        Effects of a supportive workplace environment on the success rate for smoking cessation camp

        Background: This study was conducted to identify the success rate for smoking cessation over time after participation in a therapeutic smoking cessation camp, and to identify how participant characteristics, including a supportive workplace environment for smoking cessation (SWESC), affect the success rate for smoking cessation. Methods: In all, 296 participants at smoking cessation camps in Ulsan between 2015 and 2020 were investigated. The success rates of smoking cessation after weeks 4, 6, 12, and 24 at camp were investigated. The participants were grouped as workers with an SWESC, and workers without an SWESC, and variables (age, education, household income, marital status, drinking, exercise, body mass index, morbidity, job, number of counseling sessions, cigarettes smoked per day and smoking initiation age) were investigated. Multiple logistic regression analysis was conducted at each time point. In addition, Cox regression analysis was performed to evaluate the variables affecting the success rate for smoking cessation over time. Results: The smoking cessation success rate of workers with an SWESC at week 24 (90.7%) was higher than that for workers without an SWESC (60.5%). Multiple logistic regression was performed to determine the relationship between each variable and the success rates for smoking cessation at week 6, 12, and 24. SWESC was confirmed as significant (p < 0.05) variables for increased success rate for smoking cessation at all 3 time points. After adjusting for all variables, the Cox proportional hazards survival analysis showed a hazard ratio of 6.17 for SWESC (p < 0.001,; 95% confidence interval: 3.08–12.38). Conclusions: At a professional treatment smoking cessation camp, participants with an SWESC showed a significantly higher success rate for smoking cessation. Supportive workplace environment for workers’ health is expected to be an important factor for smoking cessation projects as well as other health promotion projects at workplace.

      • Roles of Wnt Target Genes in the Journey of Cancer Stem Cells

        <P>The importance of Wnt/β-catenin signaling in cancer stem cells (CSCs) has been acknowledged; however, the mechanism through which it regulates the biological function of CSCs and promotes cancer progression remains elusive. Hence, to understand the intricate mechanism by which Wnt controls stemness, the specific downstream target genes of Wnt were established by analyzing the genetic signatures of multiple types of metastatic cancers based on gene set enrichment. By focusing on the molecular function of Wnt target genes, the biological roles of Wnt were interpreted in terms of CSC dynamics from initiation to metastasis. Wnt signaling participates in cancer initiation by generating CSCs from normal stem cells or non-CSCs and augmenting persistent growth at the primary region, which is resistant to anti-cancer therapy. Moreover, it assists CSCs in invading nearby tissues and in entering the blood stream, during which the negative feedback of the Wnt signaling pathway maintains CSCs in a dormant state that is suitable for survival. When CSCs arrive at distant organs, another burst of Wnt signaling induces CSCs to succeed in re-initiation and colonization. This comprehensive understanding of Wnt target genes provides a plausible explanation for how Wnt allows CSCs variation during cancer progression.</P>

      • Application of Newly Developed Amniotic Membrane Ointment for Photorefractive Keratectomy in Rabbits

        <P>We developed amniotic membrane ointment (AMO), and the effect of instilling the AMO after photorefractive keratectomy (PRK) was investigated with respect to inflammatory cell infiltration into the corneal stroma, apoptosis of keratocytes, and suppression of lipid peroxidation of cellular walls. The PRK procedure was performed on both eyes of 10 white rabbits. One eye of each rabbit (the experimental eye) was instilled with the AMO and the other eye of the rabbit (the control eye) with a base ointment 0, 8 and 16 h after the PRK procedure. Corneal specimens were collected 24 h after the PRK procedure. Hematoxylin-eosin stain and TUNEL assay were conducted to demonstrate polymorphonuclear and apoptotic cells, respectively. To assess lipid peroxidation, immunohistochemical staining with an antibody to malondialdehyde was undertaken. Compared to the control, the cornea instilled with the AMO had significantly less polymorphonuclear cells infiltrating into the corneal stroma as well as keratocytes subjected to apoptosis. These corneas also showed a significantly less extent of lipid peroxidation than the control. These data support that instillation of the AMO effectively reduced the recruitment of polymorphonuclear cells, the induction of apoptosis in keratocytes, and lipid peroxidation induced by PRK. Thus, this study could provide basic data on the clinical application of the AMO in the days ahead.</P><P>Copyright © 2006 S. Karger AG, Basel</P>

