노랑초파리 발생과정에서 sta 유전자의 발현 양상에 관한 연구

Drosophila stuvarista(sta) gene encodes the ribosomal protein D-p40, consisting of 270 amino acids. The D-p40 is highly conserved between such evolutionarily distinct organisms as human(63% identity) and hydra(58% identity) and has the effect of Minute phenotype such as other ribosomal proteins. We identified the sta gene in the process of Drosophila cDNA library screening and analyzed the developmental expression pattern of sta gene in the transcription level using in situ hybridization. Each animal stage and tissues of 3rd instar larva and adults were showed ubiquitously expression pattern. These results were similar to other ribosomal proteins that represented Minute phenotype. Also, we expressed the 31kDa recombinant proteins of D-p40 using his-tag expression system in E. coli and purified by affinity chromatography. Immunoblot result of the developmental stages was showedthat strongly detection only the stage of embryo and pupa. This result suggested that the expression of sta gene were regulated in the level of post-translation modification ailed considered that sta gene had to other function during development as well as to function as a core ribosomal component.

Chitosan is the deacetylated product of chitin. Chitosan and its derivatives have many properties that make them attractive for a wide variety of health applications. We reported recently that water soluble chitooligosaccharides have hepatoprotective effect on ethanol induced liver injury in the mouse. The current study examined the effect of water soluble chitooligosaccharides on CC1₄-induced liver injury in rats. Male Sprague-Dawley rats were orally treated with CC1₄without or with administration of 5% chitooligosaccharides. After 5 weeks after CC1₄treatment, the histological sections and the expression of mRNA of interleukin-12(IL-12), hepatocyte growth factor(HGF), and tumor necrosis factor(TNF- ) were examined. The liver section of rats supplemented with chitooligosaccharides showed attenuated effects of the necrosis, steatosis, cirrhosis compared with those of non-supplemented rats. The level of TNF- mRNA in CC1₄-treated rats were higher than those in the rats supplemented with chitooligosaccharides, while the levels of IL-12 and HGF mRNA were higher in the rats supplemented with chitooligosaccharides than those of in CC1₄-treated rats. These findings suggested that water soluble chitooligosaccharides may have a hepatoprotective activity against CCl₄-induced liver injury in rats.
노랑초파리 ecdysone 결핍 돌연변이(ecd1ts)에서 기능적 ecdysone수용체 구성분의 발현

In Drosophila, the steroid hormone ecdysone triggers the key regulatory cascades controlling the coordinated changes in the developmental pathway of molting and metamorphosis. Ecdysone action is mediated by heterodimer consisting of the ecdysone receptor(EcR) and ultraspiracle proteins(Usp). Heterodimers of these proteins bind to ecdysone response element and ecdsone to modulate gene transcription. In this study, the transcripts of functional ecdsone receptors and ecdysone response genes(Fbp1 and Uro) in ecdysoneless(ecd-1ts) mutation were analyzed by using a RT-PCR and competitive RT-PCR method. The expression of functional ecdysone receptor components were not correlated with tissue-specific ecdysone response genes(Fbp1 and Uro), Suggesting that several other factors involved in the complexity of various ecdysone responses in Drosphila melanogaster. But we showed that the transcripts of EcR-B1 isoforms were expressed with higher level than those of EcR-A in the late third instar larvae of ecdysoneless(ecd-1ts) mutation.
Stability Evaluation of National Reference Standards for Blood Products in Korea

