Promyelocytic leukemia (PML) gene, through alternative splicing of its Cterminal region, generates several PML isoforms that interact with specific partners and perform distinct functions. The PML protein is a tumor suppressor that plays an important role by interacting with various proteins. Herein, we investigated the effect of the PML isoforms on oncostatin M (OSM)-induced signal transducer and activator of transcription-3 (STAT-3) transcriptional activity. PML influenced OSMinduced STAT-3 activity in a cell type-specific manner, which was dependent on the p53 status of the cells but regardless of PML isoform. Interestingly, overexpression of PML exerted opposite effects on OSM-induced STAT-3 activity in p53 wild-type and mutant cells. Specifically, overexpression of PML in the cell lines bearing wild-type p53 (NIH3T3 and U87-MG cells) decreased OSM-induced STAT-3 transcriptional activity, whereas overexpression of PML increased OSM-induced STAT-3 transcriptional activity in mutant p53-bearing cell lines (HEK293T and U251-MG cells). When wild-type p53 cells were co-transfected with PML-IV and R273H-p53 mutant, OSM-mediated STAT-3 transcriptional activity was significantly enhanced, compared to that of cells which were transfected with PML-IV alone; however, when cells bearing mutant p53 were co-transfected with PML-IV and wild-type p53, OSM-induced STAT-3 transcriptional activity was significantly decreased, compared to that of transfected cells with PML-IV alone. In conclusion, PML acts together with wild-type or mutant p53 and influences OSM-mediated STAT-3 activity in a negative or positive manner, resulting in the aberrant activation of STAT-3 in cancer cells bearing mutant p53 probably might occur through the interaction of mutant p53 with PML.

Wind plays an important role in cases of unexpected radioactive pollutant dispersion, deciding distribution and concentration of the leaked substance. The accurate prediction of wind has been challenging in numerical weather prediction models, especially near the surface because of the complex interaction between turbulent flow and topographic effect. In this study, we investigated the characteristics of atmospheric dispersion of radioactive material (i.e. <sup>137</sup>Cs) according to the simulated boundary layer around the HANARO research nuclear reactor in Korea using the Weather Research and Forecasting (WRF)-Mesoscale Model Interface (MMIF)-California Puff (CALPUFF) model system. We examined the impacts of orographic drag on wind field, stability calculation methods, and planetary boundary layer parameterizations on the dispersion of radioactive material under a radioactive leaking scenario. We found that inclusion of the orographic drag effect in the WRF model improved the wind prediction most significantly over the complex terrain area, leading the model system to estimate the radioactive concentration near the reactor more conservatively. We also emphasized the importance of the stability calculation method and employing the skillful boundary layer parameterization to ensure more accurate low atmospheric conditions, in order to simulate more feasible spatial distribution of the radioactive dispersion in leaking scenarios.
3D Active Vessel Tracking Using an Elliptical Prior

<P>In this paper, we propose a novel vessel tracking method, called active vessel tracking (AVT). The proposed method retains the major advantages that most 2D segmentation methods have demonstrated for 3D tracking while overcoming the drawbacks of previous 3D vessel tracking methods. Under the assumption that the vessel is cylindrical, thereby making its cross-section elliptical, the AVT finds a plane perpendicular to the vessel axis while tracking the vessel along its length. Also, We propose a method for vessel branch detection to automatically track complete vascular networks from a single starting point, whereas the previously proposed solutions have usually been limited in handling vessel bifurcations precisely on 3D or have required considerable user interaction. Our results show that the method is robust and accurate in both synthetic and clinical cases. In an experiment on synthetic data sets, the proposed method achieved a tracking accuracy of 96.1±0.5, detecting 99.1% of the branches. In an experiment on abdominal CTA data sets, it achieved a tracking accuracy of 98.4±0.5 for six target vessels, detecting 98.3% of the branches. These results show that the proposed method can outperform previous methods for vessel tracking.</P>
Krill and Coconut Oil Attenuates Inflammation and ER Stress in Murine Adipocytes