        Modern technology has lessened the physical demand of every activities. Thus, physical inactivity has led to a rise in `Hypokinetic diseases'. The prefix, hypo, means lack of and kinetic refers to movement. Individuals who do not exercise regularly are at greater risk of developing hypokinetic diseases such as coronary heart disease, hypertension, hyperlipidemia, obesity, musculoskeletal disease. Systematic exercise program defenses development of hypokinetic diseases. Exercise program bases on basic physical fitness test. The physical fitness evaluation is used to degree of Wellness of the individuals. It includes the evaluation of Health-Related Physical Fitness - muscular strength, muscular endurance, flexibility, cardiorespiratory endurance, body composition - and skill or sports related fitness -agility, balance, coordination, reaction time, speed. In present study, the authors investigated health-related physical fitness test result and questionnaire for 133 male and 71 female from January 16 to March 15, 1997. The purpose of this study is to observe relationship health related physical fitness and self-perceived health status. The results are as follows. 1. There were statistically significant differences in BMI, grip strength, back strength, muscular endurance, cardiorespiratory endurance, and flexibility between male and female subjects. Flexibility is higher in female than male subject, but Others are higher in male than female subjects. 2. There were statistically significant differences in grip strength, muscular endurance between 30s, 40s, and 50s group in both gender subjects, and cardiorespiratory endurance between the groups especially in Female subjects. 3. For male subjects, flexibility was measured lower in group who classified low level in self-perceived health status than group of others. Also, for female subjects, Muscular endurance & flexibility were measured. 4. It was shown that grip strength, back strength, and muscular endurance were significantly lower in group who have chronic diseases than normal group for Male subjects. But, for female subjects, all the component were statistically insignificant results between normal and disease group. 5. Both male and female subjects, there were positive correlation among grip strength, back strength, muscular endurance, cardiorespiratory endurance, and flexibility but, negative correlation for age. Therefore, health-related physical fitness is very important component for Health Promotion & Wellness. Physical Fitness test is valuable test in health evaluation, health management and health promotion, so available for hospital, sports center, community health center, industrial field, school etc.

      • Highly Selective Xylene Sensor Based on NiO/NiMoO<sub>4</sub> Nanocomposite Hierarchical Spheres for Indoor Air Monitoring

        <P>Xylene is a hazardous volatile organic compound, which should be measured precisely for monitoring of indoor air quality. The selective detection of ppm-level xylene using oxide semiconductor chemiresistors, however, remains a challenging issue. In this study, NiO/NiMoO4 nanocomposite hierarchical spheres assembled from nanosheets were prepared by hydrothermal reaction, and the potential of sensors composed of these nanocomposites to selectively detect xylene gas was investigated. The sensors based on the NiO/NiMoO4 nanocomposite hierarchical spheres exhibited high responses (maximum resistance ratio =101.5) to 5 ppm p-xylene with low cross-responses (resistance ratios <30) to 5 ppm toluene, benzene, C2H5OH, CH3COCH3, HCHO, CO, trimethylamine, and NH3. In contrast, a sensor based on pure NiO hierarchical spheres exhibited negligibly low responses to all 9 analyte gases. The gas-sensing mechanism underlying the high selectivity and response to xylene in the NiO/NiMoO4 nanocomposite hierarchical spheres is discussed in relation to the catalytic promotion of the xylene-sensing reaction by synergistic combination between NiO and NiMoO4, gas-accessible hierarchical morphology, and electronic sensitization by Mo addition. Highly selective detection of xylene can pave the road toward a new solution for precise monitoring of indoor air pollution.</P>

      • Effect of antifreeze proteins on vitrification of mouse oocyte: Comparison of three different antifreeze proteins

