ray powder diffraction shows that all samples crystallize in the rhombohedral structure with R3¯c space group symmetry. The ratio of Co3+/Co4+ gradually increases with increasing Co doping composition, which was determined by XPS analysis. The temperature and applied magnetic field dependence of magnetization were carried out. All samples exhibit a FM-PM transition at the Curie temperature TC, which decreases from 312 K to 223 K with increasing Co composition. Critical analysis shows that the estimated critical exponents β, γ and δ are in between the theoretically predicted values for 3DHeisenberg and mean-field theory models for all samples. The magnetocaloric effect has been calculated from isothermal magnetization curve at various temperatures near TC. The maximum magnetic entropy changes -ΔSmax under applied magnetic field of 1.6 T were found to be 3.18 J kg−1 K−1, 2.47 J kg−1 K−1 and 2.68 J kg−1 K−1 respectively for x=0, 0.02 and 0.04 composition, which will be an interesting compound for application materials working as magnetic refrigerants near room temperature.

        First principles studies on the elastic, thermodynamic properties and electronic structure of Ti15−xMoxSn compounds

        The structural, elastic, thermodynamic and electronic properties of the Ti15−xMoxSn compounds were systematically investigated by means of first-principles calculations based on the density functional theory (DFT). The calculated results demonstrate the Ti15−xMoxSn compounds still remain the stable β phase structure. The calculation of cohesive energy shows that the structural stability of the Ti15−xMoxSn compounds increases apparently with the increase of Mo content. According to Hooke's law, the single crystal elastic constants were obtained and show that all the calculated compounds keep mechanical stability. Then the bulk modulus B, shear modulus G, Young's modulus E and Poisson's ratio ν of polycrystalline aggregates were calculated at zero pressure. The calculated results show that among these Ti15−xMoxSn compounds, Ti4Mo11Sn exhibits the largest stiffness while Ti12Mo3Sn shows the greatest ductility. The compounds Ti12Mo3Sn and Ti11Mo4Sn with the two lowest elastic Young's modulus of 61.01 GPa and 65.59 GPa are expected to be promising metallic biomaterials for implant applications. Besides, the Debye temperature ΘD and the electronic density of states (DOS) are also investigated and discussed.