Selection of which dietary fatty acids are crucial to prevent potential metabolic complications, such as CVD or obesity. Among various fatty acids, PUFAs consumption are highly important since PUFAs consumption is directly related to systemic and local inflammation. Obesity is termed as a low-grade inflammation and elevation of ER stress. In this study, Krill oil (KO) and coconut oil (CO) were treated to 3T3-L1 adipocytes to understand whether KO or CO could prevent adipocyte inflammation and ER stress. Both KO and CO treatment significantly attenuated LPS-inducible inflammation by suppressing phosphorylation of MAPK (i.e., p-JNK and p-ERK) in fully differentiated adipocytes. Moreover, KO remarkably attenuated LPS-inducible cytokine mRNA expressions (i.e., IL-6 and IL-1b). Moreover, both KO and CO also decreased thapsigargin inducible ER stress protein expressions (i.e., BiP and CHOP). To our surprise, CO prevented thapsigargin-inducible ER stress effectively in a lower concentration than KO. These results suggest that PUFAs may modulate adipocyte inflammation and ER stress in mammalian cells.
지방의회 예산·정책분석 업무지원시스템 사용 의도에 영향을 미치는 요인 연구: 수정된 기술수용모형 분석을 중심으로

지방의회의 전문성, 효율성 향상이 요구되며 예산·정책분석 업무지원시스템 도입이 제시되고 있다. 기술수용모델(TAM)은 새로운 기술이나 시스템 도입에 대한 이용자의 태도와 행위 같은 수용과정을 설명한 이론으로 현재까지 활용되고 있다. 민간부문에서는 회계시스템 및 정보시스템 도입에 TAM을 적용한 연구가 진행되었으나, 공공부문에서는 복식부기·발생주의 회계 도입 및 정보시스템 도입에 TAM을 적용한 연구만이 진행되었다. 본 연구는 예산·정책분석 업무지원시스템 도입 시 이용 요인을 알아보기 위해 지방의회 예산·정책 분석 업무 관련자를 대상으로 설문조사를 진행한 후 수정된 기술수용모델(mTAM)을 이용하여 구조모형을 실시하였다. 분석 결과, 개인의 혁신성은 지각된 용이성과 지각된 유용성에 영향을 미치는 것으로 나타났으며, 지각된 유용성은 지각된 용이성에 영향을 미치는 것으로 나타났다. 또한, 지각된 용이성, 지각된 유용성, 개인적 혁신성, 모두 수용의도에 영향을 미치는 것으로 나타났다. 이상의 연구결과의 시사점은 다음과 같다. 첫째, 원활한 시스템 도입을 위해서는 인식 및 개선되는 기능에 대한 교육지원이 필요하다. 둘째, 타 기관에서 운영 중인 시스템과의 데이터 연계·활용이 용이하여야 한다. 셋째, 시스템은 이용자의 전문성 및 특성을 고려해야 하고, 지속적인 시스템 관리가 진행되어야 한다. 본 연구는 지방자치단체 예산·정책분석 업무지원시스템의 원활한 도입 및 활용에 이바지할 수 있을 것이라 생각된다. Improvements in the expertise and efficiency of local councils are required, and the introduction of a budget and policy analysis support system is being proposed. The Technology Acceptance Model (TAM) is a theory that explains the acceptance process, such as users’ attitudes and behaviors toward the introduction of new technologies or systems, and is still used to this day. In the private sector, research was conducted on applying TAM to the introduction of accounting systems and information systems, but in the public sector, only research was conducted on applying TAM to the introduction of double-entry bookkeeping and accrual accounting and information systems. This study employed the modified Technology Acceptance Model (mTAM) to conduct a structural model in order to identify factors influencing the usage of the budget and policy analysis support system upon its introduction. Surveys were conducted among budget and policy analysts for data collection. The analysis revealed that individual innovativeness affects perceived ease of use and perceived usefulness. Additionally, perceived usefulness influences perceived ease of use. Moreover, perceived ease of use, perceived usefulness, and individual innovativeness all impact the intention to use. The implications of the research results are as follows. Firstly, educational support on perception and enhanced functionalities is essential for the smooth adoption of the system. Secondly, seamless data integration and utilization with systems operated by other agencies are crucial. Thirdly, the system should consider users’ expertise and characteristics, and ongoing system management is necessary. The findings of this study are expected to contribute to the smooth adoption and utilization of the local government budget and policy analysis support system.
