비정질 PEEK필름의 self-bonding공정시 조건 변화에 따르는 결정화도(crystallinlty)변화가 self-bonding강도에 미치는 영향을 고찰하기 위하여, 비정질 PEEK필름을 그 2차천이온도(Tg=143$^{\circ}C$)와 용융점(Tm=335$^{\circ}C$) 사이의 여러 온도에서 일정 압력 하에서 접합 시간을 달리하여 self-bonding시킨 후, 각 조건에서 개발된 self-bonding강도를 측정하고, 이에 따르는 결정화도 변화를 DSC를 이용하여 비교 분석하였다. 결정화도는 접합공정변수(시간과 온도)의 함수로서 증가하였고, 동일한 값의 결정화도를 보이는 시편들의 경우에도 접합공정의 조건에 따라 결과적인 self-bonding강도는 큰 차이가 있음을 보였다. 또한 접합후 시편을 상온으로 노냉시키는 동안에는 더 이상의 결정화 현상이 일어나지 않음이 DSC분석을 통하여 판명되었다. The relationship between the variation of crystallinity and the resultant self-bonding strength of PEEK was examined by using DSC in conjunction with the shear test. DSC measurement of the crystallinity produced at different bonding conditions demonstrated that even though PEEK specimens contain the same amount of crystallinity, the resultant self-bonding strength is sensitively dependent on bonding history. It also showed that all crystallization during the bending process occurs only in the healing and annealing stage and no additional crystallization occurs in the cooling stage.
비정질 PEEK 필름의 Self-Bonding강도에 미치는 제조공정변수의 영향

비정질 PEEK 필름의 self-bonding강도는 접합시의 공정변수(시간, 온도, 그리고 압력)와 밀접한 관계가 있다. 본 연구에서는 이러한 공정변수의 효과를 규명하기 위하여 각기 다른 접합조건하에서 개발된 시편들의 self-bonding강도를 single lap-shear test를 통하여 측정된 각각의 전단 응력(shear strength)으로 나타내었다. 개발된 self-bonding강도는 접합온도가 증가함에 따라 증가하였으며, 접합시간의 1/4승에 일차함수적으로 비례증가하였다. 접합공정 중의 압력의 효과는 단지 초기 접합단계인 wetting에 기여하였을 뿐 self-bonding강도 자체에는 거의 영향을 미치지 않는 것으로 사려되었다. 결론적으로 비정질 PEEK 필름의 self-bonding현상은 현장에서의 실제 접합공정에서 어떠한 접착재료의 사용없이도 모재와 같은 강도를 개발하는데 무한한 가능성이 있는 것으로 판단되었다. Self-bonding strength developed at the interface of amorphous PEEK films is highly sensitive to the processing variables(time, temperature, and pressure) during the bonding process. In order to examine the effects of these processing variables, amorphous PEEK films were bonded at various bonding conditions and the resultant interfacial bond strengths were measured using a modified single lap-shear test. Experimental results showed that the developed self-bonding strength increases with increase in bonding temperature and is directly proportional to the bonding time raised to the 1/4 power. The applied pressure seems only to produce better wetting at the beginning stage of the bonding process. Conclusively, the self-bonding of amorphous PEEK films provides a great potential for developing excellent bond strength approaching the strength of the parent material without any adhesives in structural applications.
Acoustic Microscopy를 이용한 무정형 PEEK의 접합 계면 연구

세라믹 강화 복합 재료의 모재로서 사용되는 무정형 PEEK가 보여주는 self-bonding 현상의 주 기구인 PEEK체인들의 확산(interdiffusion)과 뒤엉킴(entanglement)이 일어나기 위하여 PEEK의 접합 면에서 반드시 선행되어 일어나야 하는 젖음성의 정도에 미치는 접합 공정 변수의 영향을 C-mode acoustic microscopy를 이용하여 고찰하였다. 또한 self-bonding 된 PEEK시편들의 전단 변형시 전단 하중의 증가에 따라 일어나는 접합 면에서의 debonding 정도를 측정함으로써 접합 면에서 일어나는 파괴 거동을 관찰하였다. 각각의 접합 조건에서의 젖음성의 정도는 시간과 압력의 증가에 따라 다소 증가함을 보여 주었으나, 접합 온도와는 거의 무관함을 보여 주었다. 또한 전단 파괴 시험시 각각의 접합 조건 하에서 개발된 self-bonding강도의 80%-90%이상의 전단 하중이 가해진 후부터 debonding이 시작되어, 이 후 하중이 증가함에 따라 급속도로 진행되어 파괴가 일어남을 알 수 있었다.

Farnesol is a quorum-sensing molecule that inhibits biofilm formation in Candida albicans. Previous in vitro data suggest that, in combination with certain antifungals, farnesol may have an adjuvant anti-biofilm agent. However, the in vivo efficacy of farnesol is very questionable. Therefore, the in vitro and in vivo activity of fluconazole combined with farnesol was evaluated against C. albicans biofilms using fractional inhibitory concentration index (FICI) determination, time-kill experiments and a murine vulvovaginitis model. The median biofilm MICs of fluconazole-sensitive C. albicans isolates ranged between 4 -> 512 mg/L and 150–300 μM for fluconazole and farnesol, respectively. These values were 512 -> 512 mg/L and > 300 μM for fluconazole-resistant clinical isolates. Farnesol decreased the median MICs of fluconazole by 2-64-fold for biofilms. Based on FICI, synergistic interaction was observed only in the case of the sessile SC5314 reference strain (FICIs: 0.16–0.27). In time-kill studies, only the 512 mg/L fluconazole and 512 mg/L fluconazole + 75 μM farnesol reduced biofilm mass significantly at each time point in the case of all isolates. The combination reduced the metabolic activity of biofilms for all isolates in a concentration- and time-dependent manner. Our findings revealed that farnesol alone was not protective in a murine vulvovaginitis model. Farnesol was not beneficial in combination with fluconazole for fluconazole-susceptible isolates, but partially increased fluconazole activity against one fluconazole- resistant isolate, but not the other one.

We investigated the activity of caspofungin against a Candida tropicalis clinical isolate showing paradoxical growth in vitro. BALB/c mice immunosuppressed by cyclophosphamide were infected intraperitoneally using 107 CFU/mouse. Caspofungin was administered intraperitoneally once daily for 5 days or as a single dose using the following doses: 0.12, 0.25, 1, 2, 3, 5, and 15 mg/kg. The single dose of caspofungin was effective only at 5 and 15 mg/kg concentrations (100% survival). Five-day caspofungin treatment led to 100% survival at doses of 1 mg/kg or higher. Caspofungin treatment significantly decreased the number of viable yeasts in the peritoneal lavage samples as well as in the infected abscesses at doses 1, 3, 5, and 15 mg/kg caspofungin as compared to the untreated control (P<0.001 in all cases), and even to the group treated with 0.12 mg/kg caspofungin (P<0.05 in all cases). At 2 mg/kg caspofungin dose, sterilization of the internal organs was reproducibly incomplete, suggesting that the role of paradoxical growth in the late clinical failure cannot be excluded.