National reference standards (NRSs) for biologics are established through potency estimation by a multi-center joint study of standard materials used in the approval process for national lot release and quality control of vaccines, blood products, and other biologics. In this study, a stability evaluation was conducted to determine whether the potency of NRSs for six blood products was being maintained at a consistent level in Korea. The present study conducted real-time stability tests via in-vivo/in-vitro bioassay on NRSs for blood coagulation factor VIII concentrate (2nd standard), antithrombin concentrate, prekallikrein activator, anti-hepatitis B immunoglobulin, blood coagulation factor IX concentrate, and anti-tetanus human immunoglobulin, as well as a trend analysis using cumulative annual results. The real-time stability test results showed that the mean potency of six NRSs was all within the control limit. In the trend analysis, the potency of NRS for blood coagulation factor VIII concentrate (2nd standard) showed a decreasing trend, while the potency of all other products had been stably maintained. The present study confirmed that the mean potency of NRSs for six blood products had been stably maintained in Korea. The findings of the present study establish a foundation that can ensure the quality of NRSs for biologics in Korea, and it is expected to make a major contribution to the supply of high-quality biologics.
Expression of rpr, grim, dcp-1, diapl, and diap2 during Drosophila Development

초파리 발생과정에서 세포사멸에 관여하는 유전자인 reaper(rpr), grim, dcp-1, diap1, diap1, diap2의 발현양상을 경제적 RT-PCR 방법으로 분석하였다. 세포사멸 유도 유전자인 rpr, grim의 발현양상은 발생단계에 따른 ecdysone titer 변화 양상과 매우 유사하였다. Effector caspase인 dcp-1 전사체는 초기 배와 암컷 성체에서 높은 발현을 보였다. 반면에 세포사멸 억제인자인 diap1과 diap The developmental profiles of rpr, grim, dcp-1, diapl, diap2 transcripts, which were involved in programmed cell death, were analyzed using competitive RT-PCR in whole animals during Drosophila development. The fluctuation patterns of transcript levels of the apoptotic initiators(rpr and grim) were similar to those of the ecdysone titer in Drosophila life cycle. The transcript of dcp-1, which is considered as effector caspase, was expressed strong1y at early embryo and female adult stages. However, the transcript levels of anti-apoptotic factors diap1 and diap2, showed the reverse pattern comparing with those of apoptotic factors(rpr and grim). Also, the transcript levels of rpr, diap2 and dcp-1 were quantified in the salivary glands and wing discs dissected from the wandering late third instar larva. The transcript levels of rpr and diap2 were changed reversely each other in both tissues from wandering stage to puparium formation. These results suggest that the expressions of cell death related genes are regulated by the ecdysone signals during normal development.

Trialkyltin n-$Bu_3SnH$, an endocrine disruptor, was slowly converted by the catalytic action of group 4 $Cp_2MCl_2$/Red-Al (M = Ti, Zr, Hf) to produce two phases of products: one is an insoluble cross-linked solid, polystannane in 7-23% yield as minor product via redistribution/dehydrocoupling combination process, and the other is an oil, hexabutyldistannane in 69-90% yield as major product via simple dehydrocoupling process. Redistribution/dehydrocoupling process first produced a low-molecular-weight oligostannane possessing partial backbone Sn-H bonds which then underwent an extensive cross-linking reaction of backbone Sn-H bonds, leading to an insoluble polystannane. This is the first exciting example of redistribution/dehydrocoupling of a tertiary hydrostannane catalyzed by early transition metallocenes.

<P>Rapid and highly sensitive detection of duplex dye-labelled DNA sequences in a PDMS microfluidic channel was investigated using confocal surface enhanced Raman spectroscopy (SERS). This method does not need either an immobilization procedure or a PCR amplification procedure, which are essential for a DNA microarray chip. Furthermore, Raman peaks of each dye-labelled DNA can be easily resolved since they are much narrower than the corresponding broad fluorescence bands. To find the potential applicability of confocal SERS for sensitive bio-detection in a microfluidic channel, the mixture of two different dye-labelled (TAMRA and Cy3) sex determining Y genes, SRY and SPGY1, was adsorbed on silver colloids in the alligator teeth-shaped PDMS microfluidic channel and its SERS signals were measured under flowing conditions. Its major SERS peaks were observable down to the concentration of 10<SUP>−11</SUP> M.