        The aim of this study was to analyze the effect of antifreeze proteins (AFPs) on vitrification of mouse mature (MII) oocytes. We studied about 3 types of AFPs from different origins (FfIBP, LeIBPand Type III AFP). The MII oocytes were obtained from 4-week-old BD-F1 mice. Vitrification of oocyte was performed by 2 steps using the Cryotop (equilibration: 7.5% EG + 7.5% PROH for 5 min, vitrification: 15% EG + 15% PROH + 0.5M sucrose for 1 min). The concentrations of AFPs added to these solutions were 0.05 mg/ml for FfIBP and 0.1 mg/ml for LeIBP and Type III AFP. After fertilization, embryo development was assessed up to 5 days. Through immunostaining of vitrified-warmed oocytes, we assessed the normal meiotic spindle. Also, intracellular ROS and mitochondrial activity was analyzed. In the developmental stages, FfIBP and LeIBP groups showed significantly higher survival rates. In the blastocyst and apoptotic blastomere rates were significant differences in AFPs treated groups. AFPs treated groups were significantly higher in blastocyst cell numbers than control group. Among the AFPs treated groups, FfIBP, LeIBP groups were significantly higher rates. And, in cleavage rates, FfIBP group was significantly higher rates than the other groups. In vitrified-warmed MII oocytes, the normal meiotic spindle organization and chromosome alignment rate was significantly higher in FfIBP and LeIBP groups. And in intracellular ROS levels, control group was significantly increased than AFPs treated groups. However, in the mitochondrial activity, LeIBP group was significantly higher than control, FfIBP and LeIBP groups. AFPs treated groups were significant differences in development, meiotic spindle organization and intracellular ROS levels. And in the AFPs treated groups, FfIBP and LeIBP groups were significantly higher rates in normal meiotic spindle and mitochondrial activity than Type III AFP group respectively. In conclusion, FfIBP and LeIBP can be thought to improve oocyte cryopreservation efficiency.

        조기 유방암 환자를 위한 다지표 예후 예측 검사 GenesWell<sup>TM</sup> BCT의 분석적 성능 시험

        GenesWell<sup>TM</sup> BCT는 호르몬 수용체 양성, HER2 음성, 및 pN0 또는 1인 조기 유방암 환자의 10년내 타 장기 전이 재발 위험도를 제시하는 다지표 예후 예측 검사로, 예후에 대한 위험을 BCT Score로 제시한다. 본 연구에서는 GenesWell<sup>TM</sup> BCT의 분석적 성능을 검사하였다. 조기 유방암 환자의 FFPE 검체로 부터 추출한 RNA를 대상으로 GenesWell<sup>TM</sup> BCT 수행하여, 12개유전자의 발현값을 측정하였다. GenesWell<sup>TM</sup> BCT의 최소검출한계, 공란 한계 및 측정 범위는 단계 희석한 RNA 검체를 사용하여 평가하였으며, 분석적 정밀도 및 특이도 시험은 BCT Score에 따라 저위험군, 고위험군 그리고 경계선 주변으로 나누어진 3개의 RNA 검체를 이용하여 시험하였다. GenesWell<sup>TM</sup> BCT는 1 ng/μL 미만의 RNA 검체에서 RNA를 측정할 수 있었으며, 다기관에서 수행된 분석적 정밀도 시험에서 반복성 100% 및 재현성 98.3%의 결과를 확인할 수 있었다. 또한, 분석적 특이도 시험을 통해, 간섭 물질이 검체의 재발 위험성 판정에 영향을 미치지 않음을 확인할 수 있었다. 이들 결과는 GenesWell<sup>TM</sup>BCT가 95% 이상의 항상성을 나타내는 높은 분석적 성능을 가지고 있음을 제시한다. GenesWell<sup>TM</sup> BCT is a 12-gene test suggesting the prognostic risk score (BCT Score) for distant metastasis within the first 10 years in early breast cancer patients with hormone receptor-positive, HER2-negative, and pN0∼1 tumors. In this study, we validated the analytical performance of GenesWell<sup>TM</sup> BCT. Gene expression values were measured by a one-step, real-time qPCR, using RNA extracted from FFPE specimens of early breast cancer patients. Limit of Blank, Limit of Detection, and dynamic range for each of the 12 genes were assessed by serially diluted RNA pools. The analytical precision and specificity were evaluated by three different RNA samples representing low risk group, high risk group, and near-cutoff group in accordance with their BCT Scores. GenesWell<sup>TM</sup> BCT could detect gene expression of each of the 12 genes from less than 1 ng/μL of RNA. Repeatability and reproducibility across multiple testing sites resulted in 100% and 98.3% consistencies of risk classification, respectively. Moreover, it was confirmed that the potential interference substances does not affect the risk classification of the test. The findings demonstrate that GenesWell<sup>TM</sup> BCT have high analytical performance with over 95% consistency for risk classification.
